    Seed productivity of Linum usitatissimum L. in different ecological conditions of Uzbekistan

    The article reports influence of different environmental conditions on the productivity of Linum usitatissimum L. (flax or flaxseed) in Uzbekistan. Analyses of the plant productivity were carried out on the rainfed lands and in the conditions of the Tashkent Botanical Garden. Phenological studies revealed differences in various phases of the growing period. Morphometric parameters that determine yield of L. usitatissimum seeds under different conditions were determined. The seed yield per plant on average in the Botanical Garden was 79.4 ± 4.00, and at various regions in the rich lands such as Malguzar 76,8±4,33, Pishagar 74,2±4,54 and Khatyrchi district was 76,5±4,28. The results of the studies can be used by farms in cultivating L. usitatissimum in the Republic of Uzbekistan

    Aberrant chromatin landscape following loss of the H3.3 chaperone Daxx in haematopoietic precursors leads to Pu.1-mediated neutrophilia and inflammation

    Defective silencing of retrotransposable elements has been linked to inflammageing, cancer and autoimmune diseases. However, the underlying mechanisms are only partially understood. Here we implicate the histone H3.3 chaperone Daxx, a retrotransposable element repressor inactivated in myeloid leukaemia and other neoplasms, in protection from inflammatory disease. Loss of Daxx alters the chromatin landscape, H3.3 distribution and histone marks of haematopoietic progenitors, leading to engagement of a Pu.1-dependent transcriptional programme for myelopoiesis at the expense of B-cell differentiation. This causes neutrophilia and inflammation, predisposing mice to develop an autoinflammatory skin disease. While these molecular and phenotypic perturbations are in part reverted in animals lacking both Pu.1 and Daxx, haematopoietic progenitors in these mice show unique chromatin and transcriptome alterations, suggesting an interaction between these two pathways. Overall, our findings implicate retrotransposable element silencing in haematopoiesis and suggest a cross-talk between the H3.3 loading machinery and the pioneer transcription factor Pu.1

    The endothelial glycocalyx (EG) system is an important protective regulator of vascular integrity and permeability, provides cellular interaction and serves as a component of hemostasis. Damage of EG in septic shock, cardiopulmonary bypass (CPB), trauma, ischemia and in a number of other critical states leads to capillary leakage, hemodynamic and metabolic disorders.The aim of the study was to evaluate the interaction of EG components with hemodynamic and metabolic response in patients with septic shock and in cardiosurgical interventions using CPB.Materials and methods. The study included 21 patients with septic shock and 26 patients undergoing cardiac surgery with CPB. The plasma concentrations of EG components, including heparin-sulfate proteoglycan (HSPG) and syndecan 1 (S1), were determined in the group of patients with septic shock at baseline, 2 and 24 hours after the fluid load test, and in the group of cardiosurgical patients – after induction of anesthesia, at 6 and 24 hours after the end of CPB.Results. In septic shock, the concentration of S1 in blood plasma tended to increase at 2 hours after the fluid load test. In cardiosurgical patients, at 6 hours after the end of CPB, the plasma concentration of HSPG reduced from 6.13 (4.20–9.04) to 5.08 (4.18–7.21) ng/ml (p <0.01), whereas S1 increased from 0.80 (0.56–1.13) to 1.25 (1.04–1.41) ng/ml (p <0.001). At 24 hours, HSPG and S1 returned to values close to baseline. In both groups, we established the relationship of the EG components with the parameters of pre- and afterload, as well as with the concentration of lactate.Conclusion. The damage of EG in septic shock and in cardiosurgical interventions using CPB is related with disorders of hemodynamics and metabolism.Система эндотелиального гликокаликса (ЭГ) служит важным регулятором целостности и проницаемости сосудов, обеспечивает клеточное взаимодействие и является компонентом системы гемостаза. Повреждение ЭГ при септическом шоке, искусственном кровообращении (ИК), ишемии, реперфузии, а также ряде прочих критических состояний ассоциируется с капиллярной утечкой, гемодинамическими и метаболическими нарушениями.Цель исследования: оценить взаимосвязь состояния (или повреждения) компонентов ЭГ с гемодинамическим и метаболическим ответом у пациентов с септическим шоком и при кардиохирургических вмешательствах в условиях ИК.Материалы и методы. В исследование включены 21 пациент с септическим шоком и 26 пациентов, которым проводили кардиохирургические вмешательства в условиях ИК. Концентрацию в плазме крови компонентов ЭГ, включая гепарансульфат-протеогликан (HSPG) и синдекан-1 (S1), в группе пациентов с септическим шоком определяли в начале исследования, через 2 и 24 ч после теста с инфузионной нагрузкой, а в группе кардиохирургических пациентов – после индукции анестезии, а также через 6 и 24 ч после окончания ИК.Результаты. В группе пациентов с септическим шоком через 2 ч после теста с инфузионной нагрузкой обнаружена тенденция к увеличению концентрации S1 в плазме крови. В группе кардиохирургических пациентов через 6 ч после окончания ИК отмечали снижение концентрации HSPG с 6,13 (4,20–9,04) до 5,08 (4,18–7,21) нг/мл (p <0,01) и повышение концентрации S1 в плазме крови с 0,80 (0,56–1,13) до 1,25 (1,04–1,41) нг/мл (p <0,001). Через 24 ч HSPG и S1 вернулись к значениям, близким к предоперационным. В обеих группах установлена ассоциация между концентрацией компонентов ЭГ в плазме с показателями пред- и постнагрузки, а также с концентрацией лактата.Вывод. Повреждение и сброс компонентов ЭГ при септическом шоке и ИК взаимосвязаны с нарушениями гемодинамики и метаболизма
