    Defining ovine dermal papilla cell markers and identifying key signaling pathways regulating its intrinsic properties

    Dermal papilla cell (DPC), one of the key cell types during hair follicle development and regeneration, specifies hair size, shape and cycling. It is also an important in vitro screening model for hair growth. Although some characteristics of DPCs, such as agglutinative growth and marker genes, have been studied in mice and humans, the intrinsic properties of ovine DPCs and the regulatory mechanism of the intrinsic properties during continued culture in vitro remained unknown. In this study, based on our previous single-cell transcriptome sequencing on sheep lambskin, we verified SOX18 and PDGFRA as the novel marker genes of ovine DPCs through immunofluorescence staining on skin sections and cultured DPCs. Using continued cell culture and alkaline phosphatase staining, we found that different from mice and humans, ovine DPCs exhibit particularly robust and stable aggregation with unbated alkaline phosphatase activity till 30 passages during continued culture in vitro. Also, we found that the expression of some marker genes and the activity of Wnt/β-catenin signaling differ between early passaged DPCs and multiple passaged DPCs. Further, using Wnt/β-catenin agonist and antagonist, we demonstrated that Wnt/β-catenin signaling could regulate cell aggregation and alkaline phosphatase activity of ovine DPCs through regulating FGF and IGF signaling. This study provides the basis for isolating ovine DPCs and defines their intrinsic properties, which contribute to improving wool performance and medicine of hair regeneration

    Can 9q34.2 rs633862 polymorphism predict survival in epithelial ovarian cancer?

    Objective Our previous genome-wide association study (GWAS) identified that the ABO rs633862 variant in chromosome 9q34.2 was associated with the risk of epithelial ovarian cancer (EOC) in Chinese Han women. The aim of the present study was to evaluate its prognostic effect on EOC. Methods A total of 669 EOC patients were enrolled for the genotyping of rs633862 variant in 9q34.2. We used Kaplan–Meier survival curves, univariate and multivariate Cox proportional hazard models to evaluate the association of rs633862 with overall survival (OS) in EOC patients. Results We found that rs633862 variant AG/GG genotypes were significantly associated with a longer OS by using univariate Cox proportional hazards regression analysis, compared with the rs633862 AA genotype (HR = 0.69, 95% CI [0.49–0.98], p = 0.035), albeit with a boardline significance in the multivariate analysis. Similar findings were observed in the subgroup of high-grade serous ovarian carcinoma. Further expression quantitative trait loci (eQTL) analysis indicated that the rs633862 AA genotype was associated with an increased level of ABO mRNA expression (p = 1.8 × 10−11). Conclusions Supplementary to the previous GWAS, our study provides additional evidence on the prognostic value of the 9q34.2 rs633862 variant in EOC patients, and this variant may function by regulating the ABO mRNA expression

    Study on Water Purification Effect and Operation Parameters of Various Units of Wastewater Circulation

    The discharge of wastewater from aquaculture ponds causes a certain degree of damage to the environment. It is necessary to continuously improve the treatment efficiency of wastewater treatment devices. The purpose of this study is to obtain an optimal ratio of wastewater circulation devices in order to obtain the best operating parameters and to reduce the discharge of polluted water. We constructed an experimental wastewater circulation device consisting of three units. The primary unit contained modified attapulgite (Al@TCAP-N), volcanic stone, and activated carbon for precipitation. The secondary and tertiary units used biological methods to enhance removal rates of nitrogen and phosphorus. Water quality indicators of total phosphorus (TP), total nitrogen (TN), ammonia (NH3-N), permanganate (CODMn), and total suspended solids (TSS) were detected. Water quality was tested under different matching ratios for three units of different hydraulic retention time (HRT) and load Results showed that the removal rate of TP, TN, NH3-N, and TSS reached 20–60%, 20%, 30–70%, and 10–80%, respectively. The average reduction efficiencies of secondary module chlorella and filler on TP, TN, NH3-N, CODMn, and TSS were 56.88%, 30.09%, 0.43%, 46.15%, and 53.70%, respectively. The best removal rate can be achieved when the matching ratio of each unit becomes 2:1:1 and the hydraulic retention time is maintained within 2 h in the high-concentration load. Finally, the average removal rates of TP, TN, NH3-N, and TSS reached 58.87%, 15.96%, 33.99%, and 28.89%, respectively. The second unit obtained the enhanced removal effect in this wastewater treatment system when adding microorganisms and activated sludge

    Supporting workflow in a course management system

    CMS, a secure and scalable web-based course management system developed by the Cornell University Computer Science Department, helps manage the workflow associated with running a course. Our goal in designing the system was to simplify, streamline, and automate all workflow aspects, such as course creation and importing students into it, student group management, assignment submission, assignment of graders, grading, regrade requests, and preparation of final grades. In contrast, other course management systems that we are aware of provide only specialized solutions for specific components, such as grading. This system is increasingly widely used for course management at Cornell University. The system was designed to support large courses with low administrative overhead. In this paper we describe the design of the system and the features that were found to be useful, and articulate its design principles

    Barriers to community-based drug dependence treatment: implications for police roles, collaborations and performance indicators

    ntroduction: Worldwide, people who use drugs (PWUD) are among the populations at highest risk for HIV infection. In China, PWUD are primarily sentenced to compulsory detainment centres, in which access to healthcare, including HIV treatment and prevention services, is limited or non-existent. In 2008, China's 2008 Anti-Drug Law encouraged the development and use of community-based drug dependence rehabilitation, yet there is limited evidence evaluating the efficacy and challenges of this model in China. In this study, we explore these challenges and describe how cooperation between law enforcement and health departments can meet the needs of PWUD. Methods: In 2015, we conducted semi-structured, in-depth interviews with all four staff members and 16 clients of the Ping An Centre No. 1 for community-based drug treatment, three local police officers and three officials from the local Centre for Disease Control. Interviews explored obstacles in implementing community-based drug dependence treatment and efforts to resolve these difficulties. Transcripts were coded and analyzed with qualitative data analysis software (MAXQDA 11). Results: We identified three challenges to community-based drug treatment at the Ping An Centre No. 1: (1) suboptimal coordination among parties involved, (2) a divergence in attitudes towards PWUD and harm reduction between law enforcement and health officials and (3) conflicting performance targets for police and health officials that undermine the shared goal of treatment. We also identified the take-home methadone maintenance treatment model at the Ping An Centre No. 1 as an example of an early successful collaboration between the police, the health department and PWUD. Conclusions: To overcome barriers to effective community-based drug treatment, we recommend aligning the goals of law enforcement and public health agencies towards health-based performance indicators. Furthermore, tensions between PWUD and police need to be addressed and trust between them fostered, using community-based treatment centres as mediators. The preliminary success of the take-home methadone maintenance treatment pilot can serve as an example of how collaboration with the police and other government agencies can meet the needs of PWUD and contribute to the success of community-based treatment
