    Perceptions of Tenure and Promotion Guidelines and Criteria Among Athletic Training Doctoral Students

    Purpose: Evidence suggested that athletic training doctoral candidates are being socialized through a variety of formal and informal experiences to prepare them for roles in higher education. It is unclear, however, if they are aware of the expectations associated with tenure and/or promotion which are necessary for transition into higher education. Perceptions of tenure and promotion among doctoral students preparing to enter the athletic training professoriate were investigated. Methods: Twenty-four students (16=females, 8=males, average age=28Β±3 years) completed one-on-one telephone interviews. Data were analyzed using a general inductive approach, with a peer review and multiple analyst triangulation completed for rigor. Results: Four primary themes emerged from the data: 1) there is a limited understanding of the guidelines and criteria for tenure and promotion, 2) knowledge of guidelines and criteria is perpetuated by mentoring, 3) research/scholarship is perceived as paramount for success, and 4) the belief that skills gained, regardless of the domain of tenure, can facilitate success in others. Conclusions: The findings suggest that doctoral students in athletic training are partially aware of the guidelines for tenure, but could benefit from additional mentoring

    Zeroing in on Supersymmetric Radiation Amplitude Zeros

    Radiation amplitude zeros have long been used to test the Standard Model. Here, we consider the supersymmetric radiation amplitude zero in chargino-neutralino associated production, which can be observed at the luminosity upgraded LHC. Such an amplitude zero only occurs if the neutralino has a large wino fraction and hence this observable can be used to determine the neutralino eigenstate content. We find that this observable can be measured by comparing the p_T spectrum of the softest lepton in the trilepton Ο‡1Β±Ο‡20\chi_1^\pm \chi_2^0 decay channel to that of a control process such as Ο‡1+Ο‡1βˆ’\chi_1^+ \chi_1^- or Ο‡20Ο‡20\chi_2^0 \chi_2^0. We test this technique on a previously generated model sample of the 19 dimensional parameter space of the phenomenological MSSM, and find that it is effective in determining the wino content of the neutralino.Comment: 19 pages, 7 figure

    Reservoirs of faecal indicator bacteria in well-head hand pumps in Bangladesh

    The majority of the population of Bangladesh (90%) rely on untreated groundwater for drinking and domestic use. At the point of collection, 40% of these supplies are contaminated with faecal indicator bacteria (FIB). Recent studies have disproved the theory that latrines discharging to shallow aquifers are the major contributor to this contamination. In this study, we tested the hypothesis that hand pumps are a reservoir of FIB. We sampled the handle, spout, piston and seal from 19 wells in Araihazar Upazila, Bangladesh and identified that the spout and seal were reservoirs of FIB. These findings led to our recommendation that well spouts be regularly cleaned, including the removal of precipitated deposits, and that the seals be regularly changed. It is envisaged that one or both of these interventions will reduce the numbers of FIB in drinking water, thereby reducing the burden of diarrhoeal disease in Bangladesh

    Group 13 HOX proteins interact with the MH2 domain of R-Smads and modulate Smad transcriptional activation functions independent of HOX DNA-binding capability

    Interactions with co-factors provide a means by which HOX proteins exert specificity. To identify candidate protein interactors of HOXA13, we created and screened an E11.5–E12.5, distal limb bud yeast two-hybrid prey library. Among the interactors, we isolated the BMP-signaling effector Smad5, which interacted with the paralogous HOXD13 but not with HOXA11 or HOXA9, revealing unique interaction capabilities of the AbdB-like HOX proteins. Using deletion mutants, we determined that the MH2 domain of Smad5 is necessary for HOXA13 interaction. This is the first report demonstrating an interaction between HOX proteins and the MH2 domain of Smad proteins. HOXA13 and HOXD13 also bind to other BMP and TGF-Ξ²/Activin-regulated Smad proteins including Smad1 and Smad2, but not Smad4. Furthermore, HOXD13 could be co-immunoprecipitated with Smad1 from cells. Expression of HOXA13, HOXD13 or a HOXD13 homeodomain mutant (HOXD13(IQN>AAA)) antagonized TGF-Ξ²-stimulated transcriptional activation of the pAdtrack-3TP-Lux reporter vector in Mv1Lu cells as well as the Smad3/Smad4-activated pTRS(6)-E1b promoter in Hep3B cells. Finally, using mammalian one-hybrid assay, we show that transcriptional activation by a GAL4/Smad3-C-terminus fusion protein is specifically inhibited by HOXA13. Our results identify a new co-factor for HOX group 13 proteins and suggest that HOX proteins may modulate Smad-mediated transcriptional activity through protein–protein interactions without the requirement for HOX monomeric DNA-binding capability

    A Natural Plasmid Uniquely Encodes Two Biosynthetic Pathways Creating a Potent Anti-MRSA Antibiotic

    Background Understanding how complex antibiotics are synthesised by their producer bacteria is essential for creation of new families of bioactive compounds. Thiomarinols, produced by marine bacteria belonging to the genus Pseudoalteromonas, are hybrids of two independently active species: the pseudomonic acid mixture, mupirocin, which is used clinically against MRSA, and the pyrrothine core of holomycin. Methodology/Principal Findings High throughput DNA sequencing of the complete genome of the producer bacterium revealed a novel 97 kb plasmid, pTML1, consisting almost entirely of two distinct gene clusters. Targeted gene knockouts confirmed the role of these clusters in biosynthesis of the two separate components, pseudomonic acid and the pyrrothine, and identified a putative amide synthetase that joins them together. Feeding mupirocin to a mutant unable to make the endogenous pseudomonic acid created a novel hybrid with the pyrrothine via β€œmutasynthesis” that allows inhibition of mupirocin-resistant isoleucyl-tRNA synthetase, the mupirocin target. A mutant defective in pyrrothine biosynthesis was also able to incorporate alternative amine substrates. Conclusions/Significance Plasmid pTML1 provides a paradigm for combining independent antibiotic biosynthetic pathways or using mutasynthesis to develop a new family of hybrid derivatives that may extend the effective use of mupirocin against MRSA

    Allele distribution and genetic diversity of VNTR loci in Salmonella enterica serotype Enteritidis isolates from different sources

    <p>Abstract</p> <p>Background</p> <p><it>Salmonella enterica </it>serotype Enteritidis (<it>S</it>. Enteritidis) is a zoonotic pathogen, which can be found in many sources including animals and the environment. However, little is known about the molecular relatedness among <it>S</it>. Enteritidis isolates from different sources. We have applied multiple-locus variable number tandem repeat analysis (MLVA) to study the genetic diversity of <it>S</it>. Enteritidis isolates from human and non-human sources.</p> <p>Results</p> <p>We identified 38 unique MLVA types using nine VNTR loci markers for discrimination between 145 <it>S</it>. Enteritidis isolates from different sources including humans (n = 41), chickens (n = 45), and eggs (n = 40). There were 20 distinct MLVA types identified from human isolates, 17 distinct MLVA types from chicken isolates, and 5 from egg isolates. We compared allele distribution and frequency for each VNTR marker and measured allelic polymorphism within each VNTR locus of <it>S</it>. Enteritidis isolates from the sources using Nei's diversity index (<it>D</it>). Differences in allele distribution and frequency were detected in most loci of study isolates. Different genetic diversity for certain loci was identified in isolates from different sources. The average of genetic diversity (<it>D</it>) was lower in egg isolates (0.16) compared to human (0.41) and chicken (0.30). However, for loci SE3, SE7, and SE9, human isolates showed significantly higher diversity than both chicken and egg isolates. Whereas for loci SE5 and SE10, chicken isolates had significantly higher diversity than both human and egg isolates. Minimum-spanning tree (MST) comprised one major cluster, a minor cluster, and four clonal expansions. MLVA application enabled a cluster analysis by the MST of the <it>S</it>. Enteritidis isolates by sources, which allows a great insight into the genetic relatedness and the possible flow of these organisms between different reservoirs and humans.</p> <p>Conclusion</p> <p>Differences in allele distribution and genetic diversity of VNTR loci in <it>S</it>. Enteritidis isolates from different sources were found. Polymorphism in most of the VNTR loci was more frequent among human <it>S</it>. Enteritidis isolates than isolates from chickens or eggs. Therefore, VNTR profiles of <it>S</it>. Enteritidis isolates from a specific source should be further evaluated as potential markers in epidemiologic investigations to trace <it>S</it>. Enteritidis to their probable source.</p
