    Simultaneous Smelling an Incense Outdoor and Putting the Hands Together Activate Specific Brain Areas

    Mirror neurons are involved in imitation of habitual behaviors. To increase understanding of the theory of mirror neurons and the default mode network, brain activation was explored in 11 healthy adult volunteers who did or did not have a habit of putting their hands together as if praying. Magnetoencephalography (MEG) data were recorded while the participants simultaneously smelled an odor in two kinds of incenses outdoor and/or while they moved to putting their hands together. A magnetoencephalographic contour map of the recorded findings was drawn and an estimated current dipole (ECD) was set. Regardless of a habit of putting their hands together or not, the inner lobe of the frontal area, anterior area in the temporal lobe, and F5 language area in the left frontal lobe and so on were specifically activated. We used cortisol value as an index of the stress state measured in every state (before and after smelling two different incenses outdoor). These experiments suggest that simultaneous smelling an incense outdoor and the behavior of putting their hands together increased the activity of these specific areas in the human brain due to mutual interactions and enhanced interactions

    Giantin Affects Golgi Stack Connection

    Golgins are a family of Golgi-localized long coiled-coil proteins. The major golgin function is thought to be the tethering of vesicles, membranes, and cytoskeletal elements to the Golgi. We previously showed that knockdown of one of the longest golgins, Giantin, altered the glycosylation patterns of cell surfaces and the kinetics of cargo transport, suggesting that Giantin maintains correct glycosylation through slowing down transport within the Golgi. Giantin knockdown also altered the sizes and numbers of mini Golgi stacks generated by microtubule de-polymerization, suggesting that it maintains the independence of individual Golgi stacks. Therefore, it is presumed that Golgi stacks lose their independence following Giantin knockdown, allowing easier and possibly increased transport among stacks and abnormal glycosylation. To gain structural insights into the independence of Golgi stacks, we herein performed electron tomography and 3D modeling of Golgi stacks in Giantin knockdown cells. Compared with control cells, Giantin-knockdown cells had fewer and smaller fenestrae within each cisterna. This was supported by data showing that the diffusion rate of Golgi membrane proteins is faster in Giantin-knockdown Golgi, indicating that Giantin knockdown structurally and functionally increases connectivity among Golgi cisternae and stacks. This increased connectivity suggests that contrary to the cis-golgin tether model, Giantin instead inhibits the tether and fusion of nearby Golgi cisternae and stacks, resulting in transport difficulties between stacks that may enable the correct glycosylation of proteins and lipids passing through the Golgi

    Cleavage of Toll-Like Receptor 9 Ectodomain Is Required for In Vivo Responses to Single Strand DNA

    Mouse toll-like receptor 9 (TLR9) is an endosomal sensor for single-stranded DNA. TLR9 is transported from the endoplasmic reticulum to endolysosomes by a multiple transmembrane protein Unc93 homolog B1, and proteolytically cleaved at its ectodomain. The structure of TLR9 and its biochemical analyses have shown that the proteolytic cleavage of TLR9 ectodomain enables TLR9-dimerization and TLR9 activation. However, the requirement of TLR9 cleavage in vivo has not been studied. We here show that the 13 amino acids deletion at the cleavage site made TLR9 resistant to proteolytic cleavage. The deletion mutation in the Tlr9 gene impaired TLR9-dependent cytokine production in conventional dendritic cells from the mutant mice. Not only in vitro, in vivo production of inflammatory cytokines (TNF-α and IL-12p40), chemokine (CCR5/RANTES), and type I interferon (IFN-α) induced by administration of TLR9 ligand was also impaired. These results demonstrate that the TLR9 cleavage is required for TLR9 responses in vivo

    Research Activities in the Department of Medical Engineering

    The Department of Medical Engineering is dedicated to the research and educational activities to fulfill its mission as educating medical professionals in medical engineering under the diploma policy and curriculum policy, that is, "research and education aiming for fostering professionals competent in comprehensive resolving capacity based upon a wide field of knowledge and vision in clinical engineering, which can be attained by wearing the basic knowledge of medical science and engineering." For this reason, the Faculty of the Department of Medical Engineering is composed of the two areas; PhDs in engineering-based clinical medicine, and mainly MDs in medical sciences and clinical medicine. To summarize the research activities at the Department of Medical Engineering, the authors will describe the overview of research activities being performed in the Department of Medical Engineering Fields, by dividing into 1) Research in Biomedical Engineering Fields, and 2) Research in Medical Science and Clinical Engineering Fields

    Risk Factors for Bleeding After Endoscopic Submucosal Dissection for Gastric Cancer in Elderly Patients Older Than 80 Years in Japan.

    Introduction:As the aging of people in a society advances, the number of elderly patients older than 80 years in Japan with gastric cancer continues to increase. Although delayed ulcer bleeding is a major adverse event after endoscopic submucosal dissection (ESD), little is known about characteristic risk factors for bleeding in elderly patients undergoing ESD. This study aimed to evaluate risk factors for delayed bleeding after ESD for gastric cancer in elderly patients older than 80 years.Methods:We retrospectively evaluated the incidence of delayed bleeding after ESD in 10,320 patients with early-stage gastric cancer resected by ESD between November 2013 and January 2016 at 33 Japanese institutions and investigated risk factors for delayed bleeding in elderly patients older than 80 years.Results:The incidence of delayed bleeding in elderly patients older than 80 years was 5.7% (95% confidence interval [CI]: 4.6%-6.9%, 95/1,675), which was significantly higher than that in nonelderly (older than 20 years and younger than 80 years) patients (4.5%, 4.1%-5.0%, 393/8,645). Predictive factors for ESD-associated bleeding differed between nonelderly and elderly patients. On multivariate analysis of predictive factors at the time of treatment, risk factors in elderly patients were hemodialysis (odds ratio: 4.591, 95% CI: 2.056-10.248, P < 0.001) and warfarin use (odds ratio: 4.783, 95% CI: 1.689-13.540, P = 0.003).Discussion:This multicenter study found that the incidence of delayed bleeding after ESD in Japanese patients older than 80 years was high, especially in patients receiving hemodialysis and taking warfarin. Management of ESD to prevent delayed bleeding requires particular care in patients older than 80 years
