    Incidence of Bacteria with Potential Public Health Implications in Raw Lycopersicon esculentum (Tomato) Sold in Lagos State, Nigeria

    ABSTRACTLycopersicon esculentum (tomato) is a fleshy berry that is highly perishable and prone to microbial spoilage. The presence of some microorganisms of public health significance makes it a potential health hazard to consumers. There is therefore the need to determine the food safety and public health implications of some of the microorganisms present in it. The aim of the study was to isolate, identify and compare the incidence of the organisms that could be potential pathogens among the four grades of tomato samples used in the investigation. Seven batches of tomato samples were obtained from two (2) major produce markets in Lagos Nigeria. A total of 40 isolates were obtained from the tomato samples. Staphylococcus aureus (22.5%), Escherichia coli (15%), Bacillus species (20%) and Salmonella species (5%) were among the bacteria isolated from the tomato samples. The average pH values of the tomato samples ranged from 4.34 to 4.60 while the average moisture content of the tomato samples was between 12% and 95.1%. The results showed that the freshness and hardness of tomato samples were important factors which determine the types of bacteria found on them. As some of the bacterial genera identified are potential food-borne pathogens that could pose some public health challenges, proper handling and adequate cooking before consumption is suggested

    Biological Resolution of Virulence Genes of Salmonella Species from different Microbiomes

    The pathogenic promiscuity of virulence associated macromolecules in Salmonella infection is a key driver to their wide epidemiology and curtailing  such distribution is contingent upon proper clarification of these virulence genes. This study was therefore aimed at determining the virulence  genes of Salmonella species from different microbiomes. To achieve this, a total of three hundred (300) biological specimens were aseptically  collected and processed for Salmonella presence using the BAM USFDA technique prior to their genotypic characterization while virulence gene  detection was carried out in a primer specific polymerase chain reaction. Results obtained depict the distribution of the following Salmonella species  viz; Salmonella gallinarum 19(26.39%), Salmonella heidelberg 19(26.39%), Salmonella enteritidis 18(25%) and Salmonella typhimurium  16(22.22%) while the occurrence of the virulence genes (InvA, SopE, AgfA and SpvC) were Salmonella enteritidis ( 7(38.8), 6(33.3), 9(50), 3(16.7),  Salmonella typhimurium ( 5(26.3), 3(15.8), 2(10.5), 7(36.8)), Salmonella heidelberg (0(0), 8(50), 4(25), 4(25), and Salmonella gallinarum (12(63.2),  6(31.6), 2(10.5), 7(36.8)) respectively. It was however found that the different microbiomes analyzed were ubiquitously rich in virulence genes  associated Salmonella species.   La promiscuitĂ© pathogène des macromolĂ©cules associĂ©es Ă  la virulence dans l’infection Ă  Salmonella est un facteur clĂ© de leur large Ă©pidĂ©miologie  et la rĂ©duction de cette distribution dĂ©pend de la clarification appropriĂ©e de ces gènes de virulence. Cette Ă©tude visait donc Ă  dĂ©terminer les gènes  de virulence des espèces de Salmonella de diffĂ©rents microbiomes.     The global retinoblastoma outcome study : a prospective, cluster-based analysis of 4064 patients from 149 countries

    DATA SHARING : The study data will become available online once all analyses are complete.BACKGROUND : Retinoblastoma is the most common intraocular cancer worldwide. There is some evidence to suggest that major differences exist in treatment outcomes for children with retinoblastoma from different regions, but these differences have not been assessed on a global scale. We aimed to report 3-year outcomes for children with retinoblastoma globally and to investigate factors associated with survival. METHODS : We did a prospective cluster-based analysis of treatment-naive patients with retinoblastoma who were diagnosed between Jan 1, 2017, and Dec 31, 2017, then treated and followed up for 3 years. Patients were recruited from 260 specialised treatment centres worldwide. Data were obtained from participating centres on primary and additional treatments, duration of follow-up, metastasis, eye globe salvage, and survival outcome. We analysed time to death and time to enucleation with Cox regression models. FINDINGS : The cohort included 4064 children from 149 countries. The median age at diagnosis was 23·2 months (IQR 11·0–36·5). Extraocular tumour spread (cT4 of the cTNMH classification) at diagnosis was reported in five (0·8%) of 636 children from high-income countries, 55 (5·4%) of 1027 children from upper-middle-income countries, 342 (19·7%) of 1738 children from lower-middle-income countries, and 196 (42·9%) of 457 children from low-income countries. Enucleation surgery was available for all children and intravenous chemotherapy was available for 4014 (98·8%) of 4064 children. The 3-year survival rate was 99·5% (95% CI 98·8–100·0) for children from high-income countries, 91·2% (89·5–93·0) for children from upper-middle-income countries, 80·3% (78·3–82·3) for children from lower-middle-income countries, and 57·3% (52·1-63·0) for children from low-income countries. On analysis, independent factors for worse survival were residence in low-income countries compared to high-income countries (hazard ratio 16·67; 95% CI 4·76–50·00), cT4 advanced tumour compared to cT1 (8·98; 4·44–18·18), and older age at diagnosis in children up to 3 years (1·38 per year; 1·23–1·56). For children aged 3–7 years, the mortality risk decreased slightly (p=0·0104 for the change in slope). INTERPRETATION : This study, estimated to include approximately half of all new retinoblastoma cases worldwide in 2017, shows profound inequity in survival of children depending on the national income level of their country of residence. In high-income countries, death from retinoblastoma is rare, whereas in low-income countries estimated 3-year survival is just over 50%.     Metagenomic tracking of microbial consortia of cassava flakes (garri)

    The affirmation of several cross-sectional studies on the vulnerability of cassava flakes commonly called 'garri' to microbial attack has long been documented. However, longitudinal data on metagenomic tracking of microbial consortia of this important staple food are scarce. Hence, this study was aimed at tracking the microbial consortia of garri. A total of eight samples (four each from both Nigeria and Republic of Benin markets) were randomly collected aseptically using pre-sterilized aluminum pans and processed through a metagenomic approach, while both the chemical and proximate components of garri were assessed following standard techniques. The analysis of the taxonomic consortia of garri reveals the predomination of bacteria (99.82 and 99.81% for samples from Nigeria and Republic of Benin, respectively) while the remaining sequences matched with the Archae (0.07%), fungi (0.09%) and protozoa (0.09%). A large proportion of the sequences were unclassified at the phylum level (approximately 84.10 and 86.2% for Nigerian and Beninese samples, respectively). The reads of cassava flakes metagenome of both Nigeria and Republic of Benin exhibited analogous level of average GC content with sequence count of between 187773-213444 for samples from Nigeria and 157784-198763 for samples from Republic of Benin. The functional characteristics of the inhabiting metagenomes were found containing the genes encoding for adhesins, bacteriocins, resistance to antibiotics, toxic chemicals as well as toxins and superantigens. Both the chemical and the proximate compositions of the examined garri samples, though exhibited significant disparity, but without any apparent variation in the patterns of metagenomic data.
