Time evolution in visible light emission from high-power Ar pulse-modulated induction thermal plasmas
Transient behavior of a 15-kW Ar pulse-modulated induction thermal plasma (PMITP) at a pressure of 31 kPa was observed using a high-speed video camera. The PMITP method is one of new techniques for controlling temperature and radical density in time-domain in high-power thermal plasmas. The adoption of a high-power metal-oxide-semiconductor field-effect transistor (MOSFET) inverter power supply makes it possible to sustain PMITPs with high power-conversion efficiency. The high-speed video pictures provide two-dimensional, time-dependent radiation intensity distributions from PMITPs, which are useful for understanding an unique transition behavior of PMITPs
Population-level transition of capsular polysaccharide types among sequence type 1 group B Streptococcus isolates with reduced penicillin susceptibility during a long-term hospital epidemic
Over a 35-month period, group B Streptococcus isolates with reduced penicillin susceptibility (PRGBS) were detected from elderly patients at a regional hospital in Japan, accompanying population-level transition of PRGBS serotypes. The genetic relatedness of 77 non-duplicate PRGBS from 73 patients was analysed. Serotype III PRGBS predominated (16 serotype III/1 serotype Ib) in the first 9 months (period I), then 3 serotype Ib isolates appeared transiently for the next 3 months (period II), which was replaced predominantly by serotype Ia (20 serotype Ia/1 serotype III/1 non-typeable) for 9 months (period III). In the last 14 months (period IV), besides 25 serotype Ia isolates, 10 serotype III were also identified. Serotypes III and Ia isolates, belonging to ST1, shared G329V, G398A, V405A and G429D substitutions in penicillin-binding protein 2X. Of three strains subjected to whole-genome sequencing, serotype III strain SU12 (period I) had a higher degree of genomic similarity with serotype Ia strain SU97 (period III) than serotype Ib strain SU67 (period II) based on average nucleotide identity and single nucleotide polymorphisms. Analysis of the cps gene clusters and the upstream and downstream flanking sequences revealed that disruption of the hyaluronidase gene located upstream of cpsY by insertion of IS 1548 was found in strain SU12, whereas Delta ISSag8 was inserted between tRNA-Arg and rpsA genes located downstream of cpsL in strain SU97. Interestingly, most serotype III PRGBS re-emerging in period IV had this tRNA-Arg-Delta ISSag8-rpsA region. Capsular switching and nosocomial transmission may possibly contribute to population-level serotype replacement among ST1 PRGBS isolates. The Subaru Deep Field Project: Lyman Emitters at Redshift of 6.6
We present new results of a deep optical imaging survey using a narrowband
filter () centered at 9196 \AA ~ together with , ,
, , and broadband filters in the sky area of the Subaru
Deep Field which has been promoted as one of legacy programs of the 8.2m Subaru
Telescope. We obtained a photometric sample of 58 Ly emitter candidates
at 6.5 -- 6.6 among strong -excess () objects together with a color criterion of . We then obtained optical spectra of 20 objects in our -excess
sample and identified at least nine Ly emitters at -- 6.6
including the two emitters reported by Kodaira et al. (2003). Since our
Ly emitter candidates are free from strong amplification of
gravitational lensing, we are able to discuss their observational properties
from a statistical point of view. Based on these new results, we obtain a lower
limit of the star formation rate density of yr Mpc at , being
consistent with our previous estimate. We discuss the nature of star-formation

Development and characterization of transgenic dominant male sterile rice toward an outcross-based breeding system
Genomic selection is attracting attention in the field of crop breeding. To apply genomic selection effectively for autogamous (self-pollinating) crops, an efficient outcross system is desired. Since dominant male sterility is a powerful tool for easy and successive outcross of autogamous crops, we developed transgenic dominant male sterile rice (Oryza sativa L.) using the barnase gene that is expressed by the tapetum-specific promoter BoA9. Barnase-induced male sterile rice No. 10 (BMS10) was selected for its stable male sterility and normal growth characteristics. The BMS10 flowering habits, including heading date, flowering date, and daily flowering time of BMS10 tended to be delayed compared to wild type. When BMS10 and wild type were placed side-by-side and crossed under an open-pollinating condition, the seed-setting rate was <1.5%. When the clipping method was used to avoid the influence of late flowering habits, the seed-setting rate of BMS10 increased to a maximum of 86.4%. Although flowering synchronicity should be improved to increase the seed-setting rate, our results showed that this system can produce stable transgenic male sterility with normal female fertility in rice. Cellular HIV-1 DNA levels in patients receiving antiretroviral therapy strongly correlate with therapy initiation timing but not with therapy duration
<p>Abstract</p> <p>Background</p> <p>Viral reservoir size refers to cellular human immunodeficiency virus-1 (HIV-1) DNA levels in CD4<sup>+ </sup>T lymphocytes of peripheral blood obtained from patients with plasma HIV-1-RNA levels (viral load, VL) maintained below the detection limit by antiretroviral therapy (ART). We measured HIV-1 DNA levels in CD4<sup>+ </sup>lymphocytes in such patients to investigate their clinical significance.</p> <p>Methods</p> <p>CD4<sup>+ </sup>T lymphocytes were isolated from the peripheral blood of 61 patients with a VL maintained at less than 50 copies/ml for at least 4 months by ART and total DNA was purified. HIV-1 DNA was quantified by nested PCR to calculate the copy number per 1 million CD4<sup>+ </sup>lymphocytes (relative amount) and the copy number in 1 ml of blood (absolute amount). For statistical analysis, the Spearman rank or Wilcoxon signed-rank test was used, with a significance level of 5%.</p> <p>Results</p> <p>CD4 cell counts at the time of sampling negatively correlated with the relative amount of HIV-1 DNA (median = 33 copies/million CD4<sup>+ </sup>lymphocytes; interquartile range [IQR] = 7-123 copies/million CD4<sup>+ </sup>lymphocytes), but were not correlated with the absolute amounts (median = 17 copies/ml; IQR = 5-67 copies/ml). Both absolute and relative amounts of HIV-1 DNA were significantly lower in six patients in whom ART was initiated before positive seroconversion than in 55 patients in whom ART was initiated in the chronic phase, as shown by Western blotting. CD4 cell counts before ART introduction were also negatively correlated with both the relative and absolute amounts of HIV-1 DNA. Only the relative amounts of HIV-1 DNA negatively correlated with the duration of VL maintenance below the detection limit, while the absolute amounts were not significantly correlated with this period.</p> <p>Conclusions</p> <p>The amounts of cellular HIV-1 DNA in patients with VLs maintained below the detection limit by the introduction of ART correlated with the timing of ART initiation but not with the duration of ART. In addition, CD4<sup>+ </sup>T lymphocytes, which were newly generated by ART, diluted latently infected cells, indicating that measurements of the relative amounts of cellular HIV-1 DNA might be underestimated.</p
Mizoribine Suppresses the Progression of Experimental Peritoneal Fibrosis in a Rat Model
Background/Aims: Peritoneal fibrosis is a serious complication of peritoneal dialysis (PD). It has been reported that administration of mizoribine, an effective immunosuppressant, ameliorated renal fibrosis in a rat model of unilateral ureteral obstruction. We therefore examined the effects of mizoribine in an experimental model of peritoneal fibrosis. Methods: 24 rats were given a daily intraperitoneal injection of chlorhexidine gluconate and ethanol dissolved in saline. The rats were divided into three groups (n = 8 per group) that received either vehicle or mizoribine at a dose of 2 or 8 mg/kg once a day. 28 days after the start of the treatments the rats were sacrificed and peritoneal tissue samples collected. Macrophage infiltration (ED1), myofibroblast accumulation (alpha-smooth muscle actin (SMA)) and expression of type III collagen, transforming growth factor (TGF)-beta and monocyte chemotactic protein-1 (MCP-1) were examined by immunohistochemistry. Results: Mizoribine significantly suppressed submesothelial zone thickening and reduced macrophage infiltration. Mizoribine also reduced collagen III+ area and decreased the number of alpha-SMA(+), TGF-beta(+) and MCP-1(+) cells. The magnitude of the changes observed was dose-dependent. Conclusion: The administration of mizoribine prevented the progression of peritoneal fibrosis in this rat model. Transient IGF-1R inhibition combined with osimertinib eradicates AXL-low expressing EGFR mutated lung cancer
Drug tolerance is the basis for acquired resistance to epidermal growth factor receptor-tyrosine kinase inhibitors (EGFR-TKIs) including osimertinib, through mechanisms that still remain unclear. Here, we show that while AXL-low expressing EGFR mutated lung cancer (EGFRmut-LC) cells are more sensitive to osimertinib than AXL-high expressing EGFRmut-LC cells, a small population emerge osimertinib tolerance. The tolerance is mediated by the increased expression and phosphorylation of insulin-like growth factor-1 receptor (IGF-1R), caused by the induction of its transcription factor FOXA1. IGF-1R maintains association with EGFR and adaptor proteins, including Gab1 and IRS1, in the presence of osimertinib and restores the survival signal. In AXL-low-expressing EGFRmut-LC cell-derived xenograft and patient-derived xenograft models, transient IGF-1R inhibition combined with continuous osimertinib treatment could eradicate tumors and prevent regrowth even after the cessation of osimertinib. These results indicate that optimal inhibition of tolerant signals combined with osimertinib may dramatically improve the outcome of EGFRmut-LC
A Shock-Induced Pair of Superbubbles in the High-Redshift Powerful Radio Galaxy MRC 0406-244
We present new optical spectroscopy of the high-redshift powerful radio
galaxy MRC 0406244 at redshift of 2.429. We find that the two extensions
toward NW and SE probed in the rest-frame ultraviolet image are heated mainly
by the nonthermal continuum of the active galactic nucleus. However, each
extension shows a shell-like morphology, suggesting that they are a pair of
superbubbles induced by the superwind activity rather than by the interaction
between the radio jet and the ambient gas clouds. If this is the case, the
intense starburst responsible for the formation of superbubbles could occur
yr ago. On the other hand, the age of the radio jets may
be of the order of yr, being much shorter than the starburst age.
Therefore, the two events, i.e., the starburst and the radio-jet activities,
are independent phenomena. However, their directions of the expanding motions
could be governed by the rotational motion of the gaseous component in the host

Decomposition of the Superwind in M82
We present new optical images (, , and H) of the archetypical
starburst/superwind galaxy M82 obtained with the 8.2 m Subaru Telescope to
reveal new detailed structures of the superwind-driven nebula and the
high-latitude dark lanes. The emission-line nebula is decomposed into (1) a
ridge-dominated component comprising numerous filament/loop sub-structures
whose overall morphology appears as a pair of narrow cylinders, and (2) a
diffuse component extended over much wider opening angle from the nucleus. We
suggest that these two components have different origins. The ridge-dominated
component appears as a pair of cylinders rather than a pair of cones. Since
this morphological property is similar to that of hot plasma probed by soft
X-ray, this component seems to surround the hot plasma. On the other hand, the
diffuse component may arise from dust grains which scatter stellar light from
the galaxy. Since inner region of this component is seen over the prominent ^^
^^ X"-shaped dark lanes streaming out from the nuclear region and they can be
reproduced as a conical distribution of dust grains, there seems to be a dusty
cold outflow as well as the hot one probed by soft X-ray and shock-excited
optical emission lines. If this is the case, the presence of such high-latitude
dust grains implies that neutral gaseous matter is also blown out during the
