Risk management for drinking water safety in low and middle income countries: cultural influences on water safety plan (WSP) implementation in urban water utilities
We investigated cultural influences on the implementation of water safety plans (WSPs) using case studies from WSP pilots in India, Uganda and Jamaica. A comprehensive thematic analysis of semi-structured interviews (n = 150 utility customers, n = 32 WSP ‘implementers’ and n = 9 WSP ‘promoters’), field observations and related documents revealed 12 cultural themes, offered as ‘enabling’, ‘limiting’, or ‘neutral’, that influence WSP implementation in urban water utilities to varying extents. Aspects such as a ‘deliver first, safety later’ mind set; supply system knowledge management and storage practices; and non-compliance are deemed influential. Emergent themes of cultural influence (ET1 to ET12) are discussed by reference to the risk management, development studies and institutional culture literatures; by reference to their positive, negative or neutral influence on WSP implementation. The results have implications for the utility endorsement of WSPs, for the impact of organisational cultures on WSP implementation; for the scale-up of pilot studies; and they support repeated calls from practitioner communities for cultural attentiveness during WSP design. Findings on organisational cultures mirror those from utilities in higher income nations implementing WSPs – leadership, advocacy among promoters and customers (not just implementers) and purposeful knowledge management are critical to WSP success
Optical Identification of Close White Dwarf Binaries in the LISA Era
The Laser Interferometer Space Antenna (LISA) is expected to detect close
white dwarf binaries (CWDBs) through their gravitational radiation. Around 3000
binaries will be spectrally resolved at frequencies > 3 mHz, and their
positions on the sky will be determined to an accuracy ranging from a few tens
of arcminutes to a degree or more. Due to the small binary separation, the
optical light curves of >~ 30% of these CWDBs are expected to show eclipses,
giving a unique signature for identification in follow-up studies of the LISA
error boxes. While the precise optical location improves binary parameter
determination with LISA data, the optical light curve captures additional
physics of the binary, including the individual sizes of the stars in terms of
the orbital separation. To optically identify a substantial fraction of CWDBs
and thus localize them very accurately, a rapid monitoring campaign is
required, capable of imaging a square degree or more in a reasonable time, at
intervals of 10--100 seconds, to magnitudes between 20 and 25. While the
detectable fraction can be up to many tens of percent of the total resolved
LISA CWDBs, the exact fraction is uncertain due to unknowns related to the
white dwarf spatial distribution, and potentially interesting physics, such as
induced tidal heating of the WDs due to their small orbital separation.Comment: 4 pages, 2 figure

The cytochrome P450 family in the parasitic nematode <i>Haemonchus contortus</i>
<i>Haemonchus contortus</i>, a highly pathogenic and economically important parasitic nematode of sheep, is particularly adept at developing resistance to the anthelmintic drugs used in its treatment and control. The basis of anthelmintic resistance is poorly understood for many commonly used drugs with most research being focused on mechanisms involving drug targets or drug efflux. Altered or increased drug metabolism is a possible mechanism that has yet to receive much attention despite the clear role of xenobiotic metabolism in pesticide resistance in insects. The cytochrome P450s (CYPs) are a large family of drug-metabolising enzymes present in almost all living organisms, but for many years thought to be absent from parasitic nematodes. In this paper, we describe the CYP sequences encoded in the <i>H. Contortus</i> genome and compare their expression in different parasite life-stages, sexes and tissues. We developed a novel real-time PCR approach based on partially assembled CYP sequences “tags” and confirmed findings in the subsequent draft genome with RNA-seq. Constitutive expression was highest in larval stages for the majority of CYPs, although higher expression was detected in the adult male or female for a small subset of genes. Many CYPs were expressed in the worm intestine. A number of <i>H. Contortus</i> genes share high identity with <i>Caenorhabditis elegans</i> CYPs and the similarity in their expression profiles supports their classification as putative orthologues. Notably, <i>H. Contortus</i> appears to lack the dramatic CYP subfamily expansions seen in <i>C. elegans</i> and other species, which are typical of CYPs with exogenous roles. However, a small group of <i>H. Contortus</i> genes cluster with the <i>C. elegans</i> CYP34 and CYP35 subfamilies and may represent candidate xenobiotic metabolising genes in the parasite

Basal rot of narcissus : understanding pathogenicity in fusarium oxysporum f. sp. narcissi
Fusarium oxysporum is a globally distributed soilborne fungal pathogen causing root rots, bulb rots, crown rots and vascular wilts on a range of horticultural plants. Pathogenic F. oxysporum isolates are highly host specific and are classified as formae speciales. Narcissus is an important ornamental crop and both the quality and yield of flowers and bulbs can be severely affected by a basal rot caused by F. oxysporum f. sp. narcissi (FON); 154 Fusarium isolates were obtained from different locations and Narcissus cultivars in the United Kingdom, representing a valuable resource. A subset of 30 F. oxysporum isolates were all found to be pathogenic and were therefore identified as FON. Molecular characterisation of isolates through sequencing of three housekeeping genes, suggested a monophyletic origin with little divergence. PCR detection of 14 Secreted in Xylem (SIX) genes, previously shown to be associated with pathogenicity in other F. oxysporum f. spp., revealed different complements of SIX7, SIX9, SIX10, SIX12 and SIX13 within FON isolates which may suggest a race structure. SIX gene sequences were unique to FON and SIX10 was present in all isolates, allowing for molecular identification of FON for the first time. The genome of a highly pathogenic isolate was sequenced and lineage specific (LS) regions identified which harboured putative effectors including the SIX genes. Real-time RT-PCR, showed that SIX genes and selected putative effectors were expressed in planta with many significantly upregulated during infection. This is the first study to characterise molecular variation in FON and provide an analysis of the FON genome. Identification of expressed genes potentially associated with virulence provides the basis for future functional studies and new targets for molecular diagnostics
The origins of intensive marine fishing in medieval Europe: the English evidence
The catastrophic impact of fishing pressure on species such as cod and herring is well documented. However, the antiquity of their intensive exploitation has not been established. Systematic catch statistics are only available for ca. 100 years, but large-scale fishing industries existed in medieval Europe and the expansion of cod fishing from the fourteenth century (first in Iceland, then in Newfoundland) played an important role in the European colonization of the Northwest Atlantic. History has demonstrated the scale of these late medieval and post-medieval fisheries, but only archaeology can illuminate earlier practices. Zooarchaeological evidence shows that the clearest changes in marine fishing in England between AD 600 and 1600 occurred rapidly around AD 1000 and involved large increases in catches of herring and cod. Surprisingly, this revolution predated the documented post-medieval expansion of England's sea fisheries and coincided with the Medieval Warm Period-when natural herring and cod productivity was probably low in the North Sea. This counterintuitive discovery can be explained by the concurrent rise of urbanism and human impacts on freshwater ecosystems. The search for 'pristine' baselines regarding marine ecosystems will thus need to employ medieval palaeoecological proxies in addition to recent fisheries data and early modern historical records
Manipulations to the Timing and Type of Instructions to Examine Motor Skill Performance Under Pressure
There is evidence that prescriptive versus discovery methods of learning can lead to breakdowns under pressure due to “reinvestment” of knowledge and a more conscious, controlled mode of control. There is some speculation that this breakdown is mediated by the attentional focus of the instructions. We expected these effects to also be moderated by when in practice these instructions are given. Across two experiments, five groups practiced a forehand disk throwing task and we manipulated the timing and attentional focus of instructions. Internally directed instructions provided to participants early in practice resulted in a slower rate of acquisition (outcome error) and detrimental effects under stress, in comparison to the same instructions provided later in practice or not at all. Externally directed, technical instructions positively impacted rate of acquisition and regardless of when in practice they were provided, there were no adverse effects associated with instructions under pressure. These results show that the direction of attention encouraged by instructions moderates performance under stress as does the timing of presentation of these instructions
The prevalences of Salmonella Genomic Island 1 variants in human and animal Salmonella Typhimurium DT104 are distinguishable using a Bayesian approach
Throughout the 1990s, there was an epidemic of multidrug resistant Salmonella Typhimurium DT104 in both animals and humans in Scotland. The use of antimicrobials in agriculture is often cited as a major source of antimicrobial resistance in pathogenic bacteria of humans, suggesting that DT104 in animals and humans should demonstrate similar prevalences of resistance determinants. Until very recently, only the application of molecular methods would allow such a comparison and our understanding has been hindered by the fact that surveillance data are primarily phenotypic in nature. Here, using large scale surveillance datasets and a novel Bayesian approach, we infer and compare the prevalence of Salmonella Genomic Island 1 (SGI1), SGI1 variants, and resistance determinants independent of SGI1 in animal and human DT104 isolates from such phenotypic data. We demonstrate differences in the prevalences of SGI1, SGI1-B, SGI1-C, absence of SGI1, and tetracycline resistance determinants independent of SGI1 between these human and animal populations, a finding that challenges established tenets that DT104 in domestic animals and humans are from the same well-mixed microbial population
Quantifying non-star formation associated 8um dust emission in NGC 628
Combining Ha and IRAC images of the nearby spiral galaxy NGC 628, we find
that between 30-43% of its 8um dust emission is not related to recent star
formation. Contributions from dust heated by young stars are separated by
identifying HII regions in the Ha map and using these areas as a mask to
determine the 8um dust emission that must be due to heating by older stars.
Corrections are made for sub-detection-threshold HII regions, photons escaping
from HII regions and for young stars not directly associated to HII regions
(i.e. 10-100 Myr old stars). A simple model confirms this amount of 8um
emission can be expected given dust and PAH absorption cross-sections, a
realistic star-formation history, and the observed optical extinction values. A
Fourier power spectrum analysis indicates that the 8um dust emission is more
diffuse than the Ha emission (and similar to observed HI), supporting our
analysis that much of the 8um-emitting dust is heated by older stars. The 8um
dust-to-Ha emission ratio declines with galactocentric radius both within and
outside of HII regions, probably due to a radial increase in disk transparency.
In the course of this work, we have also found that intrinsic diffuse Ha
fractions may be lower than previously thought in galaxies, if the differential
