    The Effect of Pluchea indica (L.) Less. Tea on Adipogenesis in 3T3-L1 Adipocytes and Lipase Activity

    Obesity and hyperlipidemia are a major problem in the world. Pluchea indica (L.) Less. tea (PIT) is a beverage that has various indications. This study focused on the effect of the PIT on inhibiting adipogenesis of 3T3-L1 cells and pancreatic lipase enzyme activity. The viability of 3T3-L1 cells was not significantly decreased after exposure to 200 to 1000 μg mL−1 PIT compared to controls (p < 0.05). The PIT at 750 to 1000 μg mL−1 exhibited a significantly reduced lipid accumulation compared to the control (p < 0.05). The inhibitory effects of the PIT at 250 to 1000 μg mL−1 on lipase activity were significantly increased compared to control (p < 0.05). The FTIR results showed that the integrated areas of lipids, proteins, nucleic acids, glycogen, and carbohydrates of the PIT-treated 3T3-L1 adipocytes were significantly lower than the untreated 3T3-L1 adipocytes (p < 0.05). These findings may indicate that the PIT is not only capable of inhibiting lipids and carbohydrate accumulation in adipocytes but also has a potential to inhibit pancreatic lipase activity. So, the PIT may be further developed to the novel lipid-lowering herbal supplement for the management of overweight or obesity

    Intracellular ROS Scavenging and Anti-Inflammatory Activities of Oroxylum indicum Kurz (L.) Extract in LPS plus IFN-gamma-Activated RAW264.7 Macrophages

    Oroxylum indicum (L.) Kurz has been used as plant-based food and herbal medicine in many Asian countries. The aim of the present study was to examine the antioxidant and anti-inflammatory activities of O. indicum extract (O. indicum) in RAW264.7 cells activated by LPS plus IFN-Îł. The phytochemical compounds in O. indicum were identified by GC-MS and LC-MS/MS. Five flavonoids (luteolin, apigenin, baicalein, oroxylin A, and quercetin) and 27 volatile compounds were found in O. indicum. O. indicum presented antioxidant activities, including reducing ability by FRAP assay and free radical scavenging activity by DPPH assay. Moreover, O. indicum also suppressed LPS plus IFN-Îł-activated reactive oxygen species generation in RAW264.7 macrophages. It possessed the potent anti-inflammatory action through suppressing nitric oxide (NO) and IL-6 secretion, possibly due to its ability to scavenge intracellular ROS. The synchrotron radiation-based Fourier transform infrared (SR-FTIR) spectroscopy results showed the alteration of signal intensity and integrated areas relating to lipid and protein of the activated RAW264.7 macrophages compared to unactivated cells. This is the first report of an application of the SR-FTIR technique to evaluate biomolecular changes in activated RAW264.7 cells. Our results indicate that O. indicum may be used as a potential source of nutraceutical for the development of health food supplement or a novel anti-inflammatory herbal medicine

    Oroxylum indicum (L.) Kurz extract inhibits adipogenesis and lipase activity in vitro

    Background: Oroxylum indicum (L.) Kurz (O. indicum) is found in Thailand. It has been used for the treatment of obesity. This study aimed to investigate the effects of an O. indicum extract (OIE) on the adipogenic and biomolecular change in 3T3-L1 adipocytes. Methods: Initial studies examined the chemical components of OIE. The cell line 3T3-L1 was used to establish potential toxic effects of OIE during the differentiation of pre-adipocytes to adipocytes. The inhibitory effect of OIE on lipid accumulation in 3T3-L1 cells was investigated. Moreover, the impact of OIE on pancreatic lipase activity was determined. In further experiments, Fourier Transform Infrared (FTIR) was used to monitor and discriminate biomolecular changes caused by the potential anti-adipogenic effect of OIE on 3T3-L1 cells. Results: Chemical screening methods indicated that OIE was composed of flavonoids, alkaloids, steroids, glycosides, and tannins. The percentage viability of 3T3-L1 cells was not significantly decreased after exposure to either 200 or 150 Îźg/mL of OIE for 2 and 10 days, respectively compared to control cells. The OIE exhibited a dose-dependent reduction of lipid accumulation compared to the control (p < 0.05). The extract also demonstrated a dosedependent inhibitory effect upon lipase activity compared to the control. The inhibitory effect of the OIE on lipid accumulation in 3T3-L1 cells was also confirmed using FTIR microspectroscopy. The signal intensity and the integrated areas relating to lipids, lipid esters, nucleic acids, glycogen and carbohydrates of the OIE-treated 3T3-L1 adipocytes were significantly lower than the non-treated 3T3-L1 adipocytes (p < 0.05). Principal component analysis (PCA) indicated four distinct clusters for the FTIR spectra of 3T3-L1 adipocytes based on biomolecular changes (lipids, proteins, nucleic acids, and carbohydrates). This observation was confirmed using Unsupervised hierarchical cluster analysis (UHCA). Conclusions: These novel findings provide evidence that the OIE derived from the fruit pods of the plant is capable of inhibiting lipid and carbohydrate accumulation in adipocytes and also has the potential to inhibit an enzyme associated with fat absorption. The initial observations indicate that OIE may have important properties which in the future may be exploited for the management of the overweight or obese

    Drought response of rice in Northeastern Thailand assessed via fourier transform infrared spectroscopy

    Drought is a major constraint in many rainfed areas and affects rice yield. We aimed to characterize the physiological changes in rice in response to drought using Fourier transform infrared (FTIR) spectroscopy. Eighty rice landrace seedlings were subjected to drought in the greenhouse using a PEG 6000. Physiological parameters, including total chlorophyll content, relative water content, electrolyte leakage, and biochemical changes were evaluated. Based on the FTIR results, the landraces were divided into three main groups: tolerant, moderately tolerant, and susceptible. Principal component analysis revealed spectral differences between the control and drought stress treatment groups. Lipid, pectin, and lignin content increased after drought stress. The biochemical components of plants at different drought tolerance levels were also compared. The lipid (CH2 and CH3), lignin (C=C), pectin (C=O), and protein (C=O, N–H) contents were the highest in the drought-tolerant cultivars, followed by the moderately tolerant and susceptible cultivars, respectively. Cultivar 17 and 49 were the most tolerant, and the functional groups were identified and characterized using FTIR. Overall, these results will be useful in selecting parental cultivars for rice breeding programs

    Chemical sensing by cell-surface receptors to effect signal transduction is a ubiquitous biological event. Despite extensive structural biochemical study, detailed knowledge of how signal transduction occurs is largely lacking. We report herein a kinetic and structural study, obtained by stopped-flow IR spectroscopy, of the activation of the BlaR1 receptor of the Staphylococcus aureus bacterium by β-lactam antibiotics. The cell-surface BlaR1 receptor alerts the bacterium to the presence of β-lactam antibiotics, resulting in expression of the gene for a β-lactamase enzyme. This enzyme hydrolytically destroys the remaining β-lactam antibiotics. IR spectroscopic interrogation of the β-lactam–BlaR1 receptor reaction has allowed the simultaneous measurement of the chemical events of receptor recognition of the β-lactam and the characterization of the conformational changes in the BlaR1 receptor that result. The key chemical events in β-lactam recognition are serine acylation and subsequent irreversible decarboxylation of the BlaR1 active site lysine carbamate. Both events are observed by stopped-flow IR kinetics and (13)C isotope-edited IR spectroscopy. The secondary structural changes in the BlaR1 receptor conformation that occur as a consequence of this acylation/decarboxylation are predicted to correlate to the signal transduction event accomplished by this receptor

    Understanding the tsunami cycle requires a simple method for identification of tsunami backwash deposits. This study investigates Fourier transform infrared (FTIR) spectroscopy followed by careful analysis of variance (ANOVA), Gaussian distribution, hierarchical cluster analysis (HCA) and principal component analysis (PCA) for the discrimination of typical marine sediments and tsunami backwash deposits. In order to test the suitability of FTIR spectra as innovative methods for classifications of tsunami deposits, typical marine sediments and terrestrial soils were classified into three zones, namely zone-1 (i.e. typical marine sediments), zone-2 (i.e. including tsunami backwash deposits) and zone-3 (i.e. coastal terrestrial soils). HCA was performed to group the spectra according to their spectral similarity in a dendrogram and successfully separate FTIR spectra of all three sampling zones into two main clusters with five sub-clusters. The simplicifolious (i.e. single-leafed) type of dendrogram was observed with the strong dissimilarity of terrestrial components in subcluster- 5. Graphical displays of PC1 vs PC2 highlight the prominent features of zone-1, which is explicitly different from those of zone-2 and zone-3. The acceptable discrimination of typical marine sediments and tsunami backwash deposits, even six years after the tsunami on Boxing Day 2004, dramatically demonstrates the potential of the method for the identification of paleotsunami

    Phenotypic correlations between feed efficiency, growth, and meat quality of slow-growing chickens

    International audienceKorat (KR) chicken is a new meat-type chicken breed established with the purpose of developing Small and Micro Community Enterprise Production. This slow-growing chicken has been recognised for its good texture and flavour, but its low feed efficiency is associated with high cost of production which can hamper the development of local production. This has highlighted the importance of studying the trade-off between feed efficiency and meat quality in KR chicken. This study investigated the phenotypic correlations between feed efficiency, growth performance, and meat quality in KR chicken. Individual body weight and feed intake were recorded weekly for 75 male KR chickens for the calculation of feed conversion ratio (FCR) and residual feed intake (RFI). The growth curve was modelled by Gompertz function and meat quality evaluated at 10 weeks of age, through the measurement of pH (pHu), water-holding capacity (WHC) and drip loss (DL) in breast and thigh muscle.Faster growth rate at young age appeared favourable, regarding feed efficiency, since a moderate negative correlation was found between FCR and initial growth rate. Chickens with a higher initial growth and maturation rates were characterised by lower water-holding capacity in meat. This was not associated with acidic meat, since a higher speed of growth (especially in the first weeks) was positively associated with final pH in breast meat. Relationships between RFI and meat quality traits appeared to be weak. While better FCR could be achieved by increased initial growth rate, this seemed to negatively impact water-holding capacity of the meat.RFI appeared as an interesting alternative as a compromise between FCR and meat quality

    This paper focuses on providing new results relating to the impacts of Diurnal variation, Vertical distribution, and Emission source on sulfur K-edge XANES spectrum of aerosol samples. All aerosol samples used in the diurnal variation experiment were preserved using anoxic preservation stainless cylinders (APSCs) and pressure-controlled glove boxes (PCGBs), which were specially designed to prevent oxidation of the sulfur states in PM10. Further investigation of sulfur K-edge XANES spectra revealed that PM10 samples were dominated by S(VI), even when preserved in anoxic conditions. The &quot;Emission source effect&quot; on the sulfur oxidation state of PM10 was examined by comparing sulfur K-edge XANES spectra collected from various emission sources in southern Thailand, while &quot;Vertical distribution effects&quot; on the sulfur oxidation state of PM10 were made with samples collected from three different altitudes from rooftops of the highest buildings in three major cities in Thailand. The analytical results have demonstrated that neither &quot;Emission source&quot; nor &quot;Vertical distribution&quot; appreciably contribute to the characteristic fingerprint of sulfur K-edge XANES spectrum in PM10.</p

    Elongated oligomers assemble into mammalian PrP Amyloid fibrils

    In prion diseases, the mammalian prion protein PrP is converted from a monomeric, mainly α-helical state into β-rich amyloid fibrils. To examine the structure of the misfolded state, amyloid fibrils were grown from a β form of recombinant mouse PrP (residues 91–231). The β-PrP precursors assembled slowly into amyloid fibrils with an overall helical twist. The fibrils exhibit immunological reactivity similar to that of ex vivo PrPSc. Using electron microscopy and image processing, we obtained three-dimensional density maps of two forms of PrP fibrils with slightly different twists. They reveal two intertwined protofilaments with a subunit repeat of not, vert, similar60 Å. The repeating unit along each protofilament can be accounted for by elongated oligomers of PrP, suggesting a hierarchical assembly mechanism for the fibrils. The structure reveals flexible crossbridges between the two protofilaments, and subunit contacts along the protofilaments that are likely to reflect specific features of the PrP sequence, in addition to the generic, cross-β amyloid fold
