    Roles of values in the risk factors of passive suicide ideation among young adults in the US and Japan

    The present study examined how the importance of values and perceived value congruence with families, friends, and country would be associated with the risk factors of passive suicide ideation. Specifically, the study investigated the associations that the values and perceived congruence had with thwarted belongingness and perceived burdensomeness during the COVID-19 pandemic after controlling for the impact of depression levels. The data from the US and Japan demonstrated that the values such as cherishing family and friends and value congruence played a protective factor for Japanese participants; however, the associations differed among those in the US. Values such as enduring challenges played a protective factor for perceived burdensomeness in Japan whereas values such as cherishing family and friends played a protective factor and improving society was a risk factor for thwarted belongingness for those in the US. These results can be used to further understand the roles of values in mental health

    Stepwise Assembly of Fibrinogen Is Assisted by the Endoplasmic Reticulum Lectin-Chaperone System in HepG2 Cells

    The endoplasmic reticulum (ER) plays essential roles in protein folding and assembly of secretory proteins. ER-resident molecular chaperones and related enzymes assist in protein maturation by co-operated interactions and modifications. However, the folding/assembly of multimeric proteins is not well understood. Here, we show that the maturation of fibrinogen, a hexameric secretory protein (two trimers from a, b and c subunits), occurs in a stepwise manner. The ac complex, a precursor for the trimer, is retained in the ER by lectin-like chaperones, and the b subunit is incorporated into the ac complex immediately after translation. ERp57, a protein disulfide isomerase homologue, is involved in the hexamer formation from two trimers. Our results indicate that the fibrinogen hexamer is formed sequentially, rather than simultaneously, using kinetic pause by lectin chaperones. This study provides a novel insight into the assembly of most abundant multi-subunit secretory proteins

    The development of high speed mobile networks and the widespread use of smartphones have enabled users to easily obtain large data volumes via the Internet. This causes a heavy consumption of network resources, a burden on the available bandwidth. To solve such problems, a data offloading method with a wireless LAN access point has been used to distribute traffic from mobile to fixed networks. However, the method using wireless LAN access points can only change the communication paths but cannot reduce the overall traffic. This paper proposes a local cooperative data offloading system (LoCO) that reduces the overall traffic by sharing data, with direct communication between neighbors based on their location-related information. Moreover, the authors implemented the LoCO system on Android smartphones and clarified its performance in comparison with a traditional client/server system through experiments to download data in a real-world environment

    Effect of shelter acclimation on the post-release movement and putative predation mortality of hatchery-reared black-spot tuskfish Choerodon schoenleinii, determined by acoustic telemetry

    In this study, the effect of shelter acclimation on the post-release movement and putative predation mortality of hatchery-reared black-spot tuskfish Choerodon schoenleinii was examined using acoustic telemetry. We acclimated four 1-year-old fish to shelters in cages before release and compared their movements with six nonacclimated fish. Since it was not possible to compare the behavioral pattern between the former and the latter fish due to the short periods the latter fish were available to be monitored, we also compared their movements with those of large nonacclimated fish that were less likely to be preyed upon. Sixty-seven percent of the nonacclimated fish showed atypical movements before the signals ceased to be detected, a pattern that suggested a predation event had occurred, whereas none of the acclimated and large nonacclimated fish showed the atypical movements. In addition, the probability of detection cessation was about 13 times lower in the acclimated than nonacclimated fish. The signal detection patterns suggest that the acclimated fish utilized night-time shelters from the first night after release, while the large nonacclimated fish started to utilize shelters several days after release. Therefore, it is likely that the shelter acclimation enhanced the shelter utilization by tuskfish, possibly decreasing post-release predation mortality

In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection

    DOCK2 is involved in the host genetics and biology of severe COVID-19

Here we conducted a genome-wide association study (GWAS) involving 2, 393 cases of COVID-19 in a cohort of Japanese individuals collected during the initial waves of the pandemic, with 3, 289 unaffected controls. We identified a variant on chromosome 5 at 5q35 (rs60200309-A), close to the dedicator of cytokinesis 2 gene (DOCK2), which was associated with severe COVID-19 in patients less than 65 years of age. This risk allele was prevalent in East Asian individuals but rare in Europeans, highlighting the value of genome-wide association studies in non-European populations. RNA-sequencing analysis of 473 bulk peripheral blood samples identified decreased expression of DOCK2 associated with the risk allele in these younger patients. DOCK2 expression was suppressed in patients with severe cases of COVID-19. Single-cell RNA-sequencing analysis (n = 61 individuals) identified cell-type-specific downregulation of DOCK2 and a COVID-19-specific decreasing effect of the risk allele on DOCK2 expression in non-classical monocytes. Immunohistochemistry of lung specimens from patients with severe COVID-19 pneumonia showed suppressed DOCK2 expression. Moreover, inhibition of DOCK2 function with CPYPP increased the severity of pneumonia in a Syrian hamster model of SARS-CoV-2 infection, characterized by weight loss, lung oedema, enhanced viral loads, impaired macrophage recruitment and dysregulated type I interferon responses. We conclude that DOCK2 has an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19, and could be further explored as a potential biomarker and/or therapeutic target
