    Antioxidative and antibacterial activities of phenolic compounds from Ficus sur Forssk. and Ficus sycomorus L. (Moraceae) : potential for sickle cell disease treatment in Burkina Faso

    Ficus sur and Ficus sycomorus, two medicinal species used in Burkinabe traditional medicine for the treatment of sickle cell disease were investigated. The determination of total phenolic and tannins contents, using the method of Folin-Ciocalteu (FCR), indicate values significantly (p <0.05) higher in extracts of Ficus sycomorus compared to those of Ficus sur, with respective values of 336.80 ± 0.80 mg TAE/g of extract and 203.74 ± 0.9 mg TAE/g of extract against 247.00 ± 0.52 mg TAE/g of extract and 120.8 ± 0.83 mg TAE/g of extract. Similarly, using the method of 1,1-diphenyl-2-picrylhydrazyl (DPPH), the results of the analysis indicate that extracts of Ficus sycomorus produce the highest antiradical activity with IC50 value of 9.60 ± 0.02 μg/mL against 31.83 ± 0.55 μg/ mL for Ficus sur. The IC50 value of quercetin, used as reference antioxidant, was of 4.6 ± 0.08 μg/mL. The latex of Ficus sycomorus presented the lowest minimum inhibitory concentrations (MIC) against Staphylococcus aureus and Escherichia coli, with respective values of 0.13 mg/mL and 0.25 mg/mL. The difference in phenolic content could explain the difference in biological activity between the two Ficus species.Keywords: drepanocitosis, radical scavenging, tannins, flavonoids, medicinal plant

    Hepatoprotective activity of aqueous extract of Balanites aegyptiaca L. Delile (Balanitaceae) roots bark

    Balanites aegyptiaca (L.) Del (Balanitaceae) is traditionally used for the treatment of various ailments such as syphilis, jaundice and liver disorders, epilepsy, ... This study was designed to evaluate acute toxicity and hepatoprotective effect of aqueous extract of Balanites aegyptiaca on CCl4 induced hepatotoxicity in rats.Methods: Acute toxicity was assessed with the extract at a dose of 2000 mg / kg bw. The extract at doses of 25, 50 and 100 mg / kg b.w. was orally administered respectively to CC14-induced hepatotoxicity (0.5 ml / kg) animals. Silymarin (100 mg / kg) was given as a reference. Biochemical parameters such as ALT, AST, PT, ALB and ALP were assayed as well as enzymatic antioxidant activities SOD, CAT and MDA. Nitrogen monoxide (NO) involved in inflammation was also measured.Results: Activities of liver marker enzymes, ALT, AST and ALP, total protein, albumin and showed a significant hepatoprotective effect. Regarding antioxidant enzymatic activities in vivo (SOD, CAT and MDA) of aqueous extract exhibited a significant effect showing increasing levels of SOD, CAT and reducing malondialdehyde (MDA) levels. The production of NO is significantly reduced compared to the batch intoxicated by CCl4.Conclusion: Balanites aegyptiaca is endowed with hepatoprotective properties that can be attributed to antioxidant potential which could justify its use in traditional medicine in liver disorders

    Antimicrobial and antioxidant properties of methanol extract, fractions and compounds from the stem bark of Entada abyssinica Stend ex A. Satabie

    <p>Abstract</p> <p>Background</p> <p>The aim of this study was to evaluate the antimicrobial and antioxidant activities of the methanol extract, fractions and isolated compounds from <it>Entada abyssinica </it>stem bark, plant used traditionally against gastrointestinal infections.</p> <p>Methods</p> <p>The methanol extract of <it>E. abyssinica </it>stem bark was pre-dissolved in a mixture of methanol and water, and then partitioned between <it>n</it>-hexane, ethyl acetate and <it>n</it>-butanol. The ethyl acetate portion was fractionated by column chromatography and the structures of isolated compounds elucidated by analysis of spectroscopic data and comparison with literature data. Antimicrobial activity was assayed by broth microdilution techniques on bacteria and yeasts. The antioxidant activity was determined by DPPH radical scavenging method.</p> <p>Results</p> <p>Four known compounds [(5<it>S</it>,6<it>R</it>,8a<it>R</it>)-5-(carboxymethyl)-3,4,4a,5,6,7,8,8a-octahydro-5,6,8a-trimethylnaphthalenecarboxylic acid (<b>1</b>), methyl 3,4,5-trihydroxybenzoate (<b>2</b>), benzene-1,2,3-triol (<b>3</b>) and 2,3-dihydroxypropyltriacontanoate (<b>4</b>)] were isolated. Compared to the methanol extract, fractionation increased the antibacterial activities of the <it>n</it>-hexane and ethyl acetate fractions, while the antifungal activities increased in ethyl acetate, <it>n</it>-butanol and aqueous residue fractions. The isolated compounds were generally more active on bacteria (9.7 to 156.2 μg/ml) than yeasts (78.1 to 312.5 μg/ml). Apart from compound <b>1</b>, the three others displayed DPPH<sup>· </sup>scavenging activity (RSa), with RSa<sub>50 </sub>values of 1.45 and 1.60 μg/ml.</p> <p>Conclusion</p> <p>The results obtained from this study support the ethnomedicinal use of <it>E. abyssinica </it>in the treatment of gastrointestinal infections and the isolated compounds could be useful in the standardisation of antimicrobial phytomedicine from this plant.</p

    Étude des activités anti-inflammatoire, analgésique et antipyrétique des décoctés aqueux des feuilles et des racines de Pterocarpus erinaceus Poir. (Fabaceae)

    International audiencePterocarpus erinaceus Poir. (Fabaceae) est une plante médicinale utilisée dans la prise en charge de nombreuses pathologies dont les maladies à composante inflammatoire. L'objectif du travail était d'étudier les effets anti-inflammatoire, analgésique et antipyrétique des décoctés aqueux des feuilles et des écorces de racines de Pterocarpus erinaceus afin de valoriser son utilisation en médecine traditionnelle. Aux doses de 200, 400 et 600 mg/kg, les décoctés aqueux des feuilles et des écorces de racines ont réduit significativement l'œdème de la patte de la souris induit par la carragénine et les contractions abdominales produites par l'injection de l'acide acétique chez la souris. Les deux extraits ont inhibé les deux phases de douleur produites par la solution de formaldéhyde. L'hyperthermie chez la souris induite par la levure de bière a été réduite par les deux décoctés aqueux, cet effet reste cependant plus marqué pour les feuilles que les racines. Le criblage phytochimique des extraits a permis de mettre en évidence la présence de tanins et polyphénols, de flavonoïdes, de saponosides, de triterpènes et stéroïdes, qui pourraient être responsables des propriétés pharmacologiques

    Countrywide survey of plants used for liver disease management by traditional healers in Burkina Faso

    Liver disease is highly prevalent in Africa, especially in the western African country Burkina Faso, due to the presence of multiple biological and chemical aggressors of the liver. Furthermore, diagnosis and appropriate care for liver disease are uneven and usually insufficient. This drives local communities to turn to folk medicine based on medicinal plants from healers. Small scale, ethnopharmacological studies on reputed hepatoprotective plants have been carried out in defined regions worldwide, but so far, no study has been carried out on a countrywide scale. Therefore, we have explored traditional healers' practices in all thirteen regions of Burkina Faso. We interviewed 575 healers and we compiled a database with 2,006 plant entries. Here, we report results on liver nosology, liver pathologies, medicinal plants used for liver disease, and traditional practices through the lens of Burkinabe healers. Our goal was to give a full inventory of medicinal plants used to treat liver disease and to determine if there was consensus on the use of specific plants for specific symptoms. Analysis of the medicinal plants in use across the whole country provides local communities with a wider evidence base to determine which plants may be more effective in treating liver disease and could provide the scientific community, with a shortlist of plants suitable for chemical and pharmacological investigation to validate the plants' therapeutic role

    Pharmacological properties and related constituents of stem bark of Pterocarpus erinaceus Poir. (Fabaceae)

    International audienceObjective To screen methanol and dichloromethane extracts of stem bark of Pterocarpus erinaceus for anti-inflammatory, analgesic, in vitro antioxidant activities and phytochemical analysis. Methods Anti-inflammatory activity was determined by using carrageenan induced-edema of mice paw and croton oil-induced edema of mice ear; analgesic effect was evaluated using acetic acid-induced writhing. Phytochemical screening of extracts was performed by thin layer chromatography. The chromatographic fractionation led to the isolation of main active components as friedelin, lupeol and epicathechin. The structures were established by TLC and nuclear magnetic resonance studies. Results Both methanol and dichloromethane extracts, friedelin, lupeol and epicatechin showed a significant anti-inflammatory effect using croton oil induced-ear edema. Furthermore, the action of dichloromethane extract was more important. At the doses of 100 and 200 mg/kg, the methanol extract was able to reduce the carrageenan induced-hind paw edema, while at the doses of 100, 200 and 400 mg/kg, it showed an important analgesic effect against writhing induced by acetic acid injection of 38.8%, 68.0% and 74.3%, respectively. Antioxidative properties of methanol extract and its dichloromethane and ethyl acetate fractions were assessed by using the 1,1-diphenyl-2-picrylhydrazyl method. The methanol extract showed the stronger radical scavenging activity than dichloromethane and ethyl acetate fractions, with an antiradical power of 5, 3.5 and 2 respectively. The main components isolated from these extracts as friedelin, lupeol and epicathechin were responsible of these activities. Conclusions The results suggest that the stem bark extracts of Pterocarpus erinaceus possessed important anti-inflammatory, analgesic activities and strong antioxidant properties, therefore, they could be used as natural potential ingredients for pharma ceutical industry

    Lack of robust evidence for a Wolbachia infection in Anopheles gambiae from Burkina Faso

    The endosymbiont Wolbachia can have major effects on the reproductive fitness, and vectorial capacity of host insects and may provide new avenues to control mosquito‐borne pathogens. Anopheles gambiae s.l is the major vector of malaria in Africa but the use of Wolbachia in this species has been limited by challenges in establishing stable transinfected lines and uncertainty around native infections. High frequencies of infection of Wolbachia have been previously reported in An. gambiae collected from the Valle du Kou region of Burkina Faso in 2011 and 2014. Here, we re‐evaluated the occurrence of Wolbachia in natural samples, collected from Valle du Kou over a 12‐year time span, and in addition, expanded sampling to other sites in Burkina Faso. Our results showed that, in contrast to earlier reports, Wolbachia is present at an extremely low prevalence in natural population of An. gambiae. From 5341 samples analysed, only 29 were positive for Wolbachia by nested PCR representing 0.54% of prevalence. No positive samples were found with regular PCR. Phylogenetic analysis of 16S rRNA gene amplicons clustered across supergroup B, with some having similarity to sequences previously found in Anopheles from Burkina Faso. However, we cannot discount the possibility that the amplicon positive samples we detected were due to environmental contamination or were false positives. Regardless, the lack of a prominent native infection in An. gambiae s.l. is encouraging for applications utilizing Wolbachia transinfected mosquitoes for malaria control
