    Beauty from the deep: cnidarians in cosmetics

    Cnidarian proteins are considered useful for the development of therapeutics, as well as this they have also received the attention of biotechnology and the cosmetic industries. In 2017 the first ever sea anemone venom peptide cosmetic, named SensAmone P5, was launched by Mibelle Biochemistry. This synthetic peptide is based on the interaction of APHC1, from Heteractis crispa, on the pain relevant ion channel TRPV1. This peptide reduces TRPV1 signalling in-vitro and skin sensitivity in human volunteers. Aside from venoms, jellyfish mucus and collagen are both used in cosmetic preparations. Many legal definitions of animals do not include the invertebrates and thus it is likely that invertebrate proteins are more acceptable as an alternative to mammalian proteins. Mucins are important proteins for moisturisers and using jellyfish as the source appears to be a suitable alternative to bovine and porcine proteins which were previously used. The main structural protein that supports the soft bodied jellyfish is collagen. This collagen appears to be biocompatible with human tissues and thus has been successful as a cosmetic, as well as being used in-vitro for 3D tissue engineering scaffolds.     The failures of ethnobotany and phytomedicine in delivering novel treatments for snakebite envenomation

    Snakebite envenomation (SBE) is a high priority, neglected tropical disease. This devastating occupational health hazard disproportionately affects rural farming communities in tropical countries. This is exacerbated by the distribution and densities of venomous snakes, incidence of encounters and limited access to advanced healthcare, including antivenom. Before the development of antivenom, desperation and spiritual beliefs led patients to experiment with a wide range of traditional treatments. Many of these treatments still survive today, particularly in regions where access to healthcare is limited. Plants are a major source of bioactive molecules, including several lifesaving medications that are widely used to this day. However, much of the research into the use of traditional plant treatments for SBE are limited to preliminary analysis, or have focused on techniques used to confirm antibody efficacy that are not suitable for non-antibody containing treatments. Modern drugs are developed through a robust pharmaceutical drug discovery and development process, which applies as much to SBE as it does to any other disease. This review discusses specifically why research into ethnobotanical practices has failed to identify or develop a novel treatment for SBE, and proposes specific approaches that should be considered in this area of research in the future

    Recent changes in summer distribution and numbers of migratory caribou on the southern Hudson Bay coast

    The status of migratory woodland caribou inhabiting the coastal region in southern Hudson Bay is dynamic. The Pen Islands Herd within that region was defined in the 1990s, but opportunistic observations between 1999 and 2007 suggested that its status had significantly changed since the late 1980s and early 1990s. We undertook systematic surveys from the Hayes River, MB, to the Lakitusaki River, ON, in 2008 and 2009 to determine current distribution and minimum numbers of woodland caribou on the southern Hudson Bay coast from the Hayes River, Manitoba, to the Lakitusaki River, Ontario. We documented a significant change in summer distribution during the historical peak aggregation period (7-15 July) compared to the 1990s. In 2008 and 2009, respectively, we tallied 3529 and 3304 animals; however, fewer than 180 caribou were observed each year in the Pen Islands Herd’s former summer range where over 10 798 caribou were observed during a systematic survey in 1994. Over 80% of caribou were in the Cape Henrietta Maria area of Ontario. Calf proportions in herds varied from 8% of animals in the west to 20% in the east. Our 2008 and 2009 systematic surveys were focused on the immediate coast, but one exploratory flight inland suggested that more caribou may be inland than had been observed in the 1980s-1990s. The causes of change in the numbers and distribution in the coastal Hudson Bay Lowlands and the association of current caribou with the formerly large Pen Islands Herd may be difficult to determine because of gaps in monitoring, but satellite telemetry, genetic sampling, remote sensing, habitat analysis, and aboriginal knowledge are all being used to pursue answers

    Bacterial Adaptation to Venom in Snakes and Arachnida

    Animal venoms are considered sterile sources of antimicrobial compounds with strong membrane-disrupting activity against multidrug-resistant bacteria. However, venomous bite wound infections are common in developing nations. Investigating the envenomation organ and venom microbiota of five snake and two spider species, we observed venom community structures that depend on the host venomous animal species and evidenced recovery of viable microorganisms from black-necked spitting cobra (Naja nigricollis) and Indian ornamental tarantula (Poecilotheria regalis) venoms. Among the bacterial isolates recovered from , we identified two venom-resistant, novel sequence types of Enterococcus faecalis whose genomes feature 16 virulence genes, indicating infectious potential, and 45 additional genes, nearly half of which improve bacterial membrane integrity. Our findings challenge the dogma of venom sterility and indicate an increased primary infection risk in the clinical management of venomous animal bite wounds. Notwithstanding their 3 to 5% mortality, the 2.7 million envenomation-related injuries occurring annually-predominantly across Africa, Asia, and Latin America-are also major causes of morbidity. Venom toxin-damaged tissue will develop infections in some 75% of envenomation victims, with E. faecalis being a common culprit of disease; however, such infections are generally considered to be independent of envenomation. Here, we provide evidence on venom microbiota across snakes and arachnida and report on the convergent evolution mechanisms that can facilitate adaptation to black-necked cobra venom in two independent E. faecalis strains, easily misidentified by biochemical diagnostics. Therefore, since inoculation with viable and virulence gene-harboring bacteria can occur during envenomation, acute infection risk management following envenomation is warranted, particularly for immunocompromised and malnourished victims in resource-limited settings. These results shed light on how bacteria evolve for survival in one of the most extreme environments on Earth and how venomous bites must be also treated for infections

    The impact of physical inactivity on glucose homeostasis when diet is adjusted to maintain energy balance in healthy, young males

    Background & aims: It is unclear if dietary adjustments to maintain energy balance during reduced physical activity can offset inactivity-induced reductions in insulin sensitivity and glucose disposal to produce normal daily glucose concentrations and meal responses. Therefore, the aim of the present study was to examine the impact of long-term physical inactivity (60 days of bed rest) on daily glycemia when in energy balance.Methods: Interstitial glucose concentrations were measured using Continuous Glucose Monitoring Systems (CGMS) for 5 days before and towards the end of bed rest in 20 healthy, young males (Age: 34 ± 8 years; BMI: 23.5 ± 1.8 kg/m2). Energy intake was reduced during bed rest to match energy expenditure, but the types of foods and timing of meals was maintained. Fasting venous glucose and insulin concentrations were determined, as well as the change in whole-body glucose disposal using a hyperinsulinemic-euglycemic clamp (HIEC).Results: Following long-term bed rest, fasting plasma insulin concentration increased 40% (p = 0.004) and glucose disposal during the HIEC decreased 24% (p < 0.001). Interstitial daily glucose total area under the curve (tAUC) from pre-to post-bed rest increased on average by 6% (p = 0.041), despite a 20 and 25% reduction in total caloric and carbohydrate intake, respectively. The nocturnal period (00:00–06:00) showed the greatest change to glycemia with glucose tAUC for this period increasing by 9% (p = 0.005). CGMS measures of daily glycemic variability (SD, J-Index, M-value and MAG) were not changed during bed rest.Conclusions: Reduced physical activity (bed rest) increases glycemia even when daily energy intake is reduced to maintain energy balance. However, the disturbance to daily glucose homeostasis was much more modest than the reduced capacity to dispose of glucose, and glycemic variability was not negatively affected by bed rest, likely due to positive mitigating effects from the contemporaneous reduction in dietary energy and carbohydrate intake.Clinical trials record: NCT03594799 (registered July 20, 2018) (https://clinicaltrials.gov/ct2/show/NCT03594799)

    Gaps in detailed knowledge of human papillomavirus (HPV) and the HPV vaccine among medical students in Scotland

    <p>Background: A vaccination programme targeted against human papillomavirus (HPV) types16 and 18 was introduced in the UK in 2008, with the aim of decreasing incidence of cervical disease. Vaccine roll out to 12–13 year old girls with a catch-up programme for girls aged up to 17 years and 364 days was accompanied by a very comprehensive public health information (PHI) campaign which described the role of HPV in the development of cervical cancer.</p> <p>Methods: A brief questionnaire, designed to assess acquisition of knowledge of HPV infection and its association to cervical cancer, was administered to two different cohorts of male and female 1st year medical students (school leavers: 83% in age range 17–20) at a UK university. The study was timed so that the first survey in 2008 immediately followed a summer's intensive PHI campaign and very shortly after vaccine roll-out (150 students). The second survey was exactly one year later over which time there was a sustained PHI campaign (213 students).</p> <p>Results: We addressed three research questions: knowledge about three specific details of HPV infection that could be acquired from PHI, whether length of the PHI campaign and/or vaccination of females had any bearing on HPV knowledge, and knowledge differences between men and women regarding HPV. No female student in the 2008 cohort had completed the three-dose vaccine schedule compared to 58.4% of female students in 2009. Overall, participants’ knowledge regarding the sexually transmitted nature of HPV and its association with cervical cancer was high in both year groups. However, in both years, less than 50% of students correctly identified that HPV causes over 90% of cases of cervical cancer. Males gave fewer correct answers for these two details in 2009. In 2008 only around 50% of students recognised that the current vaccine protects against a limited subset of cervical cancer-causing HPV sub-types, although there was a significant increase in correct response among female students in the 2009 cohort compared to the 2008 cohort.</p> <p>onclusions: This study highlights a lack of understanding regarding the extent of protection against cervical cancer conferred by the HPV vaccine, even among an educated population in the UK who could have a vested interest in acquiring such knowledge. The intensive PHI campaign accompanying the first year of HPV vaccination seemed to have little effect on knowledge over time. This is one of the first studies to assess detailed knowledge of HPV in both males and females. There is scope for continued improvements to PHI regarding the link between HPV infection and cervical cancer.</p&gt
