    NEOADJUVANT DIAGNOSIS AND MANAGEMENT OF CHRONIC ENDOMETRITIS

    Aim. The purpose of this research was informational content assessment of the anamnestic data in diagnosing of chronic endometritis before its histologic confirmation and possibility of the timely beginning of complex therapy of pathology.Materials and methods. To achieve the aim of the retrospective analysis of 303 case reports of patients who underwent endometrium papel biopsy as a part of examination plan before assisted reproductive treatment in 2017. A particular interest, in our case, submitted the special anamnesis including consideration of menstrual, sexual and reproductive functions, past gynecologic diseases and interventions.Results. In the analysis of the received results the most constant signs of chronic endometritis: disorders of menstrual and seсretory functions; the high index of intrauterine interventions, considerable frequency of identification of endometrium polyps and the cervical channel (40,1%), pathological cervical neoplasia (30,8%) were noted.Conclusion. Thus, anamnestic data of patients with the verified endometritis, especially extended one, with inclusion of nonconventional, but relevant at the present stage additional signs, have shown high informational content for neoadjuvant diagnosis of pathology of endometrium of inflammatory character. Only against the background of timely initiated complex therapy, numerous endometrial receptivity parameters are restored, which leads to relief of clinical symptoms and lays the foundation for the successful implementation of reproductive function

    HIV Replication Enhances Production of Free Fatty Acids, Low Density Lipoproteins and Many Key Proteins Involved in Lipid Metabolism: A Proteomics Study

    BACKGROUND: HIV-infected patients develop multiple metabolic abnormalities including insulin resistance, lipodystrophy and dyslipidemia. Although progression of these disorders has been associated with the use of various protease inhibitors and other antiretroviral drugs, HIV-infected individuals who have not received these treatments also develop lipid abnormalities albeit to a lesser extent. How HIV alters lipid metabolism in an infected cell and what molecular changes are affected through protein interaction pathways are not well-understood. RESULTS: Since many genetic, epigenetic, dietary and other factors influence lipid metabolism in vivo, we have chosen to study genome-wide changes in the proteomes of a human T-cell line before and after HIV infection in order to circumvent computational problems associated with multiple variables. Four separate experiments were conducted including one that compared 14 different time points over a period of >3 months. By subtractive analyses of protein profiles overtime, several hundred differentially expressed proteins were identified in HIV-infected cells by mass spectrometry and each protein was scrutinized for its biological functions by using various bioinformatics programs. Herein, we report 18 HIV-modulated proteins and their interaction pathways that enhance fatty acid synthesis, increase low density lipoproteins (triglycerides), dysregulate lipid transport, oxidize lipids, and alter cellular lipid metabolism. CONCLUSIONS: We conclude that HIV replication alone (i.e. without any influence of antiviral drugs, or other human genetic factors), can induce novel cellular enzymes and proteins that are significantly associated with biologically relevant processes involved in lipid synthesis, transport and metabolism (p = <0.0002-0.01). Translational and clinical studies on the newly discovered proteins may now shed light on how some of these proteins may be useful for early diagnosis of individuals who might be at high risk for developing lipid-related disorders. The target proteins could then be used for future studies in the development of inhibitors for preventing lipid-metabolic anomalies. This is the first direct evidence that HIV-modulates production of proteins that are significantly involved in disrupting the normal lipid-metabolic pathways

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals &lt;1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    Reproduction and use of labor resources in agricultural region

    Воспроизводство и рациональное использование сельских трудовых ресурсов напрямую влияют на экономический рост в регионе, поэтому объективный анализ и оценка состояния трудовых ресурсов, а также их изменений в муниципальных районах региона и в агропромышленном комплексе (АПК) как гаранте продовольственной безопасности является актуальной задачей. Предлагаемая методика исследования направлена на системный анализ происходящих в этой сфере процессов как на уровне региона, так и в разрезе муниципальных районов и организаций АПК. Проводится исследование трудовых ресурсов в динамике, в том числе их распределение по организационно-правовым формам, по уровню профессионального образования как одному из основных факторов, обеспечивающих развитие человеческого потенциала территории, а также в отраслевом разрезе (сельское хозяйство, переработка сельскохозяйственной продукции, обслуживание сельского хозяйства, сельское строительство) и др. На основе этой методики выполнено исследование воспроизводства и использования трудовых ресурсов АПК Оренбургской области, имеющей свыше 40 % сельского населения. Выявлено, что структура организационно-правовых форм в 2008-2013 гг. в АПК области существенно изменилась. Общее число организаций увеличилось на треть в основном за счет роста количества обществ с ограниченной ответственностью, доля сельхозпредприятий снизилась, выросли уровень профессионального образования и численность работающих с высшим и средним профессиональным образованием в сельском хозяйстве. Заработная плата в этой ведущей для районов отрасли по-прежнему существенно ниже, чем в других отраслях региональной экономики. В организациях АПК районов наблюдается процесс резкого сокращения трудовых ресурсов, который, несомненно, приводит к их разрушению. Происходящее особенно неблагоприятно в условиях принятого в стране курса на развитие агропромышленного комплекса в целом и активного импортозамещения в этой сфере.The reproduction and rational use of rural labor resources directly influences the economic growth in the region, therefore, objective analysis and assessment of human resources, as well as of the changes in the municipal areas of the region and in the agricultural industry as a guarantor of food security is an urgent task. The proposed method of study aims the perform comprehensive analysis of developments in this field as processes at the regional level, and across municipalities and agribusiness organizations. A survey of labor resources studies the dynamics of the distribution of organizational and legal forms, the level of vocational training as one of the main factors ensuring the development of the human potential of the territory, and looks at these aspects within the context of related industries (agriculture, processing of agricultural products, service agriculture, rural construction etc.) Based on the methodology a study of the reproduction and use of the agricultural labor resources of Orenburg region, which has over 40% of the rural population, was conducted. It was found that the structure of organizational and legal forms in 2008-2013 in the field of agribusiness changed significantly. The total number of companies increased by a third mainly due to the increase in the number of limited liability companies, the share of agricultural enterprises decreased, the level of vocational education improved and the number of employees with higher and secondary vocational degrees in agriculture increased. Wages in agricultural production is still substantially lower than in other sectors of the regional economy. A sharp reduction in the labor resources has been observed n organizations of the agricultural industry in rural districts, which will undoubtedly lead to their destruction. What is happening is not helping the policy of agriculture development in general and import substitution adopted in this country
