    alpha-(Aminomethyl)acrylate: polymerization and spontaneous post-polymerization modification of beta-amino acid ester for a pH/temperature-responsive material

    First published online 16 Jun 2015Ethyl α-(aminomethyl)acrylate, a β-amino acid ester carrying a conjugated vinylidene group at the α-position, was radically polymerized. The polymerization was found to involve subsequent ester–amide exchange reaction between the amino pendants of the polymer and an ester group of the monomer, affording acrylamide-bearing units in 11–15% contents.     α-exomethylene lactone possessing acetal–ester linkage: Polymerization and postpolymerization modification for water-soluble polymer

    First published: 16 October 20152,6-Dimethyl-5-methylene-1,3-dioxa-4-one (DMDO), a cyclic acrylate possessing acetal–ester linkage, was obtained as a mixture of cis- and trans-isomers (95:5) from Baylis–Hillman reaction of an aryl acrylate. The radical and anionic polymerizations of DMDO yielded the corresponding vinyl polymers without any side reactions such as cleavage of the acetal–ester linkage. The polymerization behaviors were significantly different from that of the acyclic acrylate, α-(hydroxymethyl)acrylic acid, which was expected inactive against polymerization due to the steric hindrance around the vinylidene group by the α-substituent.     Short interfering RNA-directed inhibition of hepatitis B virus replication

    AbstractRNA interference (RNAi) is the process by which double-stranded RNA directs sequence-specific degradation of mRNA. In mammalian cells, RNAi can be triggered by 21-nucleotide duplexes of short interfering RNA (siRNA). We examined effects of siRNA on hepatitis B virus (HBV) replication. Human hepatoma cells were transfected with HBV DNA and siRNA against HBV-pregenome RNA. Transfection experiments demonstrated that the siRNA reduced the amount of HBV-pregenome RNA and resulted in reduction of the levels of replicative intermediates and viral protein. Our results indicate that siRNA-mediated gene silencing inhibits HBV replication through suppression of viral RNA, which may be useful as a potential therapeutic modality

    Immunohistochemical analysis of CYP2A13 in various types of human lung cancers

    Human CYP2A13, which is expressed in the respiratory tract, is the most efficient enzyme for the metabolic activation of tobacco-specific nitrosamines such as 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK). The relevance of CYP2A13 in carcinogenicity and toxicity in the respiratory tract has been suggested, but the expression of CYP2A13 protein in lung cancer tissues remains to be determined. We first prepared a mouse monoclonal antibody against human CYP2A13. The antibody showed no cross reactivity with the other CYP isoforms including CYP2A6. Using the specific antibody, we performed immunohistochemical analysis for human lung carcinomas. In adenocarcinomas (n=15), all specimens were positive for the staining and five samples showed strong staining. In squamous cell carcinomas (n=15) and large cell carcinomas (n=15), each 14 samples were positive for the staining and two and three samples showed strong staining, respectively. In small cell carcinoma samples (n=15), eight samples were negative for the staining and five samples showed weak or moderate staining. In conclusion, we first found that the expression of CYP2A13 was markedly increased in non-small cell lung carcinomas. The high expression might be associated with the tumor development and progression in non-small cell lung carcinomas. © 2010 Japanese Cancer Association

    We have constructed a new research environment for geo-space science based on 3-D visualization tool and network database; Virtual Earth\u27s Magnetosphere System (VEMS). With an interactive research environment researchers can visually understand structures of the Earth\u27s magnetosphere using VEMS. On the VEMS, computer simulation results and observation data are simultaneously visualized, having a potential to data assimilation for geo-space studies in the future. Since the VEMS deals with time-dependent data, it also helps researchers to study dynamics of the Earth\u27s magnetosphere. We found that immersive data analyses are possible using the VEMS on a virtual reality system

    Identification of an Anti–Integrin αvβ6 Autoantibody in Patients With Ulcerative Colitis

Although involvement of autoimmunity has been suggested in ulcerative colitis, specific autoantigens/antibodies have yet to be elucidated. Methods: Using 23 recombinant integrin proteins, we performed enzyme-linked immunosorbent assays on sera from patients with ulcerative colitis and controls. Integrin expression and IgG binding in the colon tissues of patients with ulcerative colitis and controls were examined using immunofluorescence and coimmunoprecipitation, respectively. The blocking activity of autoantibodies was examined using solid-phase binding and cell adhesion assays. Results: Screening revealed that patients with ulcerative colitis had IgG antibodies against integrin αvβ6. In the training and validation groups, 103 of 112 (92.0%) patients with ulcerative colitis and only 8 of 155 (5.2%) controls had anti–integrin αvβ6 antibodies (P < .001), resulting in a sensitivity of 92.0% and a specificity of 94.8% for diagnosing ulcerative colitis. Anti–integrin αvβ6 antibody titers coincided with ulcerative colitis disease activity, and IgG1 was the major subclass. Patient IgG bound to the integrin αvβ6 expressed on colonic epithelial cells. Moreover, IgG of patients with ulcerative colitis blocked integrin αvβ6–fibronectin binding through an RGD (Arg-Gly-Asp) tripeptide motif and inhibited cell adhesion. Conclusions: A significant majority of patients with ulcerative colitis had autoantibodies against integrin αvβ6, which may serve as a potential diagnostic biomarker with high sensitivity and specificity

    OBJECT: The interferon-induced Jak-STAT signal alone is not sufficient to explain all the biological effects of IFN. The PI3-K pathways have emerged as a critical additional component of IFN-induced signaling. This study attempted to clarify that relationship between IFN-induced PI3-K-Akt-mTOR activity and anti-viral action. RESULT: When the human normal hepatocyte derived cell line was treated with rapamycin (rapa) before accretion of IFN-alpha, tyrosine phosphorylation of STAT-1 was diminished. Pretreatment of rapa had an inhibitory effect on the IFN-alpha-induced expression of PKR and p48 in a dose dependent manner. Rapa inhibited the IFN-alpha inducible IFN-stimulated regulatory element luciferase activity in a dose-dependent manner. However, wortmannin, LY294002 and Akt inhibitor did not influence IFN-alpha inducible luciferase activity. To examine the effect of PI3-K-Akt-mTOR on the anti-HCV action of IFN-alpha, the full-length HCV replication system, OR6 cells were used. The pretreatment of rapa attenuated its anti-HCV replication effect in comparison to IFN-alpha alone, whereas the pretreatment with PI3-K inhibitors, wortmannin and LY294002 and Akt inhibitor did not influence IFN-induced anti-HCV replication. CONCLUSION: IFN-induced mTOR activity, independent of PI3K and Akt, is the critical factor for its anti-HCV activity.     Vortex-induced vibration and its effect on torsional flutter instability in the case of B/D=4 rectangular cylinder

Also, the role of vortex-generation on the torsional flutter instability was experimentally investigated by changing the rotation axis in the state without/with a splitter plate in the wake. The two different kinds of vortex-induced excitations was confirmed in the close reduced wind speed region, and the vortex-induced excitations could play an important role as triggering of torsional flutter instability when Scruton number is small enough. Furthermore, the flow pattern around the cylinder during torsional motion drastically changes between the low and high-reduced velocity
