    Life History Parameters of Gulf Flounder (Paralichthys albigutta) From Northwest Florida

    Age, growth, natural and total mortality, a length-weight relationship, reproductive seasonality, sex ratio, maturity, and reproductive potential by size were estimated for gulf flounder, Paralichthys albigutta, from northwest Florida. The study used a fishery-independent approach during three annual migratory spawning seasons to sample gulf flounder offshore (spearing by divers) and inshore (multipanel trammel nets). We found gulf flounder to be slightly smaller at L∞ than other paralichthids (using either von Bertalanffy or damped growth functions), but gulf flounder obtained larger sizes and older ages than previously thought for this species [females to 575 mm total length (TL) and age 7, males to 373 mm TL and age 11], probably due to offshore sampling targeting more of the adult component of the stock. Given the older age structure than previously noted for this species, gulf flounder maturity and mortality estimates were similar to values reported for other U.S. paralichthids. Gulf flounder exhibited fall-winter gonad development with all evidence pointing to spawning occurring offshore. We estimated batch fecundity and spawning frequency, but it was apparent that there was a seasonal effect for both of these parameters, with increased oocyte density and nearly daily spawning occurring by late October to November, the peak period of development, also reflected in the gonadosomatic index

    Intronic Cis-Regulatory Modules Mediate Tissue-Specific and Microbial Control of angptl4/fiaf Transcription

    The intestinal microbiota enhances dietary energy harvest leading to increased fat storage in adipose tissues. This effect is caused in part by the microbial suppression of intestinal epithelial expression of a circulating inhibitor of lipoprotein lipase called Angiopoietin-like 4 (Angptl4/Fiaf). To define the cis-regulatory mechanisms underlying intestine-specific and microbial control of Angptl4 transcription, we utilized the zebrafish system in which host regulatory DNA can be rapidly analyzed in a live, transparent, and gnotobiotic vertebrate. We found that zebrafish angptl4 is transcribed in multiple tissues including the liver, pancreatic islet, and intestinal epithelium, which is similar to its mammalian homologs. Zebrafish angptl4 is also specifically suppressed in the intestinal epithelium upon colonization with a microbiota. In vivo transgenic reporter assays identified discrete tissue-specific regulatory modules within angptl4 intron 3 sufficient to drive expression in the liver, pancreatic islet β-cells, or intestinal enterocytes. Comparative sequence analyses and heterologous functional assays of angptl4 intron 3 sequences from 12 teleost fish species revealed differential evolution of the islet and intestinal regulatory modules. High-resolution functional mapping and site-directed mutagenesis defined the minimal set of regulatory sequences required for intestinal activity. Strikingly, the microbiota suppressed the transcriptional activity of the intestine-specific regulatory module similar to the endogenous angptl4 gene. These results suggest that the microbiota might regulate host intestinal Angptl4 protein expression and peripheral fat storage by suppressing the activity of an intestine-specific transcriptional enhancer. This study provides a useful paradigm for understanding how microbial signals interact with tissue-specific regulatory networks to control the activity and evolution of host gene transcription

    Effects of eight neuropsychiatric copy number variants on human brain structure

    Many copy number variants (CNVs) confer risk for the same range of neurodevelopmental symptoms and psychiatric conditions including autism and schizophrenia. Yet, to date neuroimaging studies have typically been carried out one mutation at a time, showing that CNVs have large effects on brain anatomy. Here, we aimed to characterize and quantify the distinct brain morphometry effects and latent dimensions across 8 neuropsychiatric CNVs. We analyzed T1-weighted MRI data from clinically and non-clinically ascertained CNV carriers (deletion/duplication) at the 1q21.1 (n = 39/28), 16p11.2 (n = 87/78), 22q11.2 (n = 75/30), and 15q11.2 (n = 72/76) loci as well as 1296 non-carriers (controls). Case-control contrasts of all examined genomic loci demonstrated effects on brain anatomy, with deletions and duplications showing mirror effects at the global and regional levels. Although CNVs mainly showed distinct brain patterns, principal component analysis (PCA) loaded subsets of CNVs on two latent brain dimensions, which explained 32 and 29% of the variance of the 8 Cohen’s d maps. The cingulate gyrus, insula, supplementary motor cortex, and cerebellum were identified by PCA and multi-view pattern learning as top regions contributing to latent dimension shared across subsets of CNVs. The large proportion of distinct CNV effects on brain morphology may explain the small neuroimaging effect sizes reported in polygenic psychiatric conditions. Nevertheless, latent gene brain morphology dimensions will help subgroup the rapidly expanding landscape of neuropsychiatric variants and dissect the heterogeneity of idiopathic conditions

    High Outcrossing Levels among Global Macadamia Cultivars: Implications for Nut Quality, Orchard Designs and Pollinator Management

    Global fruit and nut yields are affected by shortfalls in pollinator populations, and pollen limitation is most prevalent among tropical, bee-pollinated and self-incompatible plants. Macadamia is a subtropical, bee-pollinated crop in which some cultivars have been found to be highly outcrossing. We aimed to determine the extent of outcrossing and its effects on nut quality across a wide range of international macadamia cultivars in three countries. We sampled fruit from 19 macadamia cultivars across 23 sites in Australia, Brazil and South Africa. We used genotype-by-sequencing and MassARRAY methods to assign paternity to individual fruit and we assessed pollen-parent effects on nut quality. Macadamia was highly outcrossing, producing 80–100% of fruit by cross-pollination, at 17 of the 23 sites. Mixed mating (41–72% outcrossing) was identified at five sites, and low outcrossing (10%) was identified in one cultivar at one site where it was isolated from other flowering macadamia trees. Outcrossed fruit often had significantly better quality than selfed fruit, with 1.61–3.39 g higher nut-in-shell mass, 0.53–1.55 g higher kernel mass, 3.3–6.4% higher kernel recovery, and 3.0–3.5% higher oil concentration. The differences in kernel recovery equated to differences in value of USD 433–841 per ton of nut-in-shell at prices of USD 3000 per ton. In summary, macadamia cultivars were mostly highly outcrossing, and outcrossed nuts often had higher quality than selfed nuts. Growers should consider interplanting different cultivars more closely and distributing bee hives more widely to maximise cross-pollination, produce high yields, and optimise nut quality.</p

    Animal magnetism and curriculum history

    This article elaborates the impact that crises of authority provoked by animal magnetism, mesmerism, and hypnosis in the 19th century had for field formation in American education. Four layers of analysis elucidate how curriculum history’s repetitive focus on public school policy and classroom practice became possible. First, the article surveys external conditions of possibility for the enactment of compulsory public schooling. Second, “internal” conditions of possibility for the formation of educational objects (e.g., types of children) are documented via the processes of différance that were generated from within the experiences of confinement. Third, the article maps how these were interpenetrated by animal magnetic debates that were lustered and planished in education’s emerging field, including impact upon behavior management practices, the contouring of expertise and authority, the role of Will in intelligence testing and child development theories, and the redefinition of public and private. Last, the article examines implications for curriculum history, whether policy- or practice-oriented, especially around the question of influence, the theorization of child mind, and philosophies of Being
