    The authors cover the problem of the of future educators of pre-school establishments professional training, their personal professional self-improvement in the educational space of the university. It is analyzed, the content of the categories «professional formation», «professional self-improvement», «responsibility» is specified. To reveal the phenomenon of professional self-improvement of the future specialist, a considerable body of domestic and foreign scientific literature has been used. Among the most significant factors, the authors singled out the following: the motivation of the student-future educator and his needs for achievement; content of the person’s value-content sphere and its quality (abilities, character, mental processes, states that accumulate professional-important qualities); integral characteristics of personality (responsibility, individual style, creativity as creative potential)

    Plasmid DNA initiates replication of yellow fever vaccine in vitro and elicits virus-specific immune response in mice

    AbstractYellow fever (YF) causes an acute hemorrhagic fever disease in tropical Africa and Latin America. To develop a novel experimental YF vaccine, we applied iDNA infectious clone technology. The iDNA represents plasmid that encodes the full-length RNA genome of 17D vaccine downstream from a cytomegalovirus (CMV) promoter. The vaccine was designed to transcribe the full-length viral RNA and to launch 17D vaccine virus in vitro and in vivo. Transfection with 10ng of iDNA plasmid was sufficient to start replication of vaccine virus in vitro. Safety of the parental 17D and iDNA-derived 17D viruses was confirmed in AG129 mice deficient in receptors for IFN-α/β/γ. Finally, direct vaccination of BALB/c mice with a single 20μg dose of iDNA plasmid resulted in seroconversion and elicitation of virus-specific neutralizing antibodies in animals. We conclude that iDNA immunization approach combines characteristics of DNA and attenuated vaccines and represents a promising vaccination strategy for YF

    Identification and characterization of the mouse nuclear export factor (Nxf) family members

    TAP/hNXF1 is a key factor that mediates general cellular mRNA export from the nucleus, and its orthologs are structurally and functionally conserved from yeast to humans. Metazoans encode additional proteins that share homology and domain organization with TAP/hNXF1, suggesting their participation in mRNA metabolism; however, the precise role(s) of these proteins is not well understood. Here, we found that the human mRNA export factor hNXF2 is specifically expressed in the brain, suggesting a brain-specific role in mRNA metabolism. To address the roles of additional NXF factors, we have identified and characterized the two Nxf genes, Nxf2 and Nxf7, which together with the TAP/hNXF1's ortholog Nxf1 comprise the murine Nxf family. Both mNXF2 and mNXF7 have a domain structure typical of the NXF family. We found that mNXF2 protein is expressed during mouse brain development. Similar to TAP/hNXF1, the mNXF2 protein is found in the nucleus, the nuclear envelope and cytoplasm, and is an active mRNA export receptor. In contrast, mNXF7 localizes exclusively to cytoplasmic granules and, despite its overall conserved sequence, lacks mRNA export activity. We concluded that mNXF2 is an active mRNA export receptor similar to the prototype TAP/hNXF1, whereas mNXF7 may have a more specialized role in the cytoplasm

    Induction of Somatic Embryogenesis in Siberian Dwarf Pine (Pinus pumila (Pall.) Regel) in vitro

    Индукция и пролиферация эмбриогенного каллуса являются ключевыми этапами для крупномасштабного размножения путем соматического эмбриогенеза и долгосрочного хранения зародышей хвойных видов. Однако существуют проблемы, такие как низкий отклик эксплантов на индукционных средах, низкая эффективность размножения, слабая жизнеспособность и потеря эмбриогенности каллусных культур в процессе культивирования. Для решения проблем соматического эмбриогенеза кедрового стланика (Pinus pumila (Pall.) Regel) мы варьировали концентрацию и комбинации регуляторов роста растений в средах для индукции эмбриогенного каллуса у зародышей на разных стадиях развития. Нами была установлена зависимость отклика экспланта от стадии его развития, от гормонального состава питательной среды, определены наилучшие условия культивирования эмбриогенного каллуса. Было установлено, что зиготические семядольные зародыши длиной 1 мм являются наиболее перспективным материалом для индукции каллусогенеза у P. pumila (85 %). Эмбриогенный каллус активнее образовывался на среде DCR с пониженной концентрацией регуляторов роста растений NAA (1 мг/л) и 6-BAP (0,5 и 1 мг/л). Пересадки на свежие среды того же состава каждые 14 дней позволяли не только поддерживать жизнеспособность каллусов, но также сохранять их способность к интенсивной пролиферации в течение годаInduction and proliferation of embryogenic calluses are the key stages in large-scale reproduction of coniferous species by somatic embryogenesis and long-term storage of their embryos. However, there are some issues, such as low reproduction efficiency, loss of embryogenic ability, poor viability. To solve the problems of somatic embryogenesis in the Siberian dwarf pine (Pinus pumila (Pall.) Regel), we aimed to determine the best cultivation conditions by varying the concentrations and combinations of plant growth regulators in the culture medium for inducing embryogenic calluses at different stages of embryo development. Explant response dependence on the stage of its development and the hormonal composition of the nutrient medium was established and optimal conditions for induction of somatic embryogenesis in Siberian dwarf pine were determined. Zygotic embryos with a length of 1 mm (mature embryo) were shown to be the most promising material for induction of Pinus pumila callogenesis (85 %). Embryogenic calluses were formed more actively on the DCR medium with reduced concentrations of plant growth regulators, NAA (1 mg/l) and 6-BAP (0.5 and 1 mg/l).     Methodological instruments in work became humanistic principles on the basis of integrative, anthropological approach. The aim of the article is to activate and popularize scientific researches in higher educational institutions that give an opportunity to reveal social-psychological and pedagogical factors that influencing on formation of ecological culture. We used a range of theoretical and experimental methods of investigation, namely, theoretical (analysis, synthesis, systematization) and empirical (psychological-pedagogical experiment, interview, diagnostic methods). It is proved that ecological culture of future pedagogue should be a basic component of educational process at the university. Purposeful, structured activity of pedagogical university in conditions of global ecological crisis creates conditions for development and formation of spiritual sphere of student youth, one of the components is ecological culture. Giving distressing results of research of rates of development of spiritual potential of future pedagogues, authors singled out social, psychological, pedagogical factors that have a significant influence on formation of favorable conditions in educational-culture space for accentuation of pedagogical activity on the development of spiritual potential of future pedagogue. We offer ways of solving of a problem in existing crisis situation in ecological aspect of educational process.Методологическим инструментарием в работе стали гуманистические принципы на основе интегративного антропологического подхода. Цель статьи заключается в активизации и популяризации научных исследований в высших учебных заведениях, которые дают возможность выявить социально-психологические и педагогические факторы, влияющие на формирование экологической культуры. Использован комплекс теоретических и экспериментальных методов исследования, а именно теоретические (анализ, синтез, систематизация) и эмпирические (психолого-педагогический эксперимент, опрос, диагностические методики). Доведено, что экологическая культура будущего педагога должна быть базовой составляющей учебно-воспитательного процесса в университете. Целеустремлённая, структурированная деятельность педагогического университета в условиях глобального экологического кризиса создаёт условия для развития и формирования духовной сферы студенческой молодёжи, одним из компонентов которой является экологическая культура. Предоставив неутешительные результаты исследования показателей развития духовного потенциала будущих педагогов, авторы выделили социальные, психологические, педагогическое факторы, которые оказывают существенное влияние на создание благоприятных условий в образовательно-культурном пространстве для акцентуации педагогической деятельности на развитие духовного потенциала будущего педагога.     The functioning of the banking sector of the economy in the world is characterized by a decrease in its capital intensity due to a decrease in the concentration of foreign capital, a deterioration in the quality of the loan portfolio, an outflow of urgent deposit resources and the receipt of significant losses by banking institutions during the COVID-19 pandemic. Taking into account the current trends in the spread of the virus, the development and implementation of fundamentally new approaches to anti-crisis management in the banking sector of both the world economy and Russia is of great importance today. The COVID-19 pandemic came as a big shock to the global and European economies. While banks are not hit by the pandemic as directly as other retail institutions, they are in the public eye as they provide funding to corporations and individuals. Their stability is crucial for maintaining the efficiency of the entire economic system, not only of a particular country, but also of the world. Currently, the need for a comprehensive study of the competitiveness of a commercial bank is determined by the objective modern conditions of the life of banks operating in a competitive environment. The article proposes the author’s methodology for assessing the competitiveness of a commercial bank, which includes five groups of indicators, which makes it possible to identify not only problematic, but also stable areas of activity. The proposed method for calculating the integral indicator for assessing the competitiveness of a commercial bank has been tested on the example of Russian Agricultural Bank and Sberbank, and a set of measures has been developed to increase the level of competitiveness of a commercial bank

    Problems and development trends of coal transportation infrastructure in Russia

    Analysis of the trends in the coal industry in Russia showed that, despite the stable demand in the global coal market, the growth rate of coal exports is slowing down. The main reason is the shortcomings of the transport and logistics infrastructure for the coal haulage. At the same time, the countries of the Asia-Pacific region are a promising export destination, and it is the Eastern operating domain of the Russian railway network that is most loaded and has insufficient throughput capacity. The study of the elements of the transport and logistics infrastructure revealed key problems, namely the underdevelopment of the railway infrastructure, the inconsistency of development programs for coal mining regions, port facilities and railway infrastructure, and the inconsistency of the operation of ports and the railway. Due to the fact that the development of the railway network in the eastern direction is a long-term project, and the coal haulage values will not be increased in the near future, an assessment is made of the possibility and economic efficiency of reorienting freight traffic in other directions

    The Analysis of Biocatalytic Properties of a Proteolytic Enzyme under the Influence of Physical Factors

    Due to the relevance of biotechnological methods application in the meat processing industry, consisting in the reuse of collagen-containing raw materials, as well as the use of enzymes and enzyme preparations to improve quality and extend the shelf life, which is confirmed by the research results conducted in the field of food biotechnology, the purpose of the work was formulated. It consists in studying the changes in the biocatalytic activity of the collagenase enzyme after treatment with a blue light spectrum. In the course of the work, it was revealed that the activity of the collagenase enzyme, which was treated with a blue light spectrum, has a wider optimum pH in comparison with the control samples. Treatment of the solution with blue light collagenase with a luminous flux of 35 µW / cm2 for 1 hour increases the optimum biocatalytic activity of the enzyme, which is confirmed by studies conducted in two alternative methods (according to Anson and Telishevskaya). The experimental data showed that the activity of the collagenase enzyme in the experimental samples increases from 10 to 20 % under the influence of table salt in concentrations up to 3.5% compared to the control group

    Single-Dose Immunogenic DNA Vaccines Coding for Live-Attenuated Alpha- and Flaviviruses

    Single-dose, immunogenic DNA (iDNA) vaccines coding for whole live-attenuated viruses are reviewed. This platform, sometimes called immunization DNA, has been used for vaccine development for flavi- and alphaviruses. An iDNA vaccine uses plasmid DNA to launch live-attenuated virus vaccines in vitro or in vivo. When iDNA is injected into mammalian cells in vitro or in vivo, the RNA genome of an attenuated virus is transcribed, which starts replication of a defined, live-attenuated vaccine virus in cell culture or the cells of a vaccine recipient. In the latter case, an immune response to the live virus vaccine is elicited, which protects against the pathogenic virus. Unlike other nucleic acid vaccines, such as mRNA and standard DNA vaccines, iDNA vaccines elicit protection with a single dose, thus providing major improvement to epidemic preparedness. Still, iDNA vaccines retain the advantages of other nucleic acid vaccines. In summary, the iDNA platform combines the advantages of reverse genetics and DNA immunization with the high immunogenicity of live-attenuated vaccines, resulting in enhanced safety and immunogenicity. This vaccine platform has expanded the field of genetic DNA and RNA vaccines with a novel type of immunogenic DNA vaccines that encode entire live-attenuated viruses

    Features of the growth and development of young pigs of various breeding

    The indicators of growth and development, fattening and meat qualities of pigs of the breeding center “Lozovoe” CJSC “Plemzavod-Yubileyny” of the Tyumen region were evaluated. The indicators that characterize the growth and development of young animals were taken into account: live weight, age, average daily growth. When the live weight of 100 kg was reached, an ultrasound device was used to evaluate the thickness of bacon, the depth of muscles and the yield of lean meat, which are in the database of breeding records for 2011-2020. To characterize the meat qualities, 1144 Landrace piglets were slaughtered, 275 - large white breed, 129 - Pietren breed, 339 hybrids (LxKB), 159 hybrids (LxD), 460 hybrids obtained from boars of foreign selection. A comparative analysis of commercial hybrids of various variants of crossing pigs of domestic and foreign selection is carried out.
