    Effects of Inoculating the Diazotrophic Endophyte <i>Bradyrhizobium</i> sp. AT1 on Different Cultivars of Sweet Potato (<i>Ipomoea batatas</i> [L.] Lam.)

    Owing to the worldwide shortage of nitrogen (N) fertilizers, diazotrophic endophytes have received increasing attention as biofertilizers. In this study, we investigated the inoculation effects of a diazotrophic endophyte (Bradyrhizobium sp. AT1) on three different cultivars of sweet potato (cvs. Beniazuma, Ayamurasaki, and Kokei No. 14) under pot, container, and different field conditions. Following inoculation, the root length was increased in cvs. Beniazuma and Ayamurasaki but suppressed in cv. Kokei No. 14 in pots, filled with a mixture of vermiculite, potting soil, and pearlite. AT1 inoculation also increased shoot growth in cv. Beniazuma and tuber formation in cv. Ayamurasaki in containers filled with vermiculite, potting soil, and light-colored Andosol. In field experiments, carried out at two field sites with the three cultivars, AT1 inoculation increased the growth of cvs. Beniazuma and Ayamurasaki, but it had almost no effect on cv. Kokei No. 14. In addition to growth promotion, inoculation of micropropagated sweet potato cv. Beniazuma with AT1 led to N derived from air (Ndfa) and acetylene reduction activity (ARA) five months after inoculation. Our studies indicate that AT1 inoculation can enhance the growth of sweet potato and promote N2 fixation

    Prediction of a single Gaussian shape of spectral line measured with low-dispersion spectrometer by using machine learning

    We have developed a denoising autoencoder based neural network (NN) method to determine a spectral line intensity with an uncertainty lower than the uncertainty determined by fitting the spectral line. The NN method processes the measured raw spectral line shape, providing a single Gaussian shape based on the training dataset, which consists of synthetically prepared Doppler shift and broadening free spectral lines in the present work. It is found that the uncertainty reduction level significantly depends on the training dataset. Limitations originating from the training dataset are also discussed

    Design of Heat-Resistant in-Vessel Components for Deuterium Beam-Aided Charge Exchange Recombination Spectroscopy in JT-60SA

    Heat-resistant in-vessel components, i.e., a heat sink, a front-end optics housing, and a diagnostic window have been designed in terms of heat-handling capability and thermal stress and mechanical stress by using a finite element method code. The heat sink, which is exposed to a plasma heat flux of up to 0.3 MW/m2, consists of carbon tiles, a carbon sheet, and a stainless steel heat sink with a water-cooling channel. Analysis shows that at a water flow rate of 0.9 kg/s with a water pressure of 0.5 MPa, an increase in the carbon tile temperature is mitigated below the limit related with detrimental red-hot (900°C). The front-end optics housing temperature and the diagnostic window of sapphire glass temperature are within the allowable temperature. The thermal stress and mechanical stress are less than the allowable value, respectively

    Improving reactivity of naphthalimide-based GST probe by imparting TPP cation: Development and application for live cell imaging

    Glutathione S-transferases (GSTs) are a superfamily of multifunctional enzymes comprising multiple classes and subtypes. This paper describes the synthesis and characterization of TPPBN-1, a naphthalimide derivative conjugated with a triphenylphosphonium (TPP) cation. When 4-bromonaphthalimide (BrNaph), a previously characterized GST substrate, was conjugated to a TPP cation, the conjugate showed increased reactivity towards most alpha- and mu-class GSTs, particularly the GSTA2 subtype, compared to the parent compound, but hardly towards Pi-class GSTs. Using this probe with enhanced reactivity, the enzymatic activity of endogenous GSTA1/2 in HepG2 cells was visualized by confocal fluorescence microscopy. The results demonstrated that modification with TPP cations, which are often used as tags for targeting mitochondria, can be used to enhance the reactivity of probes for specific GST subtypes

    Study of detached plasma profile in the divertor simulation experimental module of tandem mirror GAMMA 10/PDX

    A divertor simulation experimental module (D-module) in the tandem mirror GAMMA 10/PDX was used for the study of plasma detachment. In previous studies, it was difficult to measure far-upstream plasma parameters in the D-module, and only electrostatic probes on the target plate were used to perform electron temperature and density measurements. To study the detached plasma structure, a Thomson scattering (TS) system and a microwave interferometer system have been installed to measure the inside plasma parameters of the D-module, and a movable electrostatic probe has been placed at the inlet of the D-module to measure the inlet plasma density and temperature. The TS system in the central cell observed the electron temperature and density of the core plasma simultaneously. These measurements revealed the entire density and temperature structure from the core plasma to the divertor plate. The line average electron density measured by the microwave interferometer showed a rollover behavior during detachment. The results indicated that the ionization region was located around the center of the D-module, and it appears to move upstream along the axis

    Spatiotemporal Structure of Hα Emission from the Detached Plasma in GAMMA 10/PDX

    High-speed camera measurement was performed for analyses of detached plasma fluctuation in the GAMMA 10/PDX tandem mirror device. Significant spectral peaks at frequencies of 1.5 and 3 kHz were observed to have several local maxima along the vertical, near the entrance of the V-shaped target. Wavelet analysis indicated that 1.5- and 3-kHz fluctuations became non-simultaneously strong. Their phase relationships demonstrated inversion-symmetric and symmetric features with respect to the midplane. Such spectral patterns were interpreted as the line-integral of odd- and even-mode structures. It is suspected that the vertical edge part was due to radially elongated structures with an azimuthal phase shift
