    Constructing Confidence Intervals for Effect Sizes in ANOVA Designs

    A confidence interval for effect sizes provides a range of plausible population effect sizes (ES) that are consistent with data. This article defines an ES as a standardized linear contrast of means. The noncentral method, Bonett’s method, and the bias-corrected and accelerated bootstrap method are illustrated for constructing the confidence interval for such an effect size. Results obtained from the three methods are discussed and interpretations of results are offered

    AMPK- mediated formation of stress granules is required for dietary restriction- induced longevity in Caenorhabditis elegans

    Stress granules (SGs) are nonmembranous organelles that are dynamically assembled and disassembled in response to various stressors. Under stressed conditions, polyadenylated mRNAs and translation factors are sequestrated in SGs to promote global repression of protein synthesis. It has been previously demonstrated that SG formation enhances cell survival and stress resistance. However, the physiological role of SGs in organismal aging and longevity regulation remains unclear. In this study, we used TIAR- 1::GFP and GTBP- 1::GFP as markers to monitor the formation of SGs in Caenorhabditis elegans. We found that, in addition to acute heat stress, SG formation could also be triggered by dietary changes, such as starvation and dietary restriction (DR). We found that HSF- 1 is required for the SG formation in response to acute heat shock and starvation but not DR, whereas the AMPK- eEF2K signaling is required for starvation and DR- induced SG formation but not heat shock. Moreover, our data suggest that this AMPK- eEF2K pathway- mediated SG formation is required for lifespan extension by DR, but dispensable for the longevity by reduced insulin/IGF- 1 signaling. Collectively, our findings unveil a novel role of SG formation in DR- induced longevity.In addition to heat stress, starvation and dietary restriction (DR) can activate stress granule (SG) formation in Caenorhabditis elegans. HSF- 1 and AMPK are two key regulators for the SG formations. HSF- 1 is required for the SG formation in response to acute heat shock and starvation but not DR, whereas the AMPK- eEF2K pathway is required for starvation and DR- induced SG formation but not heat shock.     In the title complex, [Cu2(CN)2(C6H7N)]n, there are two copper atoms with different coordination environments. One Cu atom (Cu1) is linked to the two cyanide ligands, one N atom from a pyridine ring while the other (Cu2) is coordinated by the two cyanide ligands in a slightly distorted tetra­hedral geometry and linked to Cu1, forming a triangular coordination environment. The Cu atoms are bridged by bidentate cyanide ligands, forming an infinite Cu–CN chain. One cyanide ligand is equally disordered over two sets of sites, exchanging C and N atoms coordinated to both metal atoms. However, one cyanide group is not disordered and it coordinates to Cu1 via the N atom whereas its C-atom counterpart coordinates Cu2. The 3-methyl­pyridine (3MP) ligand coordinates through the N atom to Cu1 as a terminal ligand, which originates from decyanation of 3-pyridyl­acetonitrile under hydro­thermal conditions.     Blood-Oxygenation-Level-Dependent-(BOLD-) Based R2′ MRI Study in Monkey Model of Reversible Middle Cerebral Artery Occlusion

    Objective. To investigate the value of BOLD-based reversible transverse relaxation rate (R2′) MRI in detecting ischemic penumbra (IP) in a monkey model of reversible middle cerebral artery occlusion (MCAO) and time evolution of relative R2′ (rR2′) in infarcted core, IP, and oligemia. Materials and Methods. 6 monkeys were used to make MCAO by the microcatheter method. MR scans were performed at 0 h (1 h after MCAO), 1 h, 3 h, 6 h, 12 h, 24 h, and 48 h after reperfusion. R2′ was calculated using quantitative T2 and T2* maps. Ischemic area was subdivided into infracted core, IP and oligemia. rR2′ was calculated respectively. Results. Reversible MCAO model for 4/6 monkeys was made successfully. rR2′ values were significantly different at each time point, being highest in oligemia followed by IP and infarcted core (P < .05). With reperfusion time evolution, rR2′ in infarcted core showed a decreased trend: sharply decreased within 6 hours and maintained at 0 during 6–48 hours (P < .05). rR2′ values in IP and oligemia showed similar increased trend: sharply increased within 6 hours, maintained a plateau during 6–24 hours, and slightly increased until 48 hours. Conclusion. BOLD-based R2′ MRI can be used to describe changes of cerebral oxygen extract in acute ischemic stroke, and it can provide additional information in detecting IP. The time evolution rR2′ in infarcted core, IP, and oligemia is in accordance with the underlying pathophysiology

    Algorithms for Assessing Intervention Effects in Single-Case Studies

    Free web-based resources or popular software to assess six data features recommended by the What Works Clearinghouse: Procedures and Standards Handbook (IES, 2013 February) to determine intervention effects in a single-case study (Lambert, Cartledge, Heward, & Lo, 2006) are demonstrated. Lambert et al. (2006) employed a reversal (or ABAB) design and visual inspection to investigate the effectiveness of the report-card treatment in reducing disruptive behaviors in students. In our demonstration, we assessed each of the six data features separately; then integrated six assessments into one comprehensive analysis of the intervention effect. A simple approach to the determination of intervention effects illustrates how researchers and practitioners can be empowered to interpret data comprehensively and formulate evidence-based conclusions logically from well-designed and well-executed single-case studies

    Risperidone-Associated Neuroleptic Malignant Syndrome in an Inpatient With Schizophrenia, With Successful Rechallenge and 3 Year Follow-Up

    Neuroleptic malignant syndrome (NMS) is rare but one of the most serious adverse effects of antipsychotics. Here, we report a case of risperidone-associated NMS in which a successful rechallenge of risperidone was observed with a positive follow-up. A 47-year-old female with schizophrenia was treated with risperidone 4 mg/d for 8 months in 2009 and was admitted to our hospital in 2015 owing to violent behavior under persecutory delusions. Risperidone 2 mg/d was initiated and increased to 4 mg/d 54 days later. Further, long-acting injectable (LAI) risperidone 25 mg per 2 weeks was added on hospital day 15. On hospital day 116, NMS occurred and thus we discontinued all antipsychotics including LAI risperidone, then NMS improved. We resumed LAI risperidone 25 mg per 2 weeks on hospital day 148, thus we waited for 22 days before re-starting the drug treatment. She was discharged on hospital day 371, then switched to LAI paliperidone 150 mg per 4 weeks 2 months later. At the time of a follow-up 3 years later, NMS had not reoccurred. This case reports on an unusual presentation of NMS in which no hyperthermia was observed. Furthermore, this case indicated that NMS may occur in a dose-dependent manner. In conclusion, this case reported important information for clinicians with regard to antipsychotic drug rechallenges and proper dosing of APs to avoid or reverse NMS

    Construction and Evaluation of Chloroplast Expression Vectors in Higher Plants

    Plastid transformation has a number of advantages in comparison with nuclear transformation. Currently, only tobacco (Nicotiana tabacum) is routinely used in plastid transformation. Here we constructed a series of chloroplast expression vectors specific for spinach (pCEV1), tomato (pCEV2 and pCEV3), and N. benthamiana (pCEV4). Selection marker aminoglycoside 3′-adenyltransferase (aadA) conferring spectinomycin resistance was used in pCEV1, pCEV2, and pCEV4, while selection marker neomycin phosphotransferase II (nptII) was used in pCEV3. The expression cassette in these vectors was integrated in the intergenic spacer between trnI and trnA of plastid genome via homologous recombination. Several transgenes, including a reporter gene encoding GFP:GUS fusion protein and genes from tomato (lycopene b-cyclase, z-carotene desaturase) and bamboo mosaic virus satellite RNA (encoding coat protein CP20), were independently cloned into some of these vectors. Transient GUS expression was detected in spinach leaves bombarded by pCEV1/GFP-GUS. Functional expression of selection markers aadA and nptII was demonstrated for spinach, tomato, and N. benthamiana. Seedling assay from T0 self-pollinated plant of transplastomic N. benthamiana confirmed maternal inheritance of transgenes, and genomic PCR analysis confirmed integration of transgenic expression cassette into the plastid genome of N. benthamiana.
