    Economic effects of Pacific halibut closures on businesses on the North Coast and the age, growth, and reproductive status of Pacific halibut in northern California and central Oregon

    Traditionally, the recreational fishery for Pacific halibut has been open in California from 1 May through 31 October. In 2014, however, the Pacific halibut fishery was closed in California during the month of August for the first time in history in an effort to reduce harvest and bring total catch closer to what is allocated to our region by the Pacific Fisheries Management Council (PFMC) Catch Sharing Plan. To determine the effects that the closure had on businesses along the North Coast, I conducted an economic impact survey in 2014. The results of the survey showed that fishing-related businesses lost between 0 percent and 8 percent of their revenue in 2014, as a result of the closure; lodging and traveler service companies lost between 0.3 percent and 1 percent of their revenue in the same year. None of the businesses changed the number of employees as a result of the closure. We estimated a decrease in revenue for businesses on the North Coast to be between 189,750and189,750 and 222,250. Age and growth are important components in stock assessment models, but biological data in general are scarce on populations of Pacific halibut found in northern California. For this reason, I conducted a study that examined the age and growth of Pacific halibut landed in this region, expanding on a previous study to examine possible interannual variation in the age/growth structure, and broadened the study into central Oregon, to compare two distinct bioregions. Results from my study show that mean size-at-age of female Pacific halibut from northern California and central Oregon was larger than those from the IPHC setline surveys in most of Alaska, but similar to those from Oregon and Washington. In addition, fish from this study in northern California and central Oregon were smaller for a given age than those from the 2014 IPHC survey conducted in northern California. Possible reasons for the trend in size-at-age include poor oceanic conditions during my study, the movement of slower-growing halibut into northern Californian waters, and sampling error. The maturity stage of female gonads is also an important component in stock assessment models, but these data are also scarce for Pacific halibut populations in northern California. For this reason, I conducted a study that characterized the maturation of Pacific halibut landed in northern California and central Oregon. I also compared the macroscopic maturity staging method currently utilized by the IPHC against the more rigorous microscopic methods (microscopic staging and measuring oocyte diameter). Results of this study and that of Perkins (2015) indicate that Pacific halibut caught in northern California and central Oregon matured three years earlier than those caught during IPHC setline surveys in waters off of Alaska, and about a year earlier than those caught by the IPHC in Oregon and Washington. The length-at-50%-maturity in Pacific halibut caught in northern California and central Oregon was smaller than that of fish caught in the IPHC setline survey. In addition, for all three stages of maturity observed in females (immature, mature, and resting; spawning-stage females were not observed) there was at least 66 percent agreement between macroscopic and histological staging methods, with the highest level of agreement (94 percent) seen in mature ovaries. This study largely validated the macroscopic staging methods because of its high accuracy in identifying mature ovaries; the inaccuracy in distinguishing resting versus immature ovaries had little effect on length- and age-at-maturity analysis

    Interploidy gene flow involving the sexual-asexual cycle facilitates the diversification of gynogenetic triploid Carassius fish

We explore the mechanisms behind the generation and maintenance of genetic diversity in triploid asexual (gynogenetic) Carassius auratus fish, which is widespread in East Asian fresh waters and exhibits one of the most extensive distribution among asexual vertebrates despite its dependence on host sperm. Our analyses of genetic composition using dozens of genetic markers and genome-wide transcriptome sequencing uncover admixed genetic composition of Japanese asexual triploid Carassius consisting of both the diverged Japanese and Eurasian alleles, suggesting the involvement of Eurasian lineages in its origin. However, coexisting sexual diploid relatives and asexual triploids in Japan show regional genetic similarity in both mitochondrial and nuclear markers. These results are attributed to a unique unidirectional gene flow from diploids to sympatric triploids, with the involvement of occasional sexual reproduction. Additionally, the asexual triploid shows a weaker population structure than the sexual diploid, and multiple triploid lineages coexist in most Japanese rivers. The generated diversity via repeated interploidy gene flow as well as an increased establishment of immigrants is assumed to offset the cost of asexual reproduction and might contribute to the successful broad distribution of this asexual vertebrate

    Quality of life of children with neurodevelopmental disorders and their parents during the COVID-19 pandemic : a 1-year follow-up study

    This study aimed to reveal changes in the quality of life (QOL) of children with neurodevelopmental disorders and their parents, and the interaction between their QOL and parental mental state during the coronavirus 2019 (COVID-19) pandemic. Eighty-nine school-aged children and parents participated in surveys in May 2020 (T1) and May 2021 (T2). The parents completed questionnaires that assessed their QOL, depression, parenting stress, and living conditions. Children's temporary mood status was evaluated using the self-reported visual analog scale (VAS). Children's QOL and VAS at T2 were higher than their QOL at T1. Parents' QOL at T2 was lower than their QOL at T1. Severe parental depression at T1 had a synergistic effect on severe parenting stress and severe depressive state at T2. Additionally, children's high QOL at T1 had a synergistic effect on low parenting stress and children's high QOL at T2. Furthermore, children's low VAS scores and parents' low QOL at T2 were associated with deterioration of family economic status. Children and parents' QOL changed during the prolonged COVID-19 pandemic. Improvement in children's QOL was influenced by reduced maternal depressive symptoms. Public support for parental mental health is important to avoid decreasing QOL.Peer reviewe

    Subtype-specific gout susceptibility loci and enrichment of selection pressure on ABCG2 and ALDH2 identified by subtype genome-wide meta-analyses of clinically defined gout patients

    Objectives Genome-wide meta-analyses of clinically defined gout were performed to identify subtype-specific susceptibility loci. Evaluation using selection pressure analysis with these loci was also conducted to investigate genetic risks characteristic of the Japanese population over the last 2000–3000 years. Methods Two genome-wide association studies (GWASs) of 3053 clinically defined gout cases and 4554 controls from Japanese males were performed using the Japonica Array and Illumina Array platforms. About 7.2 million single-nucleotide polymorphisms were meta-analysed after imputation. Patients were then divided into four clinical subtypes (the renal underexcretion type, renal overload type, combined type and normal type), and meta-analyses were conducted in the same manner. Selection pressure analyses using singleton density score were also performed on each subtype. Results In addition to the eight loci we reported previously, two novel loci, PIBF1 and ACSM2B, were identified at a genome-wide significance level (p<5.0×10–8) from a GWAS meta-analysis of all gout patients, and other two novel intergenic loci, CD2-PTGFRN and SLC28A3-NTRK2, from normal type gout patients. Subtype-dependent patterns of Manhattan plots were observed with subtype GWASs of gout patients, indicating that these subtype-specific loci suggest differences in pathophysiology along patients’ gout subtypes. Selection pressure analysis revealed significant enrichment of selection pressure on ABCG2 in addition to ALDH2 loci for all subtypes except for normal type gout. Conclusions Our findings on subtype GWAS meta-analyses and selection pressure analysis of gout will assist elucidation of the subtype-dependent molecular targets and evolutionary involvement among genotype, phenotype and subtype-specific tailor-made medicine/prevention of gout and hyperuricaemia

    Role of ER Stress in Ventricular Contractile Dysfunction in Type 2 Diabetes

    BACKGROUND: Diabetes mellitus (DM) is associated with an increased risk of ischemic heart disease and of adverse outcomes following myocardial infarction (MI). Here we assessed the role of endoplasmic reticulum (ER) stress in ventricular dysfunction and outcomes after MI in type 2 DM (T2DM). METHODOLOGY AND PRINCIPAL FINDINGS: In hearts of OLETF, a rat model of T2DM, at 25∼30 weeks of age, GRP78 and GRP94, markers of ER stress, were increased and sarcoplasmic reticulum calcium ATPase (SERCA)2a protein was reduced by 35% compared with those in LETO, a non-diabetic control. SERCA2a mRNA levels were similar, but SERCA2a protein was more ubiquitinated in OLETF than in LETO. Left ventricular (LV) end-diastolic elastance (Eed) was higher in OLETF than in LETO (53.9±5.2 vs. 20.2±5.6 mmHg/µl), whereas LV end-systolic elastance and positive inotropic responses to β-adrenergic stimulation were similar in OLETF and LETO. 4-Phenylbutyric acid (4-PBA), an ER stress modulator, suppressed both GRP up-regulation and SERCA2a ubiquitination and normalized SERCA2a protein level and Eed in OLETF. Sodium tauroursodeoxycholic acid, a structurally different ER stress modulator, also restored SERCA2a protein level in OLETF. Though LV dysfunction was modest, mortality within 48 h after coronary occlusion was markedly higher in OLETF than in LETO (61.3% vs. 7.7%). Telemetric recording showed that rapid progression of heart failure was responsible for the high mortality rate in OLETF. ER stress modulators failed to reduce the mortality rate after MI in OLETF. CONCLUSIONS: ER stress reduces SERCA2a protein via its augmented ubiquitination and degradation, leading to LV diastolic dysfunction in T2DM. Even at a stage without systolic LV dysfunction, susceptibility to lethal heart failure after infarction is markedly increased, which cannot be explained by ER stress or change in myocardial response to sympathetic nerve activation

    Conjugated docosahexaenoic acid suppresses KPL-1 human breast cancer cell growth in vitro and in vivo: potential mechanisms of action

    Introduction The present study was conducted to examine the effect of conjugated docosahexaenoic acid (CDHA) on cell growth, cell cycle progression, mode of cell death, and expression of cell cycle regulatory and/or apoptosis-related proteins in KPL-1 human breast cancer cell line. This effect of CDHA was compared with that of docosahexaenoic acid (DHA). Methods KPL-1 cell growth was assessed by colorimetric 3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay; cell cycle progression and mode of cell death were examined by flow cytometry; and levels of expression of p53, p21Cip1/Waf1, cyclin D1, Bax, and Bcl-2 proteins were examined by Western blotting analysis. In vivo tumor growth was examined by injecting KPL-1 cells subcutaneously into the area of the right thoracic mammary fat pad of female athymic mice fed a CDHA diet. Results CDHA inhibited KPL-1 cells more effectively than did DHA (50% inhibitory concentration for 72 hours: 97 μmol/l and 270 μmol/l, respectively). With both CDHA and DHA growth inhibition was due to apoptosis, as indicated by the appearance of a sub-G1 fraction. The apoptosis cascade involved downregulation of Bcl-2 protein; Bax expression was unchanged. Cell cycle progression was due to G0/G1 arrest, which involved increased expression of p53 and p21Cip1/Waf1, and decreased expression of cyclin D1. CDHA modulated cell cycle regulatory proteins and apoptosis-related proteins in a manner similar to that of parent DHA. In the athymic mouse system 1.0% dietary CDHA, but not 0.2%, significantly suppressed growth of KPL-1 tumor cells; CDHA tended to decrease regional lymph node metastasis in a dose dependent manner. Conclusion CDHA inhibited growth of KPL-1 human breast cancer cells in vitro more effectively than did DHA. The mechanisms of action involved modulation of apoptosis cascade and cell cycle progression. Dietary CDHA at 1.0% suppressed KPL-1 cell growth in the athymic mouse system.</p

    Study of spin-isospin responses of radioactive nuclei with the background-reduced neutron spectrometer, PANDORA

    The status of a project to measure spin-isospin responses of neutron drip-line nuclei using a new low-energy neutron detector, PANDORA (Particle Analyzer Neutron Detector Of Real-time Acquisition), is reported. The performance of PANDORA was characterized by the 6He(p,n)6Li reaction in inverse kinematics at the HIMAC facility in Chiba. Observation of the strong transition to the ground state in 6Li is discussed. Preliminary results of 11Li(p,n)11Be and 14Be(p,n)14B experiments in inverse kinematics at RI Beam Factory (RIBF) of RIKEN Nishina Center are also presented including the exotic decay channel of 11Be→9Li + d. Details of the experimental setup based on PANDORA and the SAMURAI large-acceptance magnetic spectrometer, as well as the combined data-acquisition system are described. The neutron-gamma discrimination capability of PANDORA was evaluated, Figure-of-Merit (FoM) values higher than those found in the literature for similar materials were derived from experimental data
