    Assessment of vegetation condition and health at Claudelands Bush (Jubilee Bush; Te Papanui)

    As the largest remnant indigenous natural area, Claudelands Bush is a key asset for Hamilton city. With a history including clearance, logging and grazing; high numbers of plant species have been lost from the bush. Some of these pressures still exist today such as drainage, invasion by adventive plant species, presence of animal pests and the small size of the bush fragment. These pressures continue to contribute to native species losses. To reduce species loss and improve vegetation condition and biodiversity, management has been taking place in the area since the 1980’s. Management included planting of native species, weeding of the bush, construction of wind breaks and boardwalks

    Key ecological sites of Hamilton City: Volume 1

    Ecological sites of significance previously identified in 2000 were reviewed in 2011. Natural vegetation in areas acquired by the city since 2000 was also surveyed to identify any new key sites. In total seventy key sites that met the Waikato Regional Council Regional Policy Statement criteria for ecological significance were identified across Hamilton City. Of the original key sites, the total area covered by sites, average site size and overall quality of sites had increased between the 2000 and 2011 surveys. This was due to restoration efforts across the city by Hamilton City Council and the community. Vegetation restoration efforts have had other biodiversity and ecological benefits such as providing additional habitat for the city’s increasing tui population. Key sites are not spread evenly across the city or across landform types. Most key sites are either in gullies or adjoining the Waikato River. Less than 1% of urban alluvial plains and peat bogs are key sites. Two sites on private land have degraded and no longer meet the ecological significance criteria in 2011. The current survey utilised a standard methodology focused on vegetation types. There will be other significant sites not identified including sites with significant fauna values but a detailed and costly survey would be required to identify all such sites. The 1.5% of the city area covered by key sites is well below the 10% minimum recommended to prevent biodiversity decline in urban areas. Areas where vegetation restoration has begun in the city have the potential to expand existing key sites or develop new sites if council and community efforts continue in the future. The Council and its restoration partners should continue to seek ways of increasing native vegetation cover in Hamilton City and restoration of the distinctive gully landform remains the best option

    Vegetation restoration plan, New Plymouth Fitzroy to Bell Block coastal walkway extension

    In 1999, the New Plymouth District Council began construction of its award winning coastal walkway. Along with providing an area for recreation, this new walk and cycle path serves as an alternate route for commuting along the city away from arterial roads. The New Plymouth District Council is in the process of extending this walkway a further three kilometres from Fitzroy Motor Camp to Ellesmere Avenue, Bell Block. This will encompass Peringa Park, Hickford Park and the Mangati Walkway, with completion expected by mid 2010. As part of this $4.2 million project, the District Council aims to restore the surrounding native duneland vegetation. The Centre for Biodiversity and Ecology Research (University of Waikato) was contracted by the New Plymouth District Council to provide a vegetation restoration plan for the Fitzroy to Bell Block section of the coastal walkway. This report considers the current vegetation of this three km section of the walkway, based on a rapid qualitative assessment undertaken in June, 2010. The target ecosytems Spinifex sandfield, flax-taupata shrubland and coastal forest vegetation types once dominant in the area are described in detail. Restoration recommendations are included to assist in the recreation of these ecosystems, including planting zones, weed control strategies and ongoing monitoring objectives

    Efficient Gene Targeting Mediated by Adeno-Associated Virus and DNA Double-Strand Breaks

    Gene targeting is the in situ manipulation of the sequence of an endogenous gene by the introduction of homologous exogenous DNA. Presently, the rate of gene targeting is too low for it to be broadly used in mammalian somatic cell genetics or to cure genetic diseases. Recently, it has been demonstrated that infection with recombinant adeno-associated virus (rAAV) vectors can mediate gene targeting in somatic cells, but the mechanism is unclear. This paper explores the balance between random integration and gene targeting with rAAV. Both random integration and spontaneous gene targeting are dependent on the multiplicity of infection (MOI) of rAAV. It has previously been shown that the introduction of a DNA double-stranded break (DSB) in a target gene can stimulate gene targeting by several-thousand-fold in somatic cells. Creation of a DSB stimulates the frequency of rAAV-mediated gene targeting by over 100-fold, suggesting that the mechanism of rAAV-mediated gene targeting involves, at least in part, the repair of DSBs by homologous recombination. Absolute gene targeting frequencies reach 0.8% with a dual vector system in which one rAAV vector provides a gene targeting substrate and a second vector expresses the nuclease that creates a DSB in the target gene. The frequencies of gene targeting that we achieved with relatively low MOIs suggest that combining rAAV vectors with DSBs is a promising strategy to broaden the application of gene targeting

    Relationship of blood pressure, antihypertensive therapy, and outcome in ischemic stroke treated with intravenous thrombolysis: retrospective analysis from Safe Implementation of Thrombolysis in Stroke-International Stroke Thrombolysis Register (SITS-ISTR)

    <p><b>Background and Purpose:</b> The optimal management of blood pressure (BP) in acute stroke remains unclear. For ischemic stroke treated with intravenous thrombolysis, current guidelines suggest pharmacological intervention if systolic BP exceeds 180 mm Hg. We determined retrospectively the association of BP and antihypertensive therapy with clinical outcomes after stroke thrombolysis.</p> <p><b>Methods:</b> The SITS thrombolysis register prospectively recorded 11 080 treatments from 2002 to 2006. BP values were recorded at baseline, 2 hours, and 24 hours after thrombolysis. Outcomes were symptomatic (National Institutes of Health Stroke Scale score deterioration ≥4) intracerebral hemorrhage Type 2, mortality, and independence at (modified Rankin Score 0 to 2) 3 months. Patients were categorized by history of hypertension and antihypertensive therapy within 7 days after thrombolysis: Group 1, hypertensive treated with antihypertensives (n=5612); Group 2, hypertensive withholding antihypertensives (n=1573); Group 3, without history of hypertension treated with antihypertensives (n=995); and Group 4, without history of hypertension not treated with antihypertensives (n=2632). For 268 (2.4%) patients, these data were missing. Average systolic BP 2 to 24 hours after thrombolysis was categorized by 10-mm Hg intervals with 100 to 140 used as a reference.</p> <p><b>Results:</b> In multivariable analysis, high systolic BP 2 to 24 hours after thrombolysis as a continuous variable was associated with worse outcome (P<0.001) and as a categorical variable had a linear association with symptomatic hemorrhage and a U-shaped association with mortality and independence with systolic BP 141 to 150 mm Hg associated with most favorable outcomes. OR (95% CI) from multivariable analysis showed no difference in symptomatic hemorrhage (1.09 [0.83 to 1.51]; P=0.58) and independence (1.03 [0.93 to 1.10]; P=0.80) but lower mortality (0.82 [0.73 to 0.92]; P=0.0007) for Group 1 compared with Group 4. Group 2 had a higher symptomatic hemorrhage (1.86 [1.34 to 2.68]; P=0.0004) and mortality (1.62 [1.41 to 1.85]; P<0.0001) and lower independence (0.89 [0.80 to 0.99]; P=0.04) compared with Group 4. Group 3 had similar results as Group 1.</p> <p><b>Conclusions:</b> There is a strong association of high systolic BP after thrombolysis with poor outcome. Withholding antihypertensive therapy up to 7 days in patients with a history of hypertension was associated with worse outcome, whereas initiation of antihypertensive therapy in newly recognized moderate hypertension was associated with a favorable outcome.</p&gt

    Associations between selected demographic, biological, school environmental and physical education based correlates, and adolescent physical activity

    The study investigated associations between selected physical activity correlates among 299 adolescents (90 boys, age 12-14 years) from 3 English schools. Physical activity was assessed by self-report and accelerometry. Correlates represented biological, predisposing, and demographic factors as described in the Youth Physical Activity Promotion Model. Boys engaged in more self-reported (p < .01) and accelerometer assessed physical activity than girls (p = .02). Positive associations between sex (male), BMI, Perceived PE Ability, Perceived PE Worth, number of enrolled students, and physical activity outcomes were evident (p < .05). Schoolbased physical activity promotion should emphasize sex-specific enhancement of students' perceived PE competence and enjoyment

    An Activity Book to Teach Primary Skills to Individuals and Families Through Fun Interventions

    This thesis will focus on a new intervention approach that teaches individuals and families the skills needed to cope with life in appropriate ways. Throughout history there have been many therapeutic theories and approaches that have clinically addressed the issues facing today\u27s individuals and families. Play therapy and the family preservation model are the two theoretical approaches this thesis explores as key components in the design of a new intervention approach that meets the needs of individuals and families in their own environment. This thesis proposes play therapy techniques that can be utilized by paraprofessionals, taking the teaching approach to the family through social service programs, schools, and community organizations. The social service programs this thesis explores which will best incorporate play therapy techniques are those of the family preservation model. However, most program designs still take a clinical intervention approach and provide services only as a response to a crisis. Thus, many children and families are not receiving true prevention services. The resolution to this problem is combining the approaches of play therapy and family preservation with a program model that utilizes paraprofessionals, school, and community organizations already in place. In order to really teach the skills needed, new intervention approaches much be shared. [n the past traditional talking and l lecturing have failed to teach with most of the individuals and families being served. This thesis proposes an activity book that addresses feeling, communication, and problem solving skills in fun and interactive ways. This thesis further focuses on the theory that approaching individuals and families in this manner better teaches the primary skills needed, as well as brings the family closer. A team of five family preservation team members reviewed the activity book and conducted several interventions with individuals and families, utilizing the activity deemed appropriate. Four of the team members completed a thirty-question survey rating their opinions of the activity book, as well as discussed opinions further in a group interview. Results of the survey and interview produced significant evidence that the activity book is targeted for an appropriate audience, as well as provided new and effective approaches for working with individuals and families struggling to learn the basic feeling, communication, and problem solving skills needed to cope with life issues

    Some topics in the kinetics of protein aggregation

    Preliminary results are presented for the kinetics of phase separation in three distinct models of protein aggregation. The first is a model of the formation of spherical microcrystals of insulin via an initial formation of fractal clusters of insulin. The results of our Brownian dynamics study of this model are in qualitative agreement with a recent experimental study (Biophys. Jour. 89 (2005) 3424-3433) of microcrystal formation from aqueous mixtures of insulin. A second work involves a theory for the formation of metastable bundles of sickle hemoglobin from fibers, based on a recent generic theory of bundle formation (Phy. Rev. Lett. 99 (2007) 098101). We also discuss a model for the microscopic formation of these fibers. Finally, we discuss preliminary results for the kinetics of cluster formation for a six patch model of protein crystallization
