    High-dimensional structure estimation in Ising models: Local separation criterion

    We consider the problem of high-dimensional Ising (graphical) model selection. We propose a simple algorithm for structure estimation based on the thresholding of the empirical conditional variation distances. We introduce a novel criterion for tractable graph families, where this method is efficient, based on the presence of sparse local separators between node pairs in the underlying graph. For such graphs, the proposed algorithm has a sample complexity of n=Ω(Jmin2logp)n=\Omega(J_{\min}^{-2}\log p), where pp is the number of variables, and JminJ_{\min} is the minimum (absolute) edge potential in the model.     Application of high-resolution remote sensing technology for the iron ore deposits of the West Kunlun Mountains in China

    This study focuses on the iron ore of Taxkorgan and Heiqia in the West Kunlun mountains as a case study, for the application of WorldView−2 and IKONOS remote sensing images as major data sources in the fabrication of a standard image map and in the adoption of image enhancement methods to extract information on the ore-controlling factors and mineralization, to interpret remote sensing for the mineral resources in these areas. ASTER, WorldView−2, and IKONOS data were applied for the extraction of alteration anomaly information. With an appropriate amount of field sampling and verification tests, this was used to establish a remote sensing geology prospecting model, that would provide the basis for future remote sensing of metallogenic belts in  West Kunlun in the hope of discovering similar minerals. Survey results showed four additional iron ore mineralization belts could be delineated in the Taxkorgan area. A comparative analysis conducted for part of the field confirmation and the known mineral deposits indicated good reliability. In Heiqia, a siderite-haematite mineralization zone was observed with copperlead- zinc formation, 60-km in length and 200–500 m wide, which includes several mineralized bodies. The ore bodies, appear as stratoid, lenticular, or podiform morphologies and were located in the transition site from clastic to carbonate rocks of the D segment in the Wenquangou Group. The ore bodies generally occur within 40°–50° strike and 68°–81° dip, in accordance to the strata. The length of the single body varies from several hundred metres to more than 9500 m. Its exposed thickness on the surface ranges from 2–50 m, and the general thickness was approximately 15 m. The surface ore minerals were mainly haematite and limonite, with a small amount of siderite. Therefore, high-resolution remote sensing technology is suitable for iron ore geological and mineral remote sensing surveying. It is advantageous in both high-ground resolution of optical characteristics and a certain spectral recognition capability, and is effective not only for information extraction from a large area, but also for recognition of local mineralization outcrops. Therefore, high-resolution remote sensing technology is valuable for popularization.

    Application of an airborne hyper-spectral survey system CASI/SASI in the gold-silver-lead-zinc ore district of Huaniushan, Gansu, China

    The airborne hyper-spectral survey system CASI/SASI, which has an integrated system for gathering both image an spectral data, is at the cutting edge developments in the remote-sensing field. It can be used to directly identify surface objects based on diagnostic spectral characteristics. In this paper, the CASI/SASI were used in the Huaniushan gold-silver-lead-zinc ore district–Gansu to produce a lithologic map, identify altered minerals, and map the mineralized-alteration zones. Radiometric correction, radiometric calibration, atmospheric correction (spectral reconstruction), and geometric corrections were carried out in ENVI to pre-process the measured data. A FieldSpec ® Pro FR portable spectrometer was used to obtain the spectral signatures of all types of rock samples, ore deposits, and mineralized-alteration zones. We extracted and analyzed the spectral characteristics of typical alteration minerals. On the basis of hyper-spectral data, ground-spectral data processing, and comparative analysis of the measured image spectrum, we used the spectral-angle-mapping (SAM) and mixture-tuned matchedfiltering (MTMF) methods to perform hyperspectral-alteration mineral  mapping of wall rock and mineralized-alteration-zone hyperspectral identification. Hyperspectral- remote- sensing geological- classification maps were produced as well as distribution maps of all kinds of alteration minerals and mineralized-alteration zones. Based on geological comprehensive analysis and field investigations, the range of mineral alteration was proven to be the same as shown by the remote-sensing imagery. Indications are that airborne hyperspectral- remote-sensing -image CASI/SASI offer good application results and show a promising potential as a tool in geological investigations. The results will provide the basis for hyperspectral remote-sensing prospecting in the same or similar unexplored areas.

    Specific, simple and rapid detection of porcine circovirus type 2 using the loop-mediated isothermal amplification method

    <p>Abstract</p> <p>Background</p> <p>Porcine circovirus type 2 (PCV2) is the causative agent of postweaning multisystemic wasting syndrome (PMWS), and porcine dermatitis and nephropathy syndrome (PDNS). It has caused heavy losses in global agriculture in recent decades. Rapid detection of PCV2 is very important for the effective prophylaxis and treatment of PMWS.</p> <p>Results</p> <p>A loop-mediated isothermal amplification (LAMP) assay was used to detect PCV2 in this study. Three pairs of primers were specially designed for recognizing eight distinct sequences of the ORF2 gene. This gene lies in the PCV2 virus genome sequence, and encodes the Rep protein that is involved in virus replication. Time and temperature conditions for amplification of PCV2 genes were optimized to be 55 min at 59°C. The analysis of clinical samples indicated that the LAMP method was highly sensitive. The detection limit for PCV2 by the LAMP assay was 10 copies, whereas the limit by conventional PCR was 1000 copies. The assay did not cross-react with PCV1, porcine reproductive and respiratory syndrome virus, porcine epidemic diarrhea virus, transmissible gastroenteritis of pigs virus or rotavirus. When 110 samples were tested using the established LAMP system, 95 were detected as positive.</p> <p>Conclusion</p> <p>The newly developed LAMP detection method for PCV2 was more specific, sensitive, rapid and simple than before. It complements and extends previous methods for PCV2 detection and provides an alternative approach for detection of PCV2.</p

    Learning Segment Similarity and Alignment in Large-Scale Content Based Video Retrieval

    With the explosive growth of web videos in recent years, large-scale Content-Based Video Retrieval (CBVR) becomes increasingly essential in video filtering, recommendation, and copyright protection. Segment-level CBVR (S-CBVR) locates the start and end time of similar segments in finer granularity, which is beneficial for user browsing efficiency and infringement detection especially in long video scenarios. The challenge of S-CBVR task is how to achieve high temporal alignment accuracy with efficient computation and low storage consumption. In this paper, we propose a Segment Similarity and Alignment Network (SSAN) in dealing with the challenge which is firstly trained end-to-end in S-CBVR. SSAN is based on two newly proposed modules in video retrieval: (1) An efficient Self-supervised Keyframe Extraction (SKE) module to reduce redundant frame features, (2) A robust Similarity Pattern Detection (SPD) module for temporal alignment. In comparison with uniform frame extraction, SKE not only saves feature storage and search time, but also introduces comparable accuracy and limited extra computation time. In terms of temporal alignment, SPD localizes similar segments with higher accuracy and efficiency than existing deep learning methods. Furthermore, we jointly train SSAN with SKE and SPD and achieve an end-to-end improvement. Meanwhile, the two key modules SKE and SPD can also be effectively inserted into other video retrieval pipelines and gain considerable performance improvements. Experimental results on public datasets show that SSAN can obtain higher alignment accuracy while saving storage and online query computational cost compared to existing methods.Comment: Accepted by ACM MM 202

    Rapid detection of porcine circovirus type 2 using a TaqMan-based real-time PCR

    Porcine circovirus type 2 (PCV2) and the associated disease postweaning multisystemic wasting syndrome (PMWS) have caused heavy losses in global agriculture in recent decades. Rapid detection of PCV2 is very important for the effective prophylaxis and treatment of PMWS. To establish a sensitive, specific assay for the detection and quantitation of PCV2, we designed and synthesized specific primers and a probe in the open reading frame 2. The assay had a wide dynamic range with excellent linearity and reliable reproducibility, and detected between 102 and 1010 copies of the genomic DNA per reaction. The coefficient of variation for Ct values varied from 0.59% to 1.05% in the same assay and from 1.9% to 4.2% in 10 different assays. The assay did not cross-react with porcine circovirus type 1, porcine reproductive and respiratory, porcine epidemic diarrhea, transmissible gastroenteritis of pigs and rotavirus. The limits of detection and quantitation were 10 and 100 copies, respectively. Using the established real-time PCR system, 39 of the 40 samples we tested were detected as positive

    Involvement of histone deacetylation in MORC2-mediated down-regulation of carbonic anhydrase IX

    Carbonic anhydrase IX (CAIX) plays an important role in the growth and survival of tumor cells. MORC2 is a member of the MORC protein family. The MORC proteins contain a CW-type zinc finger domain and are predicted to have the function of regulating transcription, but no MORC2 target genes have been identified. Here we performed a DNA microarray hybridization and found CAIX mRNA to be down-regulated 8-fold when MORC2 was overexpressed. This result was further confirmed by northern and western blot analysis. Our results also showed that the protected region 4 (PR4) was important for the repression function of MORC2. Moreover, MORC2 decreased the acetylation level of histone H3 at the CAIX promoter. Meanwhile, trichostatin A (TSA) had an increasing effect on CAIX promoter activity. Among the six HDACs tested, histone deacetylase 4 (HDAC4) had a much more prominent effect on CAIX repression. ChIP and ChIP Re-IP assays showed that MORC2 and HDAC4 were assembled on the same region of the CAIX promoter. Importantly, we further confirmed that both proteins are simultaneously present in the PR4-binding complex. These results may contribute to understanding the molecular mechanisms of CAIX regulation

    Integrated metabolomics and lipidomics evaluate the alterations of flavor precursors in chicken breast muscle with white striping symptom

    White striping (WS) is the most common myopathy in the broiler chicken industry. To reveal flavor changes of WS meat objectively, flavor precursors of WS breast muscle were evaluated systematically with integrated metabolomics and lipidomics. The results showed that WS could be distinguished from normal controls by E-nose, and four volatile compounds (o-xylene, benzene, 1,3-dimethyl, 2-heptanone and 6-methyl and Acetic acid and ethyl ester) were detected as decreased compounds by gas chromatography-mass spectrometry. Lipidomic analysis showed that WS breast fillets featured increased neutral lipid (83.8%) and decreased phospholipid molecules (33.2%). Targeted metabolomic analysis indicated that 16 hydrophilic metabolites were altered. Thereinto, some water-soluble flavor precursors, such as adenosine monophosphate, GDP-fucose and L-arginine increased significantly, but fructose 1,6-bisphosphate and L-histidine significantly decreased in the WS group. These results provided a systematic evaluation of the flavor precursors profile in the WS meat of broiler chickens
