A Krónikus Chlamydia Fertőzés Transzkriptom Alapú Diagnózisa és Kezelése - ChlamyTrans - = Transcriptome-based Monitoring and Eradication of Chronic Chlamydial Infection - ChlamyTrans -
A ChlamyTrans project keretĂ©ben sikerĂĽlt kidolgoznunk egy Ăşj tĂpusĂş, PCR amplifikáciĂłn alapulĂł Chlamydiális mRNS amplifikáciĂłs mĂłdszert. A mĂłdszer szenzitivitását Ă©s specificitását rĂ©szletesen teszteltĂĽk QPCR mĂłdszerrel. Az amplifikáciĂłs mĂłdszert Chlamydia pneumoniae-val fertĹ‘zött egĂ©rtĂĽdĹ‘ben lezajlĂł bakteriális gĂ©nexpressziĂł monitorozására használtuk fel. Több olyan gĂ©nt-gĂ©ncsoportot azonosĂtottunk, amelyek in vivo a baktĂ©rium aktĂv vagy perzisztens növekedĂ©sĂ©ben szerepet játszhatnak. A bakteriális gĂ©nexpressziĂł kutatása mellett a Chlamydia fertĹ‘zĂ©s hatására bekövetkezett gazdasejti gĂ©nexpressziĂłt is monitoroztuk. Olyan gĂ©neket azonosĂtottunk, amelyek szerepet játszhatnak a Chlamydia fertĹ‘zĂ©sek által indukált szöveti patholĂłgia kialakĂtásában. VĂ©gezetĂĽl olyan mĂłdszerfejlesztĂ©sekben is rĂ©szt vettĂĽnk, amelyekkel a Chlamydia fajok funkcionális genetikáját lehet rĂ©szletesen tanulmányozni. | In the framework of ChlamyTrans project, we developed a novel PCR-based Chlamydial mRNA amplification method. The sensitivity and specificity of the method was extensively tested using the QPCR method. The amplification was used to monitor Chlamydial gene expression in Chlamydia pneumoniae infected mouse lung tissue. The gene expression screen identified Chlamydial genes that were involved in the active and persistent growth of C. pneumoniae in vivo. Besides Chlamydial gene expression profiling we monitored the host gene expression during Chlamydia infection. We could identify host genes that could potentially participate in Chlamydia-indued tissue pathology. Finally, we developed two methods with which we can characterize the functional genetics of Chlamydiae

CĂ©lul tűztĂĽk ki, hogy megvizsgáljuk: - a Chlamydia pneumoniae fertĹ‘zöttsĂ©g mĂ©rtĂ©kĂ©t AMD-ben (age related macular degeneration) szenvedĹ‘ Ă©s koszorúér betegeken. - a cytokinek kiválasztását pathologias human könnymintákban (1. postoperativ állapotnál (cataracta Ă©s PKP [perforálĂł keratoplastica] műtĂ©t), 2. corneális idegentest kapcsán, 3. bakteriális conjunctivitisben). Az IL-6 koncentráciĂł mellett meghatároztuk az IL-6 release-t is. - AION-ban (anterior ischaemiás opticus neuropathia) milyen arányban fordul elĹ‘ a ma egyik leggyakoribb thrombosist okozĂł kockázati tĂ©nyezĹ‘, az APC-rezisztencia Ă©s az ezt elĹ‘idĂ©zĹ‘ FV (Leiden)-mutáciĂł. Ăšj eredmĂ©nyek: • Chlamydia titer vizsgálataink alapján a kontrollcsoportban Ă©szlelt alacsonyabb szeropozitivitási arány alátámasztja a Chlamydia pneumoniae patholĂłgiás szerepĂ©t az AMD Ă©s a coronaria betegsĂ©g kialakulásában. Mindezek alapján feltĂ©telezzĂĽk, hogy a Chlamydia pneumoniae az egyĂ©b rizikĂłtĂ©nyezĹ‘k hatását fokozva, egyĂĽttesen hajlamosĂthat az AMD kialakulására. • Az IL-6 nemcsak a szemfelszĂni gyulladásokban, hanem postoperatĂv állapotokban is fontos szerepet játszik az inflammatorikus folyamatokban. Az IL-6 release korai markere a gyulladásos folyamatoknak. • AION-ban szenvedĹ‘ betegek jelentĹ‘s rĂ©szĂ©nĂ©l igazoltuk az aktivált protein-C rezisztencia Ă©s a Leiden-mutáciĂł jelenlĂ©tĂ©t. FeltĂ©telezzĂĽk, hogy ezen betegeknĂ©l a thrombophil állapot hozzájárult a vascularis opticus neuropathia kialakulásához. | Aims of the study were as follows: - to find correlation among chronic Chlamydia pneumoniae infection and age-related macular degeneration, AMD) in patients with coronary heart disease (CAD). - to determine the secretion kinetics of interleukin-6 (IL-6) in human tears collected from patients with different eye diseases (1. penetrating keratoplasty /PKP/ or cataract operation, 2. corneal foreign body and 3. acute bacterial conjunctivitis). The total IL-6 amount and release were calculated. - to examine the prevalence of APC resistance and Leiden mutation in patients with non-arteric ischaemic optic neuropathy (NAION). The resistance of coagulation factor V (FV) to activated protein C (APC), mostly due to FV Leiden mutation, is a common cause of inherited thombophilia. New results • Our result supports the assumption that chronic Chlamydia pneumoniae infection plays a role in the development of AMD, and CAD. • The release of IL-6 into the tears is enhanced not only in various anterior segment eye inflammations, but also in postoperative conditions. IL-6 seems to be a sensitive indicator of the different types of irritative eye diseases, especially in postoperative PKP condition. • APC resistance and Leiden mutation were found significantly more frequenently among patients with NAION than in control group members with similar risk factors. FV Leiden mutation may play an important role in the development of the disease
VĂrusok Ă©s molekuláris genetikai alteráciĂłk szerepe conjunctivalis, colorectalis, Ă©s anogenitalis elváltozások carcinogenezisĂ©ben = The role of viruses and molecular genetic alterations in conjunctival, colorectal and anogenital carcinogenesis
Colorectalis daganatok miatt eltávolĂtott biopszia mintákbĂłl Magyarországon elĹ‘ször nukleinsav hybridizáciĂłs prĂłbával mutattuk ki a human papillomavirusokat (HPV). 802 cervix mintábĂłl 432 esetben mutattunk ki nukleinsav hybridizáciĂłs prĂłbával HPV-t. Randomszerűen 105 mintát választottunk ki consensus PCR Ă©s "short" fragmenteket amplifikálĂł mĂłdszerrel a HPV-k kimutatására. Ă–sszesen 41 mintábĂłl mutattunk ki HPV-t. RFLP mĂłdszert alkalmaztunk a HPV tĂpus meghatározására. MindkĂ©t mĂłdszert adaptáltuk real-time PCR (RT-PCR) mĂłdszerhez. 191 tĂĽnetmentes páciens cervix mintáibĂłl consensus HPV PCR amplifikáciĂł után kiválasztottuk a pozitĂv mintákat, melyeket az Ăşn. "short" fragmenteket amplifikálĂł PCR mĂłdszerrel (az eredeti mintákat felhasználva) amplikonokat állĂtottunk elĹ‘. Linear array mĂłdszerrel meghatároztuk a HPV tĂpusokat. A HPV-k kimutatására Ă©s egyidejűleg törtĂ©nĹ‘ vĂrustĂpus kimutatására microchip mĂłdszert terveztĂĽnk. | Biopsy samples from operated colorectal carcinomas were used for the detection of human papillomaviruses (HPVs) by the nucleic acid hybridization probe, for the first time in Hungary. 432 of 802 cervix samples were positive by the nucleic acid hybridization probe. Randomly, 105 of the 432 samples were determined by both consensus and short fragment PCR methods. HPVs were detected in 41 samples. The HPV types were determined by the RFLP method. Both methods were adapted to the real-time PCR (RT-PCR) method. Cervix samples from asymptomatic patients (N = 191) were amplified with the consensus PCR method. In the event of positivity, the short fragment PCR method was performed to determine the HPV types by the linear array method. Simultaneously, a microchip method was designed for the detection of HPVs and determination of the HPV types
