    Variable effect of co-infection on the HIV infectivity: Within-host dynamics and epidemiological significance

    <p>Abstract</p> <p>Background</p> <p>Recent studies have implicated viral characteristics in accounting for the variation in the HIV set-point viral load (spVL) observed among individuals. These studies have suggested that the spVL might be a heritable factor. The spVL, however, is not in an absolute equilibrium state; it is frequently perturbed by immune activations generated by co-infections, resulting in a significant amplification of the HIV viral load (VL). Here, we postulated that if the HIV replication capacity were an important determinant of the spVL, it would also determine the effect of co-infection on the VL. Then, we hypothesized that viral factors contribute to the variation of the effect of co-infection and introduce variation among individuals.</p> <p>Methods</p> <p>We developed a within-host deterministic differential equation model to describe the dynamics of HIV and malaria infections, and evaluated the effect of variations in the viral replicative capacity on the VL burden generated by co-infection. These variations were then evaluated at population level by implementing a between-host model in which the relationship between VL and the probability of HIV transmission per sexual contact was used as the within-host and between-host interface.</p> <p>Results</p> <p>Our within-host results indicated that the combination of parameters generating low spVL were unable to produce a substantial increase in the VL in response to co-infection. Conversely, larger spVL were associated with substantially larger increments in the VL. In accordance, the between-host model indicated that co-infection had a negligible impact in populations where the virus had low replicative capacity, reflected in low spVL. Similarly, the impact of co-infection increased as the spVL of the population increased.</p> <p>Conclusion</p> <p>Our results indicated that variations in the viral replicative capacity would influence the effect of co-infection on the VL. Therefore, viral factors could play an important role driving several virus-related processes such as the increment of the VL induced by co-infections. These results raise the possibility that biological differences could alter the effect of co-infection and underscore the importance of identifying these factors for the implementation of control interventions focused on co-infection.</p

    An Individual-Oriented Model on the Emergence of Support in Fights, Its Reciprocation and Exchange

    Complex social behaviour of primates has usually been attributed to the operation of complex cognition. Recently, models have shown that constraints imposed by the socio-spatial structuring of individuals in a group may result in an unexpectedly high number of patterns of complex social behaviour, resembling the dominance styles of egalitarian and despotic species of macaques and the differences between them. This includes affiliative patterns, such as reciprocation of grooming, grooming up the hierarchy, and reconciliation. In the present study, we show that the distribution of support in fights, which is the social behaviour that is potentially most sophisticated in terms of cognitive processes, may emerge in the same way. The model represents the spatial grouping of individuals and their social behaviour, such as their avoidance of risks during attacks, the self-reinforcing effects of winning and losing their fights, their tendency to join in fights of others that are close by (social facilitation), their tendency to groom when they are anxious, the reduction of their anxiety by grooming, and the increase of anxiety when involved in aggression. Further, we represent the difference in intensity of aggression apparent in egalitarian and despotic macaques. The model reproduces many aspects of support in fights, such as its different types, namely, conservative, bridging and revolutionary, patterns of choice of coalition partners attributed to triadic awareness, those of reciprocation of support and ‘spiteful acts’ and of exchange between support and grooming. This work is important because it suggests that behaviour that seems to result from sophisticated cognition may be a side-effect of spatial structure and dominance interactions and it shows that partial correlations fail to completely omit these effects of spatial structure. Further, the model is falsifiable, since it results in many patterns that can easily be tested in real primates by means of existing data

    Mode coupling effects in ring-core fibres for space-division multiplexing systems

    An optical fibre with weak mode coupling is desirable for future ultra-high capacity space-division multiplexing (SDM) systems because mode coupling in an optical fibre results in extrinsic loss of the fibre and crosstalk between guided optical modes. To study the feasibility of a ring-core fibre (RCF) for SDM systems, in this paper, we investigate the mode coupling in the RCF supporting 5 or 7 guided mode groups (MGs) at a wavelength of 1550nm. For this purpose, the coupled mode/power theory (CMT/CPT) with identified spatial power spectrum of random perturbations of fibre axis is used to estimate the bend loss/crosstalk of the RCF due to microbending. It is shown that, based on the identified parameters for the spatial power spectrum in the 5/7-MG RCF, the estimated bend loss/crosstalk of the RCF agrees well with experimental results. In addition, the impact of the gradient parameter α and refractive index contrast Δ of the fibre refractive index profile on bend loss and crosstalk of the RCF is explored. Simulation results indicate that the Δ instead of the α significantly affects bend loss and crosstalk of the RCF. The magnitude improvement in bend loss by increasing the Δ is dependent on the spatial power spectrum
