    For as long as I can remember, the church, for me, has been a place characterized by shame and hurt. I remember Christian high school friends telling me that I would go to hell for being Queer. I remember hearing sermons from televangelists about the evils of homosexuality, and church leaders pressuring youth leaders to cast out their Queer members.     Specious poisons?: reputation, gender, and democratic politics

    Suggesting that reputation and gossip have been largely ignored by contemporary political theorists, I argue that both reputation and the gossip that helps to constitute it are important aspects of our communal and political lives. I begin with the work of Jean-Jacques Rousseau as representative of a larger early modern discourse that identified the desire for reputation as one that is central to human beings. Arguing that this desire for reputation simultaneously poses great dangers and great power for political communities, Rousseau's vision urges careful attention to political arrangements as a way of harnessing the positive effects of the desire for reputation. In my second chapter, I move to a focus on the relationship between reputation and gender, interrogating the necessity that women maintain spotless sexual reputations (a central feature to Rousseau's political schema) in light of both Mary Wollstonecraft's critique of Rousseau as well as my examination of the fate of Rousseau's heroines. Turning to the work of Harriet Taylor and John Stuart Mill, I contend that their arguments about the stifling effects of reputational politics for individual liberty point to a nuanced understanding of the differential effects of reputation for individuals in various echelons of society. Chapter 4 takes up George Orwell's novel Burmese Days as well as his essay "Shooting an Elephant" to think about the ways in which reputation and gossip work in contexts of radical differences of power. While both of these works are situated in the colonial context, I argue that Orwell's observations about the functions of gossip and reputation within relationships of incommensurate power are instructive beyond the colonial setting. Finally, in Chapter 5, I turn to contemporary political events to explore the previous chapters' themes of community, gender, individual liberty, and power through the lens of sexual reputation and the gossip that surfaced during the Bill Clinton and Monica Lewinsky scandal

    Propranolol Sensitizes Vascular Sarcoma Cells to Doxorubicin by Altering Lysosomal Drug Sequestration and Drug Efflux

    Angiosarcoma is a rare cancer of blood vessel–forming cells with a high patient mortality and few treatment options. Although chemotherapy often produces initial clinical responses, outcomes remain poor, largely due to the development of drug resistance. We previously identified a subset of doxorubicin-resistant cells in human angiosarcoma and canine hemangiosarcoma cell lines that exhibit high lysosomal accumulation of doxorubicin. Hydrophobic, weak base chemotherapeutics, like doxorubicin, are known to sequester within lysosomes, promoting resistance by limiting drug accessibility to cellular targets. Drug synergy between the beta adrenergic receptor (β-AR) antagonist, propranolol, and multiple chemotherapeutics has been documented in vitro, and clinical data have corroborated the increased therapeutic potential of propranolol with chemotherapy in angiosarcoma patients. Because propranolol is also a weak base and accumulates in lysosomes, we sought to determine whether propranolol enhanced doxorubicin cytotoxicity via antagonism of β-ARs or by preventing the lysosomal accumulation of doxorubicin. β-AR-like immunoreactivities were confirmed in primary tumor tissues and cell lines; receptor function was verified by monitoring downstream signaling pathways of β-ARs in response to receptor agonists and antagonists. Mechanistically, propranolol increased cytoplasmic doxorubicin concentrations in sarcoma cells by decreasing the lysosomal accumulation and cellular efflux of this chemotherapeutic agent. Equivalent concentrations of the receptor-active S-(−) and -inactive R-(+) enantiomers of propranolol produced similar effects, supporting a β-AR-independent mechanism. Long-term exposure of hemangiosarcoma cells to propranolol expanded both lysosomal size and number, yet cells remained sensitive to doxorubicin in the presence of propranolol. In contrast, removal of propranolol increased cellular resistance to doxorubicin, underscoring lysosomal doxorubicin sequestration as a key mechanism of resistance. Our results support the repurposing of the R-(+) enantiomer of propranolol with weak base chemotherapeutics to increase cytotoxicity and reduce the development of drug-resistant cell populations without the cardiovascular and other side effects associated with antagonism of β-ARs

    Microbial co-occurrences on catheters from long-term catheterized patients

    Catheter-associated urinary tract infections (CAUTIs), a common cause of healthcare-associated infections, are caused by a diverse array of pathogens that are increasingly becoming antibiotic resistant. We analyze the microbial occurrences in catheter and urine samples from 55 human long-term catheterized patients collected over one year. Although most of these patients were prescribed antibiotics over several collection periods, their catheter samples remain colonized by one or more bacterial species. Examination of a total of 366 catheter and urine samples identify 13 positive and 13 negative genus co-occurrences over 12 collection periods, representing associations that occur more or less frequently than expected by chance. We find that for many patients, the microbial species composition between collection periods is similar. In a subset of patients, we find that the most frequently sampled bacteria, Escherichia coli and Enterococcus faecalis, co-localize on catheter samples. Further, co-culture of paired isolates recovered from the same patients reveals that E. coli significantly augments E. faecalis growth in an artificial urine medium, where E. faecalis monoculture grows poorly. These findings suggest novel strategies to collapse polymicrobial CAUTI in long-term catheterized patients by targeting mechanisms that promote positive co-associations
