The epigenetic control of E-box and Myc-dependent chromatin modifications regulate the licensing of lamin B2 origin during cell cycle
Recent genome-wide mapping of the mammalian replication origins has suggested the role of transcriptional regulatory elements in origin activation. However, the nature of chromatin modifications associated with such trans-factors or epigenetic marks imprinted on cis-elements during the spatio-temporal regulation of replication initiation remains enigmatic. To unveil the molecular underpinnings, we studied the human lamin B2 origin that spatially overlaps with TIMM 13 promoter. We observed an early G1-specific occupancy of c-Myc that facilitated the loading of mini chromosome maintenance protein (MCM) complex during subsequent mid-G1 phase rather stimulating TIMM 13 gene expression. Investigations on the Myc-induced downstream events suggested a direct interaction between c-Myc and histone methyltransferase mixed-lineage leukemia 1 that imparted histone H3K4me3 mark essential for both recruitment of acetylase complex HBO1 and hyperacetylation of histone H4. Contemporaneously, the nucleosome remodeling promoted the loading of MCM proteins at the origin. These chromatin modifications were under the tight control of active demethylation of E-box as evident from methylation profiling. The active demethylation was mediated by the Ten-eleven translocation (TET)-thymine DNA glycosylase-base excision repair (BER) pathway, which facilitated spatio-temporal occupancy of Myc. Intriguingly, the genome-wide 43% occurrence of E-box among the human origins could support our hypothesis that epigenetic control of E-box could be a molecular switch for the licensing of early replicating origins
A Casual Study on Impact of Green HRM Practices on Organisation Sustainability
Nowadays, our business environment has changed itsview from a traditional financial perspective tocompetency based and strategic based perspective. Thegreen movement across the world has given birth to theconcept of Green HRM. The concept became popularowing to the issues in the present business world, rangingfrom excessive consumption of natural resources, carboncredits, global warming, to pollution. All these results inserious industrial accidents decrease in life span, changesin climate, etc. In general, Green HR deals with practicingall HR activities with environment-friendly concerns. This, in turn, helps in the sustainability of businesses aswell as the employees. From the above result it can be concluded that six weeks pre - strenthening training is more effective in improving the performance in the tournament the Female wheelchair basketball players. Hence it can be recommended that 6 weeks pre – strengthening training is more effective, useful & performance oriented rather than the usual training program
Detect and Classify the Unpredictable Cyber-Attacks by using DNN Model
Machine learning techniques are often used to develop IDS by detecting and deploying fast and automated network attacks to torpedoes and host standards. However, there are many problems, as severe attacks change all the time and occur at very high levels that require a lot of resolution. There are many malicious packages available for further investigation by the cybersecurity community. However, one completed study did not provide a complete analysis to apply different machine learning algorithms on different media packages. Because of the persistent methods of attack and the dynamic nature of malware, it is important to systematically update and approve malicious packages that are available to the public. This paper explores the DNN, a type of comprehensive learning model, promoting flexible and appropriate IDS for detecting and deploying expected and unpredictable online attacks. Sustainable industrial development and rapid development of attacks need evaluation for some data developed over the years using static and dynamic methods. This type of research can help determine the best algorithm to identify future attacks. Comparative data for some commonly available malware provides a comprehensive comparison of DNN experiences with other class machine learning classifications. The best network parameters and network topologies for DNN are selected using the KDDCup 99 package with this hyperparameter selection method. The DNN model, which works well on KDDCup 99, works on other data, such as the NSL-KDD memory test. Our DNN model teaches how to transfer IDS information functions from multicultural.Multidisciplinary representations in a variety of encryption. Complex tests have shown that DNN performs better than conventional machine learning classification. Finally, we present a large and hybrid DNN torrent structure called Scale-Hybrid-IDS-AlertNet, which can be used to effectively monitor the impact of network traffic and host-level events to warn directly about cyber-attacks
“Do you think it is a Pandemic?” Apocalypse, Anxiety and the Environmental Grotesque in Paolo Bacigalupi’s The Windup Girl
Paolo Bacigalupi’s The Windup Girl (2009), set in the post fossil-fuel, post turbo-capitalist country of Thailand, portrays the shocking after effects of bioengineering and gene-hack modifications in food crops. The narrative depicts a country tottering on the brink of an agricultural apocalypse on account of food production being severely affected by crop driven anomalies and rogue diseases such as “cibiscosis” and “blister rust” transmitted by variants of mutating pests. Natural seed stock becomes completely supplanted by the new genetically engineered seeds which become sterile after a single seasonal cycle of sowing and harvesting. The native population of Thailand is adversely affected by the pandemic scenario, which becomes aggravated by an expedient “scientocracy” that is at the heart of the neocolonial enterprises of American megacorporations and calorie companies like Agrigen, PurCal and Redstar who hail gene hacking as the new future of food resources and market profiteering. The consumption of the gene-hacked produce spreads through crops and affects the human body in unimaginable ways thereby resulting in a considerable rise of health issues including digestive and respiratory failures.
This paper intends to articulate the idea of a pandemic, its historical understanding and affective influences in the context of a post techno-fossil fuel economy set in Thailand. It will analyse the idea of epidemiological colonialism; diseases introduced by colonising forces that reshape the natives’ existing environment thereby bringing forth a deep pandemic anxiety that percolates the collective memory of the Thai people. It also highlights how the novel portrays the conflict between traditional ecological knowledge systems and modern extractive enterprises that acts as a catalyst to hasten the destruction of sustainable systems of agriculture and food production that have endured the impact of climate change and ecological fallout. The paper will study the relevance of the pandemic as an agency of ecocatastrophe and its function in an eco-speculative science fictional narrative. Finally, the paper looks into the concept of the posthuman android, genetically modified humans in a “technologiade”, a society reconfigured by technoscience to resist the impact of environmental collapse, and explores how this trope is incorporated in Bacigalupi’s narrative to celebrate human striving for hope and survival in an imagined environmental future marked by a self-created agro-scientific grotesqueness
FORMULATION, IN VITRO, AND IN VIVO EVALUATION OF TASTE-MASKED ORAL DISINTEGRATING TABLETS OF FEXOFENADINE HYDROCHLORIDE USING SEMISYNTHETIC AND NATURAL SUPERDISINTEGRANTS
Objective: The aim of the present research work was to prepare and evaluate taste-masked oral disintegrating tablets (ODT) of Fexofenadine hydrochloride.
Methods: In the present work, Eudragit EPO, a taste masking agent and Karaya gum (GK) (natural), Sodium starch glycolate, and Croscarmellose sodium (CCS) (semi-synthetic) super disintegrants in three ratios (3, 6,9%) were used. Taste masked granules were prepared by different ratios of the drug: Eudragit EPO (1:1, 1:1.5, 1:2) by wet granulation method. The optimized taste-masked granules (1:2) were selected by sensory evaluation test to prepare 9 Fexofenadine ODT (FH1-FH9) formulations. These were evaluated for different parameters. Then desirability function (DF) was calculated for all formulations using disintegration time (DT), time taken for the tablet to release 90% of the drug (t 90%), and % drug dissolved in 10 min (Q10) as significant parameters.
Results: The best formulation (FH6) showed the highest DF value due to less DT and 100% in vitro drug release within 15 min. Thus, FH6 formulation containing 9% CCS was selected as the best among the prepared formulations to which in vivo studies were performed on rabbits to find maximum plasma concentration (Cmax), time taken to reach maximum concentration (tmax), area under the curve (AUC), rate of elimination (Kel), absorption rate (Ka) and half-life(t1/2) and compared with Fexofenadine (Allegra) marketed tablets. Total bioavailability was increased for the test formulation compared to the reference formulation.
Conclusion: Fexofenadine was successfully prepared as ODT with increased AUC and decreased tmax to which stability studies were conducted which were found to be stable
