    Online Education: Transferring Personal Experiences to Professional Development

    In this paper, we discuss how one candidate’s experience as she participated an online mathematics specialist program bolstered her confidence and ability to provide online professional development for her teachers. We include personal accounts by the mathematics specialist program instructors, the mathematics specialist candidate, and an elementary school teacher to illustrate how the experience of completing online graduate courses led to the candidate providing online learning opportunities for teachers. In particular, we highlight the importance of building relationships and using high-quality mathematical tasks in both the online preparation program and the online professional development. This case study provides evidence that exposure to online learning environments as a learner can help lower the barrier of entry for planning and providing online learning experiences as a teacher

    Zika virus preferentially replicates in the female reproductive tract after vaginal inoculation of rhesus macaques.

    Zika virus (ZIKV) is a mosquito-transmitted virus that can cause severe defects in an infected fetus. ZIKV is also transmitted by sexual contact, although the relative importance of sexual transmission is unclear. To better understand the role of sexual transmission in ZIKV pathogenesis, a nonhuman primate (NHP) model of vaginal transmission was developed. ZIKV was readily transmitted to mature cycling female rhesus macaque (RM) by vaginal inoculation with 104-106 plaque-forming units (PFU). However, there was variability in susceptibility between the individual RM with 1->8 vaginal inoculations required to establish infection. After treatment with Depoprovera, a widely used contraceptive progestin, two RM that initially resisted 8 vaginal ZIKV inoculations became infected after one ZIKV inoculation. Thus, Depoprovera seemed to enhance susceptibility to vaginal ZIKV transmission. Unexpectedly, the kinetics of virus replication and dissemination after intravaginal ZIKV inoculation were markedly different from RM infected with ZIKV by subcutaneous (SQ) virus inoculation. Several groups have reported that after SQ ZIKV inoculation vRNA is rapidly detected in blood plasma with vRNA less common in urine and saliva and only rarely detected in female reproductive tract (FRT) secretions. In contrast, in vaginally inoculated RM, plasma vRNA is delayed for several days and ZIKV replication in, and vRNA shedding from, the FRT was found in all 6 animals. Further, after intravaginal transmission ZIKV RNA shedding from FRT secretions was detected before or simultaneously with plasma vRNA, and persisted for at least as long. Thus, ZIKV replication in the FRT was independent of, and often preceded virus replication in the tissues contributing to plasma vRNA. These results support the conclusion that ZIKV preferentially replicates in the FRT after vaginal transmission, but not after SQ transmission, and raise the possibility that there is enhanced fetal infection and pathology after vaginal ZIKV transmission compared to a mosquito transmitted ZIKV

    Results from the Palo Verde neutrino oscillation experiment

    The ν̅e flux and spectrum have been measured at a distance of about 800 m from the reactors of the Palo Verde Nuclear Generating Station using a segmented Gd-loaded liquid scintillator detector. Correlated positron-neutron events from the reaction ν̅ep→e+n were recorded for a period of 200 d including 55 d with one of the three reactors off for refueling. Backgrounds were accounted for by making use of the reactor-on and reactor-off cycles, and also with a novel technique based on the difference between signal and background under reversal of the e+ and n portions of the events. A detailed description of the detector calibration, background subtraction, and data analysis is presented here. Results from the experiment show no evidence for neutrino oscillations. ν̅e→ν̅x oscillations were excluded at 90% C.L. for Δm2>1.12×10-3 eV2 for full mixing and sin22θ>0.21 for large Δm2. These results support the conclusion that the observed atmospheric neutrino oscillations do not involve νe

    MicroRNA profiling reveals marker of motor neuron disease in ALS models

    Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder marked by the loss of motor neurons (MNs) in the brain and spinal cord, leading to fatally debilitating weakness. Because this disease predominantly affects MNs, we aimed to characterize the distinct expression profile of that cell type to elucidate underlying disease mechanisms and to identify novel targets that inform on MN health during ALS disease time course. microRNAs (miRNAs) are short, noncoding RNAs that can shape the expression profile of a cell and thus often exhibit cell-type-enriched expression. To determine MN-enriched miRNA expression, we used Cre recombinase-dependent miRNA tagging and affinity purification in mice. By defining thein vivomiRNA expression of MNs, all neurons, astrocytes, and microglia, we then focused on MN-enriched miRNAs via a comparative analysis and found that they may functionally distinguish MNs postnatally from other spinal neurons. Characterizing the levels of the MN-enriched miRNAs in CSF harvested from ALS models of MN disease demonstrated that one miRNA (miR-218) tracked with MN loss and was responsive to an ALS therapy in rodent models. Therefore, we have used cellular expression profiling tools to define the distinct miRNA expression of MNs, which is likely to enrich future studies of MN disease. This approach enabled the development of a novel, drug-responsive marker of MN disease in ALS rodents.SIGNIFICANCE STATEMENTAmyotrophic lateral sclerosis (ALS) is a neurodegenerative disease in which motor neurons (MNs) in the brain and spinal cord are selectively lost. To develop tools to aid in our understanding of the distinct expression profiles of MNs and, ultimately, to monitor MN disease progression, we identified small regulatory microRNAs (miRNAs) that were highly enriched or exclusive in MNs. The signal for one of these MN-enriched miRNAs is detectable in spinal tap biofluid from an ALS rat model, where its levels change as disease progresses, suggesting that it may be a clinically useful marker of disease status. Furthermore, rats treated with ALS therapy have restored expression of this MN RNA marker, making it an MN-specific and drug-responsive marker for ALS rodents.</jats:p

    Progress in cavity QED with single trapped atoms

    We report on recent progress in our lab involving cavity quantum electrodynamics with optically trapped atoms. In particular, we will focus on a recent measurement of the Vacuum-Rabi splitting for one atom strongly coupled to the field of a high finesse optical resonator. This splitting is characteristic of the normal modes in the eigenvalue spectrum of the atom-cavity system. A new Raman scheme for cooling atomic motion along the cavity axis enables a complete spectrum to be recorded for an individual atom trapped within the cavity mode, in contrast to all previous measurements in cavity QED that have required averaging over many atoms

    Dispersed Crude Oil Induces Dysbiosis In the Red Snapper \u3ci\u3eLutjanus campechanus\u3c/i\u3e External Microbiota

    The fish external microbiota competitively excludes primary pathogens and prevents the proliferation of opportunists. A shift from healthy microbiota composition, known as dysbiosis, may be triggered by environmental stressors and increases host susceptibility to disease. The Deepwater Horizon (DWH) oil spill was a significant stressor event in the Gulf of Mexico. Despite anecdotal reports of skin lesions on fishes following the oil spill, little information is available on the impact of dispersed oil on the fish external microbiota. In this study, juvenile red snapper (Lutjanus campechanus) were exposed to a chemically enhanced water-accommodated fraction (CEWAF) of Corexit 9500/DWH oil (CEWAF) and/or the bacterial pathogen Vibrio anguillarum in treatments designed to detect changes in and recovery of the external microbiota. In fish chronically exposed to CEWAF, immunoglobulin M (IgM) expression significantly decreased between 2 and 4 weeks of exposure, coinciding with elevated liver total polycyclic aromatic hydrocarbons (PAHs). Dysbiosis was detected on fish chronically exposed to CEWAF compared to seawater controls, and addition of a pathogen challenge altered the final microbiota composition. Dysbiosis was prevented by returning fish to clean seawater for 21 days after 1 week of CEWAF exposure. Four fish exhibited lesions during the trial, all of which were exposed to CEWAF but not all of which were exposed to V. anguillarum. This study indicates that month-long exposure to dispersed oil leads to dysbiosis in the external microbiota. As the microbiota is vital to host health, these effects should be considered when determining the total impacts of pollutants in aquatic ecosystems
