Estimation of genetic parameters for feed efficiency traits using random regression models in dairy cattle.
Feed efficiency has become an increasingly important research topic in recent years. As feed costs rise and the environmental impacts of agriculture become more apparent, improving the efficiency with which dairy cows convert feed to milk is increasingly important. However, feed intake is expensive to measure accurately on large populations, making the inclusion of this trait in breeding programs difficult. Understanding how the genetic parameters of feed efficiency and traits related to feed efficiency vary throughout the lactation period is valuable to gain understanding into the genetic nature of feed efficiency. This study used 121,226 dry matter intake (DMI) records, 120,500 energy corrected milk (ECM) records, and 98,975 metabolic body weight (MBW) records, collected on 7,440 first lactation Holstein cows from 6 countries (Canada, Denmark, Germany, Spain, Switzerland, and United States of America), from January 2003 to February 2022. Genetic parameters were estimated using a multiple-trait random regression model with a fourth order Legendre polynomial for all traits. Weekly phenotypes for DMI were re-parameterized using linear regressions of DMI on ECM and MBW, creating a measure of feed efficiency that was genetically corrected for ECM and MBW, referred to as genomic residual feed intake (gRFI). Heritability (SE) estimates varied from 0.15 (0.03) to 0.29 (0.02) for DMI, 0.24 (0.01) to 0.29 (0.03) for ECM, 0.55 (0.03) to 0.83 (0.05) for MBW, and 0.12 (0.03) to 0.22 (0.06) for gRFI. In general, heritability estimates were lower in the first stage of lactation compared with the later stages of lactation. Additive genetic correlations between weeks of lactation varied, with stronger correlations between weeks of lactation that were close together. The results of this study contribute to a better understanding of the change in genetic parameters across the first lactation, providing insight into potential selection strategies to include feed efficiency in breeding programs
Effect of alloy treatment and coiling temperature on microstructure and bending performance of ultra-high strength strip steel
Two different high strength B-containing microalloyed steel strips produced in industrial processing conditions, one treated with Ti and the other treated with Al, processed by controlled rolling, accelerated cooling and coiling in two different temperatures ranges [723 K to 733 K (450 °C to 460 °C)] and [633 K to 653 K (360 °C to 380 °C)] were subjected to bend testing. The Ti treated steel coiled at the higher temperature 733 K (460 °C) showed the best bending performance. The relatively softer (tensile strength of and even {112} in the sub-surface region as well as uniformity of through thickness texture of the rolled sheet improve the bendability. In the presence of crack initiators, like coarse and brittle TiN particles found in the Ti treated steel, a harder microstructure and the presence of Cube and Goss texture in the sub-surface layer, seen for the lower coiling temperature can cause local transgranular cleavage cracking. Finally the post-uniform elongation obtained from tensile testing and bendability follow a good correlation
Safety and efficacy of a dapivirine vaginal ring for HIV prevention in women
BACKGROUND
The incidence of human immunodeficiency virus (HIV) infection remains high
among women in sub-Saharan Africa. We evaluated the safety and efficacy of extended
use of a vaginal ring containing dapivirine for the prevention of HIV infection
in 1959 healthy, sexually active women, 18 to 45 years of age, from seven communities
in South Africa and Uganda.
METHODS
In this randomized, double-blind, placebo-controlled, phase 3 trial, we randomly
assigned participants in a 2:1 ratio to receive vaginal rings containing either 25 mg
of dapivirine or placebo. Participants inserted the rings themselves every 4 weeks
for up to 24 months. The primary efficacy end point was the rate of HIV type 1
(HIV-1) seroconversion.
RESULTS
A total of 77 participants in the dapivirine group underwent HIV-1 seroconversion
during 1888 person-years of follow-up (4.1 seroconversions per 100 person-years),
as compared with 56 in the placebo group who underwent HIV-1 seroconversion
during 917 person-years of follow-up (6.1 seroconversions per 100 person-years).
The incidence of HIV-1 infection was 31% lower in the dapivirine group than in
the placebo group (hazard ratio, 0.69; 95% confidence interval [CI], 0.49 to 0.99; P = 0.04). There was no significant difference in efficacy of the dapivirine ring
among women older than 21 years of age (hazard ratio for infection, 0.63; 95% CI,
0.41 to 0.97) and those 21 years of age or younger (hazard ratio, 0.85; 95% CI, 0.45
to 1.60; P = 0.43 for treatment-by-age interaction). Among participants with HIV-1
infection, nonnucleoside reverse-transcriptase inhibitor resistance mutations were
detected in 14 of 77 participants in the dapivirine group (18.2%) and in 9 of 56
(16.1%) in the placebo group. Serious adverse events occurred more often in the
dapivirine group (in 38 participants [2.9%]) than in the placebo group (in 6 [0.9%]).
However, no clear pattern was identified.
CONCLUSIONS
Among women in sub-Saharan Africa, the dapivirine ring was not associated with
any safety concerns and was associated with a rate of acquisition of HIV-1 infection
that was lower than the rate with placebo.Supported by the International Partnership for Microbicides
