PrestoBlue for Quantifying Rat Islet Viability and Count
Rat pancreatic islets obtained from the Joslin Diabetes Center were size filtered to 70-140 um diameter range. Islets were handpicked and placed in a 24-well culture plate. Each well contained 880 uL of RPMI (with 0.125% BSA). Islets were collected in a total volume of 200 uL and added to the well. 120 uL of PrestoBlue (Thermo) was added to each well and incubated for either 1 or 2 hours. 100 uL was collected and the fluorescence signal was measured in a clear bottom black-walled 96 well plate. N = 4, error bars represent standard deviation. <div><br></div><div>All work was conducted at the Massachusetts Institute of Technology, Cambridge, MA.</div
