    Six adult lorises (Loris tardigradus lydekkerianus) in anoestrus were selected for study. Four of the animals were pretreated with an injection of 0·5 mg oestradiol-17β, two were given FSH (2 × 50 i.u. with a 12-hr interval), while the remaining two were not treated with hormones. All of the animals were subsequently given a subcutaneous injection of 0·5 mCi [3H]thymidine and their ovaries were recovered for autoradiography 24 hr later.It was found that the non-follicular germinal cells within 'nests' in the ovarian cortex actively incorporated the radioisotope. Some of the labelled germinal cells appeared to reach the pachytene-diplotene stages of meiotic prophase, although this would not be expected to occur in such a short time after treatment. None of the labelled germinal cells appeared to enter upon the phase of follicular growth, although a few (especially in ovaries treated with oestrogen and FSH) seemed to be in primordial follicles consisting of an incomplete layer of flattened cells

    The present study was carried out to determine the detailed histological and cytological features of the excurrent ducts of the male reproductive system in the rhesus monkey. The excurrent ducts show a regional difference in their histological features. The use of some of these features as histological markers and their possible functional significance are discussed. The epithelial cells in the different components of the excurrent duct system possess cytological features which suggest their involvement in absorption and the secretion of different products into the lumen

    Ultrastructural features of the principal cell in the epididymis of the rhesus monkey

    The ultrastructural features of the principal cell in the epididymal epithelium of the monkey epididymis are suggestive of the cell carrying out a dual function of absorption and secretion. Both these functions occur on the luminal surface of the cell as well as on the lateral and basal aspects of the cell which face the intercellular spaces. Transmision Electron Microscopic studies of epididymal tissues following their impregnation with lanthanum nitrate indicated that the intercellular spaces are effectively sealed-off from the luminal space by the apically situated tight junctions between adjoining principal cells. The intercellular spaces are contiguous with the perivascular spaces of the subepithelial blood capillaries. It is suggested that the absorptive and secretory functions occuring on the apical surface of cells may be related to the creation of an appropriate intraluminal milieu for the maturation of spermatozoa while the occurrence of these functions in the intercellular spaces may represent an exchange of substances between the principal cells and the subepithelial capillaries

    Ultrastructural studies on the epididymal spermatozoa in the rhesus monkey

    Ultrastructural studies on the spermatozoa in different regions of the epididymis of the rhesus monkey have shown that the process of sperm maturation is associated with the caudad migration of the cytoplastmic droplet, a reduction in the volume of the cytoplasmic droplet, and an obvious wrinkling of the plasma membrane surrounding the head of the spermatozoa. These changes are completed by the time the spermatozoa reach the distal-middle segment of the epididymis. The present studies also indicate that spermatozoa are incorporated into the intraepithelial cells in the epidymis. This finding suggests that spermiophagy is a normal occurrence in the epidymis of rhesus monkey

    Reproductive endocrine effects of intranasal administration of norethisterone to adult female rhesus monkeys (Macaca mulatta)

    Intranasal administration of norethisterone at a daily dose of 9 μg between Days 5 and 14 of the menstrual cycles blocked ovulation in 10 out of 17 adult female monkeys. Serum concentrations of hormones indicated that ovulation was blocked due to a suppression of the mid-cycle, oestradiol-induced LH surge. Ovarian follicular activity in the treated menstrual cycles was not affected by norethisterone but there was a marked delay in the onset of the mid-cycle oestradiol surge in most of the treated animals. The duration of the menstrual cycle length after the oestradiol peak was significantly reduced in all the treated monkeys, indicative of a shortened luteal phase

    Reproductive exocrine and endocrine profile of female langur monkeys, Presbytis entellus

    In 5 caged langurs menstrual cycle length (for 125 cycles) varied from 18 to 45 days with a mean ± s.d. of 26·3 ± 3·6 days. Menstruation lasted for 2 days. A distinct cycle of vaginal smear cells was observed with a maximum cornification on Day 10 coinciding with the serum oestradiol peak. A monophasic pattern of vaginal temperature with a significant nadir on Day 11, 1 day after the oestradiol peak, was observed. Serum sialic acid concentrations directly reflected the oestrogenic activity and showed a significant peak on Day 10, coinciding with the oestradiol peak and was followed by a progesterone rise. Langurs menstruated throughout the year without showing any sign of summer amenorrhoea. We suggest that maximum cornification, vaginal temperature nadir and serum sialic acid peak could be used as markers for prediction and detection of the time of ovulation in langur monkeys

    Anti-inflammatory and analgesic effects of coral reef associated gastropod, Trochus tentorium from Tuticorin coastal waters, Southeastern India

    The aim of this work was to investigate extensively, the biomedical potential of the mollusc Trochus tentorium which are abundantly associated with coral reef of the Tuticorin coastal water. The 100% acetone fraction of the gastropod tested for its analgesic effect on Swiss mice model and anti-inflammatory activity on albino rat showed promising results. T. tentorium at the concentration of 25 and 50 mg/kg (p.o) showed significant decrease in the paw thickness (41.15 and 73.6%, respectively) at the 5th hour of the experiment. The 100% column-purified fraction of the T. tentorium (200 mg/kg p.o) exhibited significant (p < 0.001) inhibition of 79.22% against acetic acid induced abdominal constrictions. The dose of 25 mg/kg showed the inhibitions in the writhings of 67.86% (p<0.001) of animals when compared to the standard (diclofenac sodium), and 56.83% (50 mg/kg) inhibition was observed. These facts suggest that T. tentorium is a potential source for anti-inflammatory and analgesic compounds.Key words: Analgesic activity, anti-inflammatory activity, mollusc, southeastern India

    Circadian and circannual changes in the testicular function of adult rhesus monkeys

    The endocrine and gametogenic status of the testes were studied in 9 healthy adult rhesus monkeys of proven fertility throughout a one-year period. Testosterone levels were estimated by radioimmunoassay in blood samples collected at 4 h intervals over a 24 h period once a month. Semen samples and testicular biopsies were also examined once a month. A well-defined circandian rhythm was evident in serum levels of testosterone. The rhythmicity was less pronounced in February and September. The 24 h mean levels of serum testosterone were high between the months of August to March and low in the months of May to July. All animals did not uniformly respond to electro-ejaculation in April and May. Semen volume and total number of spermatozoa were maximal between September and March and least from April to August. Testicular biopsies indicated that all stages of spermatogenesis were evident between September and March and the spermatogenic activity was less evident between April and August. The contents of Sertoli cells showed a seasonal cyclicity; they were laden with lipid droplets during April to August when spermatogenesis was quiescent and vacuolated during September to March when spermatogenesis was active. These studies indicate that the testing of contraceptive drugs needs to be restricted to months of September to March in male rhesus monkeys otherwise, it is possible that the naturally occurring reproductive quiscence may be attributed to the effect of the drug being tested. The data accrued from the present studies also provide quantitative information on circulating levels of testosterone which could be used as a reference background while evaluating the contraceptive drug-effects in male rhesus monkeys

    Occurrence & nucleotide sequence analysis of hepatitis G virus in patients with acute viral hepatitis & fulminant hepatitis

    Background & objectives: Association of hepatitis G virus (HGV) with acute viral hepatitis (AVH) and fulminant hepatitis (FH) is not clearly understood.This study was designed to asses the occurrence of HGV infection and its relationship with other hepatotropic viruses in patients with FH and AVH and also to determine the nucleotide sequence of HGV isolates. Methods: The study included 100 patients of FH and 125 of AVH on the basis of clinical examination, liver function test and serology for hepatitis A, B, C and E virus. HGV RNA was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and direct sequencing for 4 randomly selected samples followed by phylogenetic analysis. Results: Of the 100 patients with FH, 30 were negative for hepatitis viruses A, B, C and E by serology (non A - non E) while 60 were negative in the AVH group. In the non A- non -E hepatitis group, HGV was positive in 16.66 per cent (5/30) cases of FH, 10 per cent (6/60) cases of AVH and 6 per cent (6/100) of healthy controls. The difference in HGV seropositivity between FH and AVH patients was statistically not significant compared to healthy controls, while HBV and HCV infections were significant. The four isolates sequenced seemed to be of same type and close to Chinese strain of HGV (Y13755.1 Y13756.1 Y15407, and U67782) on phylogeny. Interpretation & conclusion: In HGV infection was not found to be clinically significant as well as nonpathogenic in the patients of FH and AVH and appeared to be an innocent bystander in the course of the disease. The four sequenced HGV isolates showed close pairing with Chinese strains
