    Suggesting outsider status by behaving improperly: the linguistic realization of a populist rhetorical strategy in Dutch Parliament

    One of the rhetorical strategies employed by populist politicians to create and maintain an anti-elitistprofile, is the use of ‘bad manners’. By violating decency standards that politicians are supposed toobserve in political debates, populists create the image of being different from the ‘political elite’. Asyet, however, little research has been carried out focusing on the question of exactly how populistsviolate the decency standards of a specific political debating culture verbally.This chapter aims to fill this gap, by focusing on improper behaviour in Dutch parliamentary debates.We will discuss how the Dutch populist politician Geert Wilders violates four decency standards forDutch parliamentary debate to create and maintain the image of a political outsider, and show howhe systematically uses a variety of conspicuous as well as inconspicuous linguistic choices to fulfil thisrhetorical strategy.Language Use in Past and Presen

    Binnen het kader van de uitgebreide pragma-dialectiek wordt ervan uitgegaan dat taalgebruikers, wanneer zij iemand willen overtuigen, hun argumentatieve zetten zo kiezen en vormgeven dat zij strategisch manoeuvreren tussen enerzijds de wens om te voldoen aan eisen van redelijkheid en anderzijds de wens om zo effectief mogelijk te zijn. Presentatiemogelijkheden vormen een van de drie aspecten van strategisch manoeuvreren. Talige stijlmiddelen vormen een belangrijk onderdeel van deze presentatiemogelijkheden. De analyses van de wijze waarop taalgebruikers strategisch manoeuvreren met stijlmiddelen zijn echter veelal ad hoc: een systematische analyse van stijl ontbreekt vaak.Dit artikel heeft twee nauw met elkaar verbonden doelstellingen. Ten eerste willen we laten zien hoe een systematische analyse van stijlmiddelen kan worden geïntegreerd in een pragma-dialectische analyse van argumentatief taalgebruik. Daartoe maken we gebruik van methodische inzichten uit de taalkundige stilistiek. Ten tweede willen wij betogen dat de door ons voorgestelde integratie van uitgebreide pragma-dialectiek en taalkundige stilistiek voor beide benaderingen waardevol is.     Time-resolved X-Shooter spectra and RXTE light curves of the ultra-compact X-ray binary candidate 4U 0614+091

    In this paper we present X-Shooter time resolved spectroscopy and RXTE PCA light curves of the ultra-compact X-ray binary candidate 4U 0614+091. The X-Shooter data are compared to the GMOS data analyzed previously by Nelemans et al. (2004). We confirm the presence of C III and O II emission features at ~ 4650 {\AA} and ~ 5000 {\AA}. The emission lines do not show evident Doppler shifts that could be attributed to the motion of the donor star/hot spot around the center of mass of the binary. We note a weak periodic signal in the red-wing/blue-wing flux ratio of the emission feature at ~ 4650 {\AA}. The signal occurs at P = 30.23 +/- 0.03 min in the X-Shooter and at P = 30.468 +/- 0.006 min in the GMOS spectra when the source was in the low/hard state. Due to aliasing effects the period in the GMOS and X-Shooter data could well be the same. We deem it likely that the orbital period is thus close to 30 min, however, as several photometric periods have been reported for this source in the literature already, further confirmation of the 30 min period is warranted. We compare the surface area of the donor star and the disc of 4U 0614+091 with the surface area of the donor star and the disc in typical hydrogen-rich low-mass X-ray binaries and the class of AM Canum Venaticorum stars and argue that the optical emission in 4U 0614+091 is likely dominated by the disc emission. Additionally, we search for periodic signals in all the publicly available RXTE PCA light curves of 4U 0614+091 which could be associated with the orbital period of this source. A modulation at the orbital period with an amplitude of ~ 10% such as those that have been found in other ultra-compact X-ray binaries (4U 0513-40, 4U 1820-30) is not present in 4U 0614+091.Comment: Accepted for publication in MNRAS, 11 pages, 7 figure

    An integrated bioinformatics approach to improve two-color microarray quality-control: impact on biological conclusions

    Omics technology used for large-scale measurements of gene expression is rapidly evolving. This work pointed out the need of an extensive bioinformatics analyses for array quality assessment before and after gene expression clustering and pathway analysis. A study focused on the effect of red wine polyphenols on rat colon mucosa was used to test the impact of quality control and normalisation steps on the biological conclusions. The integration of data visualization, pathway analysis and clustering revealed an artifact problem that was solved with an adapted normalisation. We propose a possible point to point standard analysis procedure, based on a combination of clustering and data visualization for the analysis of microarray data

    Peribiliary glands are key in regeneration of the human biliary epithelium after severe bile duct injury

    Peribiliary glands (PBG) are a source of stem/progenitor cells organized in a cellular network encircling large bile ducts. Severe cholangiopathy with loss of luminal biliary epithelium has been proposed to activate PBG, resulting in cell proliferation and differentiation to restore biliary epithelial integrity. However, formal evidence for this concept in human livers is lacking. We, therefore, developed a novel ex vivo model using precision-cut slices of extrahepatic human bile ducts obtained from discarded donor livers, providing an intact anatomical organization of cell structures, to study spatiotemporal differentiation and migration of PBG cells after severe biliary injury. Post-ischemic bile duct slices were incubated in oxygenated culture medium for up to a week. At baseline, severe tissue injury was evident with loss of luminal epithelial lining and mural stroma necrosis. In contrast, PBG remained relatively well preserved and different reactions of PBG were noted, including PBG dilatation, cell proliferation and maturation. Proliferation of PBG cells increased after 24 h of oxygenated incubation, reaching a peak after 72 h. Proliferation of PBG cells was paralleled by a reduction in PBG apoptosis and differentiation from a primitive and pluripotent (Nanog+/Sox9+) to a mature (CFTR+/secretin receptor+) and activated phenotype (increased expression of HIF-1α, Glut-1, and VEGF-A). Migration of proliferating PBG cells in our ex vivo model was unorganized, but resulted in generation of epithelial monolayers at stromal surfaces. CONCLUSION: Human PBG contain biliary progenitor cells and are able to respond to bile duct epithelial loss with proliferation, differentiation, and maturation to restore epithelial integrity. The ex vivo spatiotemporal behaviour of human PBG cells provides evidence for a pivotal role of PBG in biliary regeneration after severe injury. This article is protected by copyright.     Secondary school teachers expect students to write comprehensible paragraphs in their texts. However, an explicit way of teaching to write a paragraph doesn’t exist. This article describes a text analytic study of compliance with language norms in paragraphs in two genres: e-mails with a request and persuasive texts. It addresses the question to what extent the paragraphs of thirteen- and seventeen-year-old students comply with the paragraph norms constructed on the basis of Dutch professional advisory books. Our findings indicate that students in Dutch secondary school have difficulty writing adequate paragraphs.     Partner independent fusion gene detection by multiplexed CRISPR-Cas9 enrichment and long read nanopore sequencing

    Fusion genes are hallmarks of various cancer types and important determinants for diagnosis, prognosis and treatment. Fusion gene partner choice and breakpoint-position promiscuity restricts diagnostic detection, even for known and recurrent configurations. Here, we develop FUDGE (FUsion Detection from Gene Enrichment) to accurately and impartially identify fusions. FUDGE couples target-selected and strand-specific CRISPR-Cas9 activity for fusion gene driver enrichment - without prior knowledge of fusion partner or breakpoint-location - to long read nanopore sequencing with the bioinformatics pipeline NanoFG. FUDGE has flexible target-loci choices and enables multiplexed enrichment for simultaneous analysis of several genes in multiple samples in one sequencing run. We observe on-average 665 fold breakpoint-site enrichment and identify nucleotide resolution fusion breakpoints within 2 days. The assay identifies cancer cell line and tumor sample fusions irrespective of partner gene or breakpoint-position.
