We examined the method for determining microquantities of lipids, including cholesterol, cholesteryl esters and phospholipids. A standard colorimetric procedure of cholesteryl esters was modified to accommodate a quantitative thin-layer chromatography. This method involved the following steps. (1) Separation of lipids by a thin-layer chromatography: Lipids were applied to Silica gel G plates. Plates were developed with petroleum ether-diethyl etheracetic acid (82: 18: 2, vIvIv). (2) Elution of cholesterol and its esters from scraped silica gel: After scraping the silica gel with adhered cholesterol and its esters, they were eluted with chloroform-methanol (4: 1, v,tv). In the case of phspholipids, the silica gel was calcified. (3) Colorimetric determination of the lipids: Cholesterol and its esters eluted from the silica gel were determined by the method of ZAK with ROSENTHAL'S color reagent directly and after saponification, respectively. Phospholipids were calculated from the phosphorous content determined by the method of KATES. On the basis of examination of recovery and analyses of lipids extracted from tissue, it was concluded that this method permitted a reliable estimation of microquantities of cholesterol, its esters and phospholipids from small amounts of biological materials.</p

Zone Diagrams in Euclidean Spaces and in Other Normed Spaces

Zone diagram is a variation on the classical concept of a Voronoi diagram. Given n sites in a metric space that compete for territory, the zone diagram is an equilibrium state in the competition. Formally it is defined as a fixed point of a certain "dominance" map. Asano, Matousek, and Tokuyama proved the existence and uniqueness of a zone diagram for point sites in Euclidean plane, and Reem and Reich showed existence for two arbitrary sites in an arbitrary metric space. We establish existence and uniqueness for n disjoint compact sites in a Euclidean space of arbitrary (finite) dimension, and more generally, in a finite-dimensional normed space with a smooth and rotund norm. The proof is considerably simpler than that of Asano et al. We also provide an example of non-uniqueness for a norm that is rotund but not smooth. Finally, we prove existence and uniqueness for two point sites in the plane with a smooth (but not necessarily rotund) norm.Comment: Title page + 16 pages, 20 figure

This paper aims that recognition about Disaster prevention of the New Students entered the faculty of engineering the University of Tokushima in April, 2006. The authors used a questionnaire for investigation and analyzed it. As a result of having analyzed it, most of students cannot understand disaster prevention and more than half of them are single lives, but the students are interested in volunteer activity. By a disaster of the past, a youth is a lot dead next to an old man, too. The reason is because the students were crushed under furniture when an earthquake was generated a student lives in a simple building and a room is small. If students do volunteer activity of a disaster, it is very dangerous. The reason is because it leads to not only a primary disaster but also a second disaster. In other words, the students improve disaster prevention recognition and must understand disasters

Studies of interaction of homo-dimeric ferredoxin-NAD(P)+ oxidoreductases of Bacillus subtilis and Rhodopseudomonas palustris, that are closely related to thioredoxin reductases in amino acid sequence, with ferredoxins and pyridine nucleotide coenzymes

金沢大学理工研究域物質化学系Ferredoxin-NADP+ oxidoreductases (FNRs) of Bacillus subtilis (YumC) and Rhodopseudomonas palustris CGA009 (RPA3954) belong to a novel homo-dimeric type of FNR with high amino acid sequence homology to NADPH-thioredoxin reductases. These FNRs were purified from expression constructs in Escherichia coli cells, and their steady-state reactions with [2Fe-2S] type ferredoxins (Fds) from spinach and R. palustris, [4Fe-4S] type Fd from B. subtilis, NAD(P)+/NAD(P)H and ferricyanide were studied. From the Km and kcat values for the diaphorase activity with ferricyanide, it is demonstrated that both FNRs are far more specific for NADPH than for NADH. The UV-visible spectral changes induced by NADP+ and B. subtilis Fd indicated that both FNRs form a ternary complex with NADP+ and Fd, and that each of the two ligands decreases the affinities of the others. The steady-state kinetics of NADPH-cytochrome c reduction activity of YumC is consistent with formation of a ternary complex of NADPH and Fd during catalysis. These results indicate that despite their low sequence homology to other FNRs, these enzymes possess high FNR activity but with measurable differences in affinity for different types of Fds as compared to other more conventional FNRs. © 2008 Elsevier B.V. In the following Nankai Earthquake Tsunami, the characteristics of the tsunami height and its arrival time in the coastal area of the Seto Inland Sea was discussed. It found that their values could be estimated by the simple operation of Aida’s Ansei Nankai Earthquake Model(1981). Next, it proposed the evacuation model as the death toll increases as to be behind in the taking evacuation beginning time for the example of Usa , Kochi Prefecture . Moreover, based on this method, it measured the opening and shutting effect of the gates which was put to the embankment to exert on the death toll, and then it evaluated the effectivity of the protection against disasters

Continuous Flow Synthesis of ZSM-5 Zeolite on the Order of Seconds

    Get PDF
Zeolites have typically been synthesized via hydrothermal treatment, a process designed to artificially mimic the geological formation conditions of natural zeolites. This synthesis route, typically carried out in batch reactors like autoclaves, takes a time so long (typically, on the order of days) that the crystallization of zeolites had long been believed to be very slow in nature. Long periods of hydrothermal treatment also cause a burden on both energy efficiency and operational costs. Recently, we have reported the ultrafast syntheses of a class of industrially important zeolites within several minutes.[1,2] Further shortening the crystallization time to the order of seconds would be a great challenge but can significantly benefit the mass product of zeolites as well as the fundamental understanding of the crystallization mechanism

Ubiquitin-Specific Protease 2 Modulates the Lipopolysaccharide-Elicited Expression of Proinflammatory Cytokines in Macrophage-like HL-60 Cells

We investigated the regulatory roles of USP2 in mRNA accumulation of proinflammatory cytokines in macrophage-like cells after stimulation with a toll-like receptor (TLR) 4 ligand, lipopolysaccharide (LPS). Human macrophage-like HL-60 cells, mouse macrophage-like J774.1 cells, and mouse peritoneal macrophages demonstrated negative feedback to USP2 mRNA levels after LPS stimulation, suggesting that USP2 plays a significant role in LPS-stimulated macrophages. USP2 knockdown (KD) by short hairpin RNA in HL-60 cells promoted the accumulation of transcripts for 25 of 104 cytokines after LPS stimulation. In contrast, limited induction of cytokines was observed in cells forcibly expressing the longer splice variant of USP2 (USP2A), or in peritoneal macrophages isolated from Usp2a transgenic mice. An ubiquitin isopeptidase-deficient USP2A mutant failed to suppress LPS-induced cytokine expression, suggesting that protein ubiquitination contributes to USP2-mediated cytokine repression. Although USP2 deficiency did not accelerate TNF receptor-associated factor (TRAF) 6-nuclear factor-κB (NF-κB) signaling, it increased the DNA binding ratio of the octamer binding transcription factor (Oct)-1 to Oct-2 in TNF, CXCL8, CCL4, and IL6 promoters. USP2 decreased nuclear Oct-2 protein levels in addition to decreasing the polyubiquitination of Oct-1. In summary, USP2 modulates proinflammatory cytokine induction, possibly through modification of Oct proteins, in macrophages following TLR4 activation
