    Photoreactions of retinochrome at very low temperatures

    AbstractRetinochrome is one of the retinal proteins found in the retina of cephalopods. It catalyses the light isomerization of retinal from the all-trans to the 11-cis form. On cooling to 25 K, the absorption peak of retinochrome (λmax 490 nm) was broadened with a shoulder, showing the spectrum steepened on the long wavelength side. On irradiation with yellow-green light (550 nm), retinochrome produced an intermediate with λmax at a shorter wavelength, around 465 nm, and a lower extinction coefficient than lumiretinochrome. It changed to lunuretinochrome (λmax 475 nm) in the dark on warming to liquid nitrogen temperature. We shall call this new intermediate prelunuretinochrome

    The significant expression of TRPV3 in nasal polyps of eosinophilic chronic rhinosinusitis

    BackgroundThe number of patients with eosinophilic chronic rhinosinusitis (ECRS) has been increasing in recent years in Japan. In ECRS, nasal polyps recur immediately after endoscopic sinus surgery. The molecular biological mechanism underlying the refractoriness of ECRS is unclear.MethodsWhole-transcriptome analysis with next-generation sequencing (RNA-seq) was conducted to investigate the molecular biological mechanism of ECRS. Real-time PCR, immunohistochemical staining, and immunofluorescence staining were performed to validate the results of RNA-seq.ResultsRNA-seq analysis revealed that in the nasal polyps of ECRS, the levels of 3 transcripts were elevated significantly and those of 7 transcripts were diminished significantly. Among the genes encoding these transcripts, TRPV3 (transient receptor potential cation channel, subfamily V, member 3) was identified as the only gene that is highly expressed in ECRS nasal polyps but this gene\u27s expression was not previously detected using DNA microarray analysis in peripheral blood eosinophils. TRPV3 is newly identified here as a gene transcribed in ECRS. Our analysis also revealed that TRPV3 was highly expressed in the infiltrating eosinophils and mucosal epithelium of the nasal polyps of ECRS, and further that the more severe the refractoriness was after surgery, the higher the TRPV3 expression was in nasal polyps.ConclusionsTRPV3 might play a role in the refractoriness of ECRS. Additional studies are required to evaluate the function of TRPV3 in ECRS

    HLA-VBSeq v2: improved HLA calling accuracy with full-length Japanese class-I panel

    HLA-VBSeq is an HLA calling tool developed to infer the most likely HLA types from high-throughput sequencing data. However, there is still room for improvement in specific genetic groups because of the diversity of HLA alleles in human populations. Here, we present HLA-VBSeq v2, a software application that makes use of a new Japanese HLA reference panel to enhance calling accuracy for Japanese HLA class-I genes. Our analysis showed significant improvements in calling accuracy in all HLA regions, with prediction accuracies achieving over 99.0, 97.8, and 99.8% in HLA-A, B and C, respectively

    Identifying participants with knee osteoarthritis likely to benefit from physical therapy education and exercise: A hypothesis-generating study

    Background: The purpose of this investigation was to undertake a hypothesis generating study to identify candidate variables that characterize people with knee osteoarthritis who are most likely to experience a positive response to exercise. Methods: One hundred and fifty participants with knee osteoarthritis participated in this observational, longitudinal study. All participants received a standard exercise intervention that consisted of 20-min sessions two to three times a week for three months. The classification and regression tree methodology (CART) was used to develop prediction of positive clinical outcome. Positive pain and disability outcomes (dependent variables) were defined as an improvement in pain intensity by \u3e50% or an improvement of five or more on the Oxford knee score, respectively. The predictor variables considered included age, sex, body mass index, knee osteoarthritis severity (Kellgren/Lawrence grade), pain duration, use of medication, range of knee motion, pain catastrophizing, self-efficacy and knee self-perception. Results: Fifty-five participants (36.6%) were classified as responders for pain intensity and 36.6% were classified as responders for disability. The CART model identified impairments in knee self-perception and knee osteoarthritis severity as the discriminators for pain intensity reduction following exercise. No variables predicted reduction of disability level following exercise. Conclusions: Such findings suggest that both body perception and osteoarthritis severity may play a role in treatment outcome with exercise. It also raises the possibility that those with higher levels of disrupted body perception may need additional treatment targeted at restoring body perception prior to undertaking exercise. Significance: Regardless age, sex, body mass index, pain duration, use of medication, knee range of motion, pain catastrophizing and self-efficacy, participants with knee osteoarthritis who report low levels of body perception disruption (a FreKAQ score ≦ 17) and minimal structural changes (KL grade I) demonstrate significantly better outcomes from exercise therapy than other participants

    Possible helimagnetic order in Co4+-containing perovskites Sr1-xCaxCoO3

    We systematically synthesized perovskite-type oxides Sr1-xCaxCoO3 containing unusually high valence Co4+ ions by a high pressure technique, and investigated the effect of systematic lattice change on the magnetic and electronic properties. As the Ca content x exceeds about 0.6, the structure changes from cubic to orthorhombic, which is supported by the first-principles calculations of enthalpy. Upon the orthorhombic distortion, the ground state remains to be apparently ferromagnetic with a slight drop of the Curie temperature. Importantly, the compounds with x larger than 0.8 show antiferromagnetic behavior with positive Weiss temperatures and nonlinear magnetization curves at lowest temperature, implying that the ground state is noncollinear antiferromagnetic or helimagnetic. Considering the incoherent metallic behavior and the suppression of the electronic specific heat at high x region, the possible emergence of a helimagnetic state in Sr1-xCaxCoO3 is discussed in terms of the band-width narrowing and the double-exchange mechanism with the negative charge transfer energy as well as the spin frustration owing to the next-nearest neighbor interaction.Comment: 13 pages, 4 figure

    Basic Study on Positive Effects of Radon Inhalation on Pet's Health

    著者らは今までに,共同開発したラドン吸入装置を用いマウスにラドン吸入をさせた場合,諸臓器中の抗酸化機能が亢進する可能性などを明らかにしてきた。本研究では,ラドン吸入の獣医療への応用の可能性について新たに検討するため,健常なイヌ5頭(オス:2(1,9才),メス:3(1~5才))及び慢性腎不全症のネコ8頭(オス:3(2~6才),メス:5(5~7才))を対象に基礎的な検討をした。すなわち,約5500Bq/m3のラドンを1回30分で隔日に30日間(計15回)それぞれ吸入させた。その結果,イヌにおいて,中性脂肪が減少する可能性が示された。また,その効果は吸入開始20~30日後に現れることも示唆できた。他方,ネコにおいて,飲水量が改善し血清中クレアチニンが基準値内に減少する症例がみられるなど,慢性腎不全症に対し一定の効果が期待できる可能性が示唆された。Radon inhalation using our radon exposure device activated anti-oxidative function in some organs of mouse. To assess the possibility of its application to veterinary care, healthy dogs and cats with chronic renal failure were inhaled radon at a concentration of 5500Bq/m3 for 30 minutes every 2 days for 30 days. In result, radon inhalation within a relatively long time period significantly decreased the triglyceride level of dogs. On the other hand, some cats increased the volume of drinking water by radon inhalation and the creatinine level in blood of these cats was decreased to normal level. These findings suggest that radon inhalation may have curative properties against chronic renal failure

    Periostin as a novel biomarker for postoperative recurrence of chronic rhinosinitis with nasal polyps

    We previously reported that chronic rhinosinusitis with nasal polyps (CRSwNP) was subdivided into four chronic rhinosinusitis (CRS) subtypes using the JESREC scoring system. We sought to identify the gene expression profile and biomarkers related with CRSwNP by RNA-sequence. RNA-sequencing was performed to identify differentially expressed genes between nasal polyps (NPs) and inferior turbinate mucosa from 6 patients with CRSwNP, and subsequently, quantitative real-time PCR was performed to verify the results. ELISA was performed to identify possible biomarkers for postoperative recurrence. In the RNA-sequencing results, periostin (POSTN) expression was the highest in NP. We focused on POSTN and investigated the protein level of POSTN by immunohistochemistry and ELISA. POSTN was diffusely expressed in moderate and severe eosinophilic CRS using immunohistochemistry, and its staining pattern was associated with the severity of the phenotype of the CRSwNP (P < 0.05). There was a significant difference between the POSTN high/low groups for postoperative recurrence when the cutoff point was set at 115.5 ng/ml (P = 0.0072). Our data suggests that the protein expression level of POSTN was associated with the severity of CRSwNP, and serum POSTN can be a novel biomarker for postoperative recurrence of CRSwNP

    A comparative method for evaluating ecosystem services from the viewpoint of public works

    Nature-based solutions (NbS), such as the implementation of environmental conservation and restoration as public works projects, require accurate and cost-effective assessments of the values related to the projects. The values should represent collective ecosystem services, individual services such as food provision and water purification, and other intangible services. To comprehensively assess such services, we proposed a novel method, which we call the comparative evaluation method. Our method is able to assess the value of each service category of an NbS project from a single questionnaire survey. Survey participants are asked to compare values of multiple services having anchoring prices. Our method determines the permissible economic value of environmental public works (PEP) in response to the quantity of service. The questionnaire results used for analysis are limited to those from respondents who made their PEP evaluation on the basis of their consideration of the appropriate expenditure of taxes. In addition, the method controls for the effect of the satisfaction that a person experiences from doing good deeds to reduce an overestimation of the values of services. Moreover, PEPs are not influenced by the respondent's annual income, age, sex, or educational background, and are based on personal values. Applying this new method, we surveyed residents of the watersheds of Tokyo Bay and Osaka Bay and evaluated nine ecosystem services. Overall, our new method is shown to be an effective method for evaluating the ecosystem services of NbS projects from the viewpoint of public works
