Content of selected macroelements and microelements in acid casein and caseinates
Celem pracy było określenie zawartości wybranych makroelementów i mikroelementów w kazeinie
kwasowej i kazeinianach. Materiał badawczy stanowiła kazeina kwasowa otrzymana metodą tradycyjną oraz kazeiniany: sodu, wapnia i sodowo-wapniowy otrzymane metodą ekstruzji. Badane preparaty białkowe pochodziły z produkcji przemysłowej. W kazeinie kwasowej i kazeinianach oznaczono zawartość wody, związków mineralnych w postaci popiołu oraz zmierzono kwasowość czynną. Próbki preparatów białkowych mineralizowano na mokro, a następnie oznaczano w nich zawartość wybranych makroelementów (Ca, P, Mg, K, Na) i mikroelementów (Fe, Zn, Cu, Mn). Zawartość Ca, Mg, Fe, Zn, Cu i Mn oznaczono techniką płomieniowej spektrometrii absorpcji atomowej (płomień: acetylen – powietrze). K i Na oznaczano techniką emisyjną (płomień: acetylen – powietrze). Zawartość P oznaczano metodą kolorymetryczną. Wykazano, że produkowane ekstruzyjnie kazeiniany zawierały średnio mniej wody (5,63 ÷ 6,28 %) w porównaniu z otrzymaną tradycyjnie kazeiną kwasową (8,15 %). Kazeiniany zawierały średnio kilkakrotnie więcej popiołu (3,80 ÷ 4,54 %) w porównaniu z kazeiną kwasową (0,77 %). Badane preparaty białkowe różniły się istotnie pod względem pH. Średnia wartość pH kazeiny kwasowej wynosiła 5,10 a kazeinianu wapnia – 6,92. Kazeiniany, niezależnie od rodzaju, w porównaniu z kazeiną kwasową zawierały
znacznie więcej wapnia (odpowiednio: 1,196 ÷ 22,004 mg/g s.m. wobec 0,499 mg/g s.m.), potasu
(0,136 ÷ 0,262 mg/g s.m. wobec 0,005 mg/g s.m.) i sodu (0,279 ÷ 12,665 mg/g s.m. wobec 0,005 mg/g s.m.) oraz zbliżoną ilość fosforu (8,084 ÷ 8,177 mg/g s.m. wobec 8,205 mg/g s.m.) i magnezu (0,033 ÷ 0,040 mg/g s.m. wobec 0,039 mg/g s.m.), z wyjątkiem kazeinianu wapnia (0,201 mg/g s.m.). Największą zawartością wapnia oraz żywieniowo korzystnym stosunkiem jego zawartości względem fosforu (2,70 : 1) charakteryzował się kazeinian wapnia, co predysponuje ten preparat do stosowania w postaci składnika do wzbogacania żywności w pełnowartościowe białko i wapń. Pod względem mikroelementów kazeiniany, w porównaniu z kazeiną kwasową, zawierały więcej żelaza (odpowiednio: 76,234 ÷ 89,325 μg/g s.m. wobec 8,090 μg/g s.m.) i manganu (0,654 ÷ 6,068 μg/g s.m. wobec 0,155 μg/g s.m.). Kazeinian sodowo-wapniowy charakteryzował się mniejszą zawartością cynku (17,042 μg/g s.m.) w porównaniu z kazeiną kwasową (27,101 μg/g s.m.), kazeinianem sodu (29,508 μg/g s.m.) i kazeinianem wapnia (28,602 μg/g s.m.). Kazeinian wapnia zawierał więcej miedzi (1,801 μg/g s.m.) od kazeiny kwasowej (1,284 μg/g s.m.), kazeinianu sodu (1,234 μg/g s.m.) i od kazeinianu sodowo-wapniowego (1,120 μg/g s.m.).The objective of the research study was to determine the content of selected macroelements and microelements in acid casein and caseinates. The research materials comprised acid casein produced using a traditional method and sodium caseinate, calcium caseinate, and sodium-calcium caseinate produced using a method of extrusion. The analyzed protein preparations came from the industrial manufacturing. In the acid casein and caseinates, the water content was determined as were the mineral compounds in the form of ash, and the active acidity was measured. The samples of protein preparations were wet-mineralized, and, next, the contents of selected macroelements (Ca, P, Mg, K, Na) and microelements (Fe, Zn, Cu, Mn) were determined. The contents of Ca, Mg, Fe, Zn, Cu, and Mn were determined using a flame atomic absorption spectroscopy (flame: acetylene-air). The contents of K and Na were determined using an emission spectrometry (flame: acetylene-air). The content of P was determined using a colorimetric method. It was proved that the caseinates produced by extrusion had, on average, a lower water content (5.63 ÷ 6.28 %) than the acid casein produced by a traditional method (8.15 %). The caseinates contained a several times larger amount of ash (3.80 ÷ 4.54 %) compared to the acid casein (0.77 %). The protein preparations analyzed differed significantly in the pH value. The mean pH value of the acid casein was 5.10 and that of calcium caseinate: 6.92. Compared to the acid casein and regardless of their type, the caseinates
contained much more calcium (respectively: 1.196 ÷ 22.004 mg/g d.m. vs. 0.499 mg/g d.m.), potassium (0.136 ÷ 0.262 mg/g d.m. vs. 0.005 mg/g d.m.), and sodium (0.279 ÷ 12.665 mg/g d.m. vs. 0.005 mg /g d.m.), and the similar concentrations of phosphorus (8.084 ÷ 8.177 mg/g d.m. vs. 8.205 mg/g d.m.) and magnesium (0.033 ÷ 0.040 mg/g d.m. vs. 0.039 mg/g d.m.), except for the calcium caseinate. The calcium caseinate was characterized by the highest content of calcium and a nutritionally desirable calcium-tophosphorus ratio (2.70 : 1), which indicated that it could be used as a food additive to enrich food products with the high-quality protein and calcium. Regarding the microelements, the caseinates contained higher amounts of iron (respectively: 76.234 ÷ 89.325 μg/g d.m. vs. 8.090 μg/g d.m.) and manganese (0.654 ÷ 6.068 μg/g d.m. vs. 0.155 μg/g d.m.) compared to the acid casein. The sodium-calcium caseinate was characterized by lower contents of zinc (17.042 μg/g d.m.), sodium caseinate (29.508 μg/g d.m.), and calcium caseinate (28.602 μg/g d.m.) compared to the acid casein (27.101 μg/g d.m.). The calcium caseinate had more copper (1.801 μg/g d.m.) than the acid casein (1.284 μg/g d.m.), sodium caseinate (1.234 μg/g d.m.), and sodium-calcium caseinate (1.120 μg/g d.m.)
Zawartość wybranych makroelementów i mikroelementów w twarogu kwasowym o różnej zawartości tłuszczu
Przeprowadzono analizę zawartości wybranych makroelementów i mikroelementów
w twarogu kwasowym o różnej zawartości tłuszczu. Materiał badawczy stanowił twaróg
kwasowy chudy (0% tłuszczu), półtłusty (4% tłuszczu) i tłusty (8% tłuszczu), pochodzący od
jednego producenta. W badanym twarogu oznaczono zwartość suchej masy oraz popiołu
całkowitego. Próbki twarogu poddano mineralizacji na mokro, a następnie oznaczano w nich
zawartość wybranych makroelementów (P, Ca, Mg, K, Na) i mikroelementów (Zn, Fe, Cu, Mn).
Zawartość Ca, Mg, Zn, Fe, Cu i Mn oznaczono techniką płomieniowej spektrometrii absorpcji
atomowej (płomień acetylen – powietrze). K i Na oznaczano techniką emisyjną (płomień
acetylen – powietrze). Do oznaczenia zawartości P zastosowano metodę kolorymetryczną.
Badany twaróg kwasowy chudy, półtłusty i tłusty różnił się istotnie pod względem zawartości
suchej masy. Twaróg kwasowy chudy charakteryzował się istotnie większą zawartością popiołu
całkowitego niż twaróg kwasowy półtłusty i tłusty. W przeprowadzonych badaniach wykazano
istotne różnice między badanym twarogiem kwasowym w zawartości P, Ca, K, Na, Zn, Fe, Cu
i Mn, nie wykazano natomiast istotnych różnic w przypadku zawartości Mg. Bez względu na
zawartość tłuszczu w badanym twarogu kwasowym w największych ilościach spośród
oznaczonych makroelementów (około lub powyżej 100 mg · 100 g–1 produktu) występował:
fosfor, potas i wapń, natomiast spośród mikroelementów (w ilości powyżej 600 μg · 100 g–1
produktu) – cynk.The objective of this study was to determine the content of selected
macroelements and microelements in acid tvorogs with a different fat content. The
analyzed material consisted of skim (0% fat), medium-fat (4% fat) and full-fat (8% fat)
acid set tvorogs supplied by the same manufacturer. Dry matter content and total ash
content was determined in the analyzed cheeses. Tvorog samples were subjected to
wet mineralization, and the content of selected macroelements (P, Ca, Mg, K, Na) and
microelements (Zn, Fe, Cu, Mn) was determined. The content of Ca, Mg, Zn, Fe, Cu
and Mn were determined by atomic absorption spectrometry (flame acetylene-air).
K and Na content was determined by atomic emission spectrometry (flame acetylene-air).
The content of P were determined by colorimetric detection. The analyzed skim,
medium-fat and full-fat tvorogs differed significantly in their dry matter content. Skim
tvorog was characterized by a significantly higher total ash content than medium-fat
and full-fat tvorogs. The evaluated tvorogs differed significantly in their P, Ca, K, Na,
Zn, Fe, Cu and Mn content, but no significant differences in Mg content were observed.
In all analyzed tvorogs, the most abundant macroelements (about 100 mg · 100 g–1
or higher) were phosphorus, potassium and calcium, and the most abundant
microelement (600 μg · 100 g–1 or higher) was zinc
Assessment of selected minerals content in acid whey powder
Celem badań była ocena zawartości wybranych składników mineralnych w suszonej
serwatce kwasowej uzyskanej podczas produkcji twarogu. W serwatce kwasowej w
proszku oznaczono zawartość wody, białka, laktozy, tłuszczu, popiołu oraz kwasu mlekowego.
Celem oznaczenia składników mineralnych próbki badanej serwatki mineralizowano na mokro.
Ca, Mg, Zn, Fe, Mn i Cu oznaczano techniką płomieniowej spektrometrii absorpcji atomowej
(płomień acetylen – powietrze). K i Na oznaczano techniką emisyjną (płomień acetylen –
powietrze). P oznaczano metodą kolorymetryczną. Oceny zawartości oznaczonych składników
mineralnych dokonano, obliczając i analizując stopień pokrycia przez 10 g badanej serwatki
kwasowej w proszku uśrednionego zalecanego spożycia (w przypadku Ca, P, Mg, Zn, Fe, Mn,
Cu) lub wystarczającego spożycia (w przypadku K, Na), oddzielnie dla populacji kobiet i mężczyzn
w wieku 19-50 lat o umiarkowanej aktywności fizycznej. Badana serwatka kwasowa
w proszku zawierała średnio: 2,19% wody, 12,81% białka, 66,65% laktozy, 1,50% tłuszczu,
10,53% popiołu oraz 7,01% kwasu mlekowego. W suszonej serwatce kwasowej spośród oznaczonych
makroelementów w największych ilościach występował K (średnio 2252,90 mg/100g),
Ca (średnio 1845,16 mg/100g) i P (średnio 1031,30 mg/100g), natomiast z grupy mikroelementów
− Zn (średnio 6806,56 μg/100g). Według wyliczeń 10 g badanej serwatki kwasowej
w proszku pokrywa dzienne spożycie u kobiet i mężczyzn w wieku 19-50 lat o umiarkowanej
aktywności fizycznej w 18,45% na Ca, w 14,73% na P, odpowiednio w 5,12% i 3,94% na Mg,
w 4,79% na K, w 3,36% na Na, odpowiednio w 8,51% i 6,19% na Zn, oraz w stopniu mniejszym
niż 1% na Fe, Mn i Cu. Serwatka kwasowa potwarogowa w proszku zawiera Ca, P i Zn
w ilościach ważnych dla dziennego spożycia. Ca i P oraz Na i K w serwatce kwasowej występują
w korzystnych proporcjach z punktu widzenia żywienia i zdrowia.The aim of this study was the assessment of selected minerals content in acid whey powder produced during the making of tvorog. The content of moisture, total protein, lactose, fat, ash and lactic acid were determined in acid whey powder. Samples of acid whey powder were subjected to wet mineralization to determine the content of selected mineral nutrients. The content of Ca, Mg, Zn, Fe, Mn and Cu were determined by atomic absorption spectrometry (acetylene/air flame), K and Na content was determined by atomic emission spectrometry (acetylene/air flame), and P content was determined by colorimetry. The percentage of the Recommended Dietary Intakes of Ca, P, Mg, Zn, Fe, Mn and Cu, and the percentage of the Adequate Intake of K and Na provided by 10 g of acid whey powder were calculated and compared separately for moderately active males and females aged 19-50 years. The analyzed acid whey powder contained on average: 2,19% water, 12,81% protein, 66,65% lactose, 1,50% fat, 10,53% ash and 7,01% lactic acid. In the group of macroelements, K, Ca and P were present in the largest amounts (K – 2252,90 mg/100g, Ca – 1845,16 mg/100g, P – 1031,30 mg/100g on average). The most abundant microelement was Zn (6806,56 μg/100g on average). In a population of moderately active males and females aged 19-50 years, 10 g of acid whey powder supplied the following percentages of the recommended daily intakes of mineral nutrients: Ca – 18,45%, P – 14,73%, Mg – 5,12% and 3,94% respectively, K – 4,79%, Na – 3,36%, Zn – 8,51% and 6,19% respectively, Fe, Mn and Cu – less than 1%. Acid whey powder supplies a considerable amount of the recommended daily intakes of Ca, P and Zn, and it has nutritionally desirable Ca:P and Na:K ratios, which is an important consideration for human health
RNase H1 Regulates Mitochondrial Transcription and Translation via the Degradation of 7S RNA
RNase H1 is able to recognize DNA/RNA heteroduplexes and to degrade their RNA component. As a consequence, it has been implicated in different aspects of mtDNA replication such as primer formation, primer removal, and replication termination, and significant differences have been reported between control and mutant RNASEH1 skin fibroblasts from patients. However, neither mtDNA depletion nor the presence of deletions have been described in skin fibroblasts while still presenting signs of mitochondrial dysfunction (lower mitochondrial membrane potential, reduced oxygen consumption, slow growth in galactose). Here, we show that RNase H1 has an effect on mtDNA transcripts, most likely through the regulation of 7S RNA and other R-loops. The observed effect on both mitochondrial mRNAs and 16S rRNA results in decreased mitochondrial translation and subsequently mitochondrial dysfunction in cells carrying mutations in RNASEH1
Rheological properties, texture and color of ricotta available on the Polish market
Celem badań była analiza właściwości reologicznych, tekstury oraz barwy ricott
dostępnych na polskim rynku. Materiał badawczy stanowiły ricotty niedojrzewające pochodzące
od czterech producentów, zakupione w handlu detalicznym na terenie Olsztyna. W badanych
ricottach oznaczono zawartość suchej masy, białka ogółem, tłuszczu, laktozy, popiołu oraz zmierzono
kwasowość czynną. Do badań reologicznych wykorzystano reometr rotacyjny, a pomiary
prowadzono przy rosnącej szybkości ścinania w zakresie od 0,2 do 50 sˉ¹. W badaniach tekstury
prowadzonych przy użyciu teksturometru wyznaczono twardość, adhezyjność, spójność, sprężystość
i gumiastość ricott. Przy użyciu spektrofotometru mierzono parametry barwy L*, a*, b*, a następnie
wyznaczono nasycenie barwy C*. Między analizowanymi ricottami stwierdzono istotne różnice
w składzie chemicznym oraz kwasowości czynnej. Lepkość ricott spadała ze wzrostem szybkości ścinania.
W przypadku ricott od dwóch producentów zaobserwowano wzrost naprężenia ścinającego przy
większych szybkościach ścinania. Stwierdzono istotnie dodatnią korelację liniową pomiędzy zawartością
suchej masy i zawartością tłuszczu w suchej masie a naprężeniem ścinającym i lepkością przy maksymalnej
szybkości ścinania. Między badanymi ricottami występowały istotne różnice w twardości, adhezyjności
i gumiastości. Produkty o większej adhezyjności charakteryzowały się większym stosunkiem zawartości
tłuszczu do białka. Badane ricotty wykazywały istotne różnice w parametrach barwy L*, a*, b* i C*.
Stwierdzono istotnie ujemną korelację liniową pomiędzy zawartością białka w suchej masie a parametrami
a*, b*, C*, oraz istotnie dodatnią korelację liniową pomiędzy zawartością tłuszczu w suchej masie
a parametrem a*.The aim of this study was to analyze the rheological properties, texture and color of ricotta
available on the Polish market. The experiment was performed on fresh (unripened) ricotta supplied
by four manufacturers, purchased in retail in the city of Olsztyn, Poland. Ricotta samples were analyzed to
determine their content of dry matter, total protein, fat, lactose and ash. Active acidity was also measured.
The rheological properties of ricotta were determined with the use of a rotary rheometer. Measurements
were carried out at increasing shear rates, from 0.2 to 50 sˉ¹. Texture, including hardness, adhesiveness,
cohesiveness, springiness and gumminess, was evaluated with a texture analyzer. Color parameters L*,
a*, b* were measured with a spectrophotometer, and chroma (C*) was determined. The evaluated ricotta
products were characterized by significant differences in chemical composition and active acidity. Their
viscosity decreased with increasing shear rates. In ricotta from two manufacturers, shear stress increased
at higher shear rates. A significant positive linear correlation was found between dry matter content and
fat in dry matter content vs. shear stress and viscosity at the maximum shear rate. Significant differences
in hardness, adhesiveness and gumminess were noted between the analyzed ricotta samples. The products
characterized by higher adhesiveness had a higher protein to fat ratio. Significant differences were also
observed between the analyzed products with regard to the values of color parameters L*, a*, b* and C*.
A significant negative linear correlation was found between protein in dry matter content vs. color parameters
a*, b* and C*, whereas a significant positive linear correlation was noted between fat in dry matter
content and parameter a*
