    Familial thymic aplasia - Attempted reconstitution with fetal thymus in a Millipore diffusion chamber

    A 10-week-old female infant with familial congenital thymic aplasia without delayed hypersensitivity to common skin-test antigens underwent fetal-thymus implantation. Six hours after the implantation of a fetal thymus enclosed in a Millipore chamber phytohemagglutinin responsiveness was demonstrable in the patient's peripheral lymphocytes. The infant's death of aspiration pneumonia nine days after implantation did not allow evaluation of the extent of the immunologic reconstitution.     Convergence calls: multimedia storytelling at British news websites

    This article uses qualitative interviews with senior editors and managers from a selection of the UK's national online news providers to describe and analyse their current experimentation with multimedia and video storytelling. The results show that, in a period of declining newspaper readership and TV news viewing, editors are keen to embrace new technologies, which are seen as being part of the future of news. At the same time, text is still reported to be the cornerstone for news websites, leading to changes in the grammar and function of news video when used online. The economic rationale for convergence is examined and the article investigates the partnerships sites have entered into in order to be able to serve their audience with video content. In-house video is complementing syndicated content, and the authors examine the resulting developments in newsroom training and recruitment practices. The article provides journalism and interactive media scholars with case studies on the changes taking place in newsrooms as a result of the shift towards multimedia, multiplatform news consumption

    This report represents a scientific and working clinical consensus statement on seizure management in dogs based on current literature and clinical expertise. The goal was to establish guidelines for a predetermined, concise, and logical sequential approach to chronic seizure management starting with seizure identification and diagnosis (not included in this report), reviewing decision‐making, treatment strategies, focusing on issues related to chronic antiepileptic drug treatment response and monitoring, and guidelines to enhance patient response and quality of life. Ultimately, we hope to provide a foundation for ongoing and future clinical epilepsy research in veterinary medicine

    A gas chromatograph has been assembled which provides computer control of sample injection, column temperature, and flow rate, plus direct computer readout of inlet pressure, mass flow rate, and detector response. Data processing yields, in real-time, a standard deviation of less than 0.05% in retention time, which is comparable to previous results obtained using an off-line computer. However, corrected retention volumes determined in real-time had a standard deviation of about 0.4% which reflected primarily the uncertainty in flow measurement

    AbstractBackground & Aims: Ethanol causes both tolerance and sensitization of Kupffer cells. This study was designed to evaluate temporal effects of ethanol in an attempt to understand this paradox. Methods: Rats were given ethanol (4 g/kg body wt) intragastrically, and Kupffer cells were isolated 0–48 hours later. After addition of lipopolysaccharide (LPS), intracellular calcium concentration ([Ca2+]i) was measured using a microspectrofluorometer with the fluorescent indicator fura-2, and tumor necrosis factor α (TNF-α) was measured by enzyme-linked immunosorbent assay. CD14 was evaluated by Western and Northern analysis. Results: Two hours after ethanol administration, the LPS-induced increase in [Ca2+]i and TNF-α release by Kupffer cells was diminished by 50%, and these parameters were reciprocally enhanced twofold at 24 hours. Sterilization of the gut with antibiotics blocked all effects of ethanol on [Ca2+]i and TNF-α release completely. Twenty-four hours after ethanol, CD14 in Kupffer cells was elevated about fivefold. Conclusions: Kupffer cells isolated from rats early after ethanol exhibited tolerance to LPS, whereas sensitization was observed later. It is likely that both of these phenomena are caused by gut-derived endotoxin and that sensitization in Kupffer cells is caused by increases in CD14.GASTROENTEROLOGY 1998;115:443-45

    Patterns of regulatory activity across diverse human cell types predict tissue identity, transcription factor binding, and long-range interactions

    Regulatory elements recruit transcription factors that modulate gene expression distinctly across cell types, but the relationships among these remains elusive. To address this, we analyzed matched DNase-seq and gene expression data for 112 human samples representing 72 cell types. We first defined more than 1800 clusters of DNase I hypersensitive sites (DHSs) with similar tissue specificity of DNase-seq signal patterns. We then used these to uncover distinct associations between DHSs and promoters, CpG islands, conserved elements, and transcription factor motif enrichment. Motif analysis within clusters identified known and novel motifs in cell-type-specific and ubiquitous regulatory elements and supports a role for AP-1 regulating open chromatin. We developed a classifier that accurately predicts cell-type lineage based on only 43 DHSs and evaluated the tissue of origin for cancer cell types. A similar classifier identified three sex-specific loci on the X chromosome, including the XIST lincRNA locus. By correlating DNase I signal and gene expression, we predicted regulated genes for more than 500K DHSs. Finally, we introduce a web resource to enable researchers to use these results to explore these regulatory patterns and better understand how expression is modulated within and across human cell types
