Current understanding of imeglimin action on pancreatic β‐cells: Involvement of mitochondria and endoplasmic reticulum homeostasis

    Get PDF
Recent preclinical studies have provided insight on imeglimin's action on pancreatic β-cells and the mechanisms underlying its clinical benefits. Imeglimin may enhance glucose-induced insulin secretion (GIIS) and inhibit apoptosis of pancreatic ß-cells leading to preserved β-cell mass by maintaining or restoring the functional and structural integrity of the mitochondria and the endoplasmic reticulum homeostasis in pancreatic β-cells

High-temperature thermoelectric properties of the double-perovskite ruthenium oxide (Sr1x_{1-x}Lax_x)2_2ErRuO6_6

We have prepared polycrystalline samples of (Sr1x_{1-x}Lax_x)2_2ErRuO6_6 and (Sr1x_{1-x}Lax_x)2_2YRuO6_6, and have measured the resistivity, Seebeck coefficient, thermal conductivity, susceptibility and x-ray absorption in order to evaluate the electronic states and thermoelectric properties of the doped double-perovskite ruthenates. We have observed a large Seebeck coefficient of -160 μ\muV/K and a low thermal conductivity of 7 mW/cmK for xx=0.1 at 800 K in air. These two values are suitable for efficient oxide thermoelectrics, although the resistivity is still as high as 1 Ω\Omegacm. From the susceptibility and x-ray absorption measurements, we find that the doped electrons exist as Ru4+^{4+} in the low spin state. On the basis of the measured results, the electronic states and the conduction mechanism are discussed.Comment: 6 pages, 4 figures, J. Appl. Phys. (accepted

Background: Desmopressin orally disintegrating tablet (ODT) was approved in March 2012 in Japan; the post-market safety reports, which warned about adequate initial dose of desmopressin ODT, were published in 2014. However, it is unclear how the warning affected physician and patient behavior. Methods: We performed a retrospective single-center study to compare the clinical situation of Japanese central diabetes insipidus patients before and after the report. Results: Thirty-four patients before October 2014 and 16 patients after November 2014 switched from intranasal desmopressin to desmopressin ODT. The mean follow-up period after the switch to desmopressin ODT was 38 ± 3 months. Patients switching after November 2014 tended to have lower ratios of oral to nasal desmopressin dose at switching and 3 months after the switch (at switching; P = 0.20, 3 months; P = 0.42, respectively), and higher ratios from 6 to 12 months than before October 2014 (6 months; P = 0.93, 9 months; P = 0.52, 12 months; P = 0.80, respectively). Relative doses per initial desmopressin ODT at 9 and 12 months were significantly higher in patients switching after November 2014 than in patients switching before October 2014 (9 months; P = 0.02, 12 months; P = 0.04, respectively). Moreover, logistic regression analysis revealed that the incidence of hyponatremia was dependent on the ratio of nasal to oral desmopressin dose (P = 0.02). In addition, in four out of six patients who had serum sodium level reduced below 130 mEq/L, hyponatremia occurred within 1 month after the switch. Conclusions: A more gradual dose titration after the safety reports was performed, which involved the long-term safety of desmopressin ODT use. Vigilance of hyponatremia in early phase of desmopressin ODT use should be noted

Disability Prevention Programs for Older People: Factors Associated with Medical and Nursing Care Costs

    Get PDF
This study aimed to clarify factors associated with medical and nursing care costs for older people living in community and to suggest an effective disability prevention programs. Total of participants in this study was 83 individuals (29 men and 54 women; mean age 81.2 ± 6.3 years old) on November 1st – December 28th, 2014. This study compared the average medical and nursing care costs per month with national average for those aged ≥ 65 years old. Logistic regression test was conducted to examine its association with medical and nursing care costs. Those who had outing activities ≥ 3 times a week were approximately three times less likely to reduce medical and nursing care costs than those who had outing activities < 3 times a week despite three controlled covariates (OR = 3.23 and 95% CI = 1.03 – 10.42). Disability prevention programs that improve frequency of outing at least three times in a week may become a valid economic approach to older people who do not live in nursing home

Noninvasive quantitative evaluation of viable islet grafts using ¹¹¹In-exendin-4 SPECT/CT

Islet transplantation (IT) is an effective β-cell replacement therapy for patients with type 1 diabetes; however, the lack of methods to detect islet grafts and evaluate their β-cell mass (BCM) has limited the further optimization of IT protocols. Therefore, the development of noninvasive β-cell imaging is required. In this study, we investigated the utility of the ¹¹¹Indium-labeled exendin-4 probe {[Lys12(111In-BnDTPA-Ahx)] exendin-4} (¹¹¹In exendin-4) to evaluate islet graft BCM after intraportal IT. The probe was cultured with various numbers of isolated islets. Streptozotocin-induced diabetic mice were intraportally transplanted with 150 or 400 syngeneic islets. After a 6-week observation following IT, the ex-vivo liver graft uptake of ¹¹¹In-exendin-4 was compared with the liver insulin content. In addition, the in-vivo liver graft uptake of ¹¹¹In exendin-4 using SPECT/CT was compared with that of liver graft BCM measured by a histological method. As a result, probe accumulation was significantly correlated with islet numbers. The ex-vivo liver graft uptake in the 400-islet-transplanted group was significantly higher than that in the control and the 150-islet-transplanted groups, consistent with glycemic control and liver insulin content. In conclusion, in-vivo SPECT/CT displayed liver islet grafts, and uptakes were corroborated by histological liver BCM. ¹¹¹In-exendin-4 SPECT/CT can be used to visualize and evaluate liver islet grafts noninvasively after intraportal IT

A consortium study of Antarctic micrometeorites recovered from the Dome Fuji Station

Deposits in the water tank at the Dome Fuji Station were collected by the 37th Japanese Antarctic Research Expedition team in 1996. We recovered 233 micrometeorites from the deposits. A consortium study was started in late 1998 to investigate mineralogy, petrology, bulk chemistry, and isotopic compositions of the micrometeorites. This is the first case of an organized study of micrometeorites in Japan, in order to establish the methods to investigate micrometeorites routinely. Consortium results on mineralogy, petrology, minor and trace element compositions, isotopic compositions of noble gases of the micrometeorites are reported in this volume. We also found a sequence of mineralogical and compositional changes of micrometeorites experienced from frictional heating during atmospheric entry. INAA and ion probe studies are now in progress

Bidirectional Longitudinal Association between Back Pain and Loneliness in Later Life: Evidence from English Longitudinal Study of Ageing

Background This study examined the bidirectional and temporal-ordinal relationship between loneliness and back pain. Methods Data from 7,730 participants in waves 6 (2012–2013), 7 (2014–2015), and 8 (2016–2017) of the national English Longitudinal Study of Ageing were analyzed. Back pain was graded on a scale of 0–10 (0, no discomfort; 10, unbearable pain). Loneliness was measured using the Revised University of California Los Angeles Loneliness Scale. A targeted minimum loss-based estimator was used to examine the bidirectional longitudinal associations between back pain and loneliness. Results No loneliness in waves 6 and 7 (relative risk [RR]=0.76; 95% confidence interval [CI], 0.61–0.94), no loneliness in wave 6 but loneliness in wave 7 (RR=0.58; 95% CI, 0.50–0.68), and loneliness in wave 6 but not in wave 7 (RR=0.69; 95% CI, 0.57–0.86) were associated with significant risk reductions of back pain in wave 8 compared with the scenario of loneliness in waves 6 and 7. Mild back pain in wave 6 but moderate back pain (RR=0.55; 95% CI, 0.35–0.86) or severe back pain in wave 7 (RR=0.49; 95% CI, 0.34–0.72) showed a significant risk reduction of loneliness in wave 8 compared with severe back pain in waves 6 and 7. Conclusion Loneliness may be a risk factor for back pain, and back pain may be a risk factor for loneliness. The results of this study will inform the development of more effective interventions for loneliness and back pain

    Quantum Corrections on Fuzzy Sphere

We investigate quantum corrections in non-commutative gauge theory on fuzzy sphere. We study translation invariant models which classically favor a single fuzzy sphere with U(1) gauge group. We evaluate the effective actions up to the two loop level. We find non-vanishing quantum corrections at each order even in supersymmetric models. In particular the two loop contribution favors U(n) gauge group over U(1) contrary to the tree action in a deformed IIB matrix model with a Myers term. We further observe close correspondences to 2 dimensional quantum gravity.Comment: 27 pages, 1 figure, published version in Nucl Phys.

Antarctic micrometeorites collected at the Dome Fuji Station

    Get PDF
Antarctic micrometeorites (AMMs) were found among the precipitated fine particles recovered from a water tank in the Dome Fuji Station. These AMMs had been contained in the recent fallen snow around the station. Initial processing of the precipitated particles revealed that they were dominated by natural and artificial terrestrial materials, thus a series of processes were developed to separate AMMs from terrestrial particles. The recovery rate of AMMs by the processes was approximately 45% in weight, which was determined from a weight ratio of recovered/accreted AMMs. The micro-morphology and major-element concentration of the recovered AMMs were characterized. They appear to have been heated upon atmospheric entry to varying temperatures and can be classified into two major types based on the degree of heating : (1) fine-grained, irregular-shaped, partial-melted micrometeorites with chondritic composition, and (2) total-melted spherical micrometeorites with chondritic composition except for volatile elements. A digital catalog for the AMMs identified in this study was established on the web site [URL : http : //dust. cc. gakushuin. ac. jp/], in which optical characteristics, high-resolution images, and chemical compositions of individual AMMs are presented. The AMMs listed in the catalog are the first Japanese collection of extraterrestrial dust. The criterion and techniques developed for the selection and initial analysis of AMMs are applicable for the dust samples that are being collected by the 39th Japanese Antarctic Research Expedition team
