Phamacogenomics of Clozapine-Induced Agranulocytosis
Background: Clozapine-induced agranulocytosis (CIA)/clozapine-induced granulocytopenia (CIG) (CIAG) is a life-threatening event for schizophrenic subjects treated with clozapine.
Methods: To examine the genetic factor for CIAG, a genome-wide pharmacogenomic analysis was conducted using 50 subjects with CIAG and 2905 control subjects.
Results: We identified a significant association in the human leukocyte antigen (HLA) region (rs1800625, p = 3.46 × 10−9, odds ratio [OR] = 3.8); therefore, subsequent HLA typing was performed. We detected a significant association of HLA-B*59:01 with CIAG (p = 3.81 × 10−8, OR = 10.7) and confirmed this association by comparing with an independent clozapine-tolerant control group (n = 380, p = 2.97 × 10−5, OR = 6.3). As we observed that the OR of CIA (OR: 9.3~15.8) was approximately double that in CIG (OR: 4.4~7.4), we hypothesized that the CIG subjects were a mixed population of those who potentially would develop CIA and those who would not develop CIA (non-CIA). This hypothesis allowed the proportion of the CIG who were non-CIA to be calculated, enabling us to estimate the positive predictive value of the nonrisk allele on non-CIA in CIG subjects. Assuming this model, we estimated that 1) ~50% of CIG subjects would be non-CIA; and 2) ~60% of the CIG subjects without the risk allele would be non-CIA and therefore not expected to develop CIA.
Conclusions: Our results suggest that HLA-B*59:01 is a risk factor for CIAG in the Japanese population. Furthermore, if our model is true, the results suggest that rechallenging certain CIG subjects with clozapine may not be always contraindicated
A Self-Deformation Robot Design Incorporating Bending-Type Pneumatic Artificial Muscles
With robots becoming closer to humans in recent years, human-friendly robots made of soft materials provide a new line of research interests. We designed and developed a soft robot that can move via self-deformation toward the practical application of monitoring children and the elderly on a daily basis. The robot’s structure was built out of flexible frames, which are bending-type pneumatic artificial muscles (BPAMs). We first provide a description and discussion on the nature of BPAM, followed by static characteristics experiment. Although the BPAM theoretical model shares a similar tendency with the experimental results, the actual BPAMs moved along the depth direction. We then proposed and demonstrated an effective locomotion method for the robot and calculated its locomotion speed by measuring its drive time and movement distance. Our results confirmed the reasonability of the robot’s speed for monitoring children and the elderly. Nevertheless, during the demonstration, some BPAMs were bent sharply by other activated BPAMs as the robot was driving, leaving a little damage on these BPAMs. This will be addressed in our future work
Water-Assisted Perovskite Quantum Dots with High Optical Properties
Lead halide perovskite quantum dots (PeQDs) have excellent optical properties, such as narrow emission spectra (FWHM: 18–30 nm), a tunable bandgap (λPL: 420–780 nm), and excellent photoluminescence quantum yields (PLQYs: >90%). PeQDs are known as a material that is easily decomposed when exposed to water in the atmosphere, resulting in causing PeQDs to lower performance. On the other hand, according to the recent reports, adding water after preparing the PeQD dispersion decomposed the PeQD surface defects, resulting in improving their PLQY. Namely, controlling the amount of assisting water during the preparation of the PeQDs is a significantly critical factor to determining their optical properties and device applications. In this paper, our research group discovered the novel effects of the small amount of water to their optical properties when preparing the PeQDs. According to the TEM Images, the PeQDs particle size was clearly increased after water-assisting. In addition, XPS measurement showed that the ratio of Br/Pb achieved to be close to three. Namely, by passivating the surface defect using Ostwald ripening, the prepared PeQDs achieved a high PLQY of over 95%
Novel Reassortant Influenza A(H1N2) Virus Derived from A(H1N1)pdm09 Virus Isolated from Swine, Japan, 2012
We isolated a novel influenza virus A(H1N2) strain from a pig on January 13, 2012, in Gunma Prefecture, Japan. Phylogenetic analysis showed that the strain was a novel type of double-reassortant virus derived from the swine influenza virus strains H1N1pdm09 and H1N2, which were prevalent in Gunma at that time
Molecular Evolutionary Analyses of the Pseudomonas-Derived Cephalosporinase Gene
Despite the increasing evidence of the clinical impact of Pseudomonas-derived cephalosporinase (PDC) sequence polymorphisms, the molecular evolution of its encoding gene, blaPDC, remains elusive. To elucidate this, we performed a comprehensive evolutionary analysis of blaPDC. A Bayesian Markov Chain Monte Carlo phylogenetic tree revealed that a common ancestor of blaPDC diverged approximately 4660 years ago, leading to the formation of eight clonal variants (clusters A–H). The phylogenetic distances within clusters A to G were short, whereas those within cluster H were relatively long. Two positive selection sites and many negative selection sites were estimated. Two PDC active sites overlapped with negative selection sites. In docking simulation models based on samples selected from clusters A and H, piperacillin was bound to the serine and the threonine residues of the PDC active sites, with the same binding mode for both models. These results suggest that, in P. aeruginosa, blaPDC is highly conserved, and PDC exhibits similar antibiotic resistance functionality regardless of its genotype
