Microbiological tests of fresh bull semen collected at breeding establishment
Bacterial and fungal contamination of the semen collected in production environment largely depends on the sanitary conditions of its collection as well as on the bacteria carrier state in breeding bulls. Since antimicrobials contained in the diluent used during semen product cryopreservation do not allow an objective assessment of semen contamination, a microbiological testing of fresh undiluted bull semen was carried out at the AO «Krasnoyarskagroplem» breeding establishment to identify the contamination source. The isolated opportunistic microorganism cultures were tested for their susceptibility to antibiotics for the purpose of effective treatment of bacteria carriers. The experiment was performed at the Department for Epizootiology, Microbiology, Parasitology and Veterinary and Sanitary Expertise of the Institute of Applied Biotechnology and Veterinary Medicine of the FSBEI HE «Krasnoyarsk State Agrarian University» and at the Veterinary Laboratory of the AO «Krasnoyarskagroplem» in 2017 and 2018. Semen was collected in accordance with GOST 32222-2013 and tested for veterinary and sanitary parameters according to GOST 32198-2013. Isolated microorganism cultures were tested for their susceptibility to antibiotics with disc-diffusion method according to the Methodical Guidelines 4.2.1890-04 «Testing of microorganisms for their susceptibility to antimicrobials» using discs containing eight antimicrobials. Analysis of microbiological test results showed that semen was rejected for sanitary reasons at the breeding establishment due to isolation of the following opportunistic microorganisms: Pseudomonas aeruginosa (6.4% samples) and Proteus vulgaris (8.5% sample) in 2017 and Pseudomonas aeruginosa (2.4% samples) in 2018. Other test parameters (total microbial count, coliform count) were within admissible limits. № anaerobes and pathogenic fungi were detected. Four Pseudomonas aeruginosa isolates and three Proteus vulgaris isolates recovered during the test have demonstrated susceptibility to ciprofloxacin that can be used for etiotropic treatment of bulls identified as bacteria carriers

Impact of feldspar and liquid Bio-fertilizer on fruit quantity and quality of olive trees Picual cv.
The present investigation was carried out in two consecutive seasons (2022 and 2023) on eight-year-old olive trees “Picual” cv., planted at 4×6m apart around (420 trees / Fed). Trees irrigated with drip irrigation system, at a private orchard. The experiment included 3 rates of Feldspar as source of naturel potassium at 2, 4, and 6 kg/ tree and potassium dissolving bacteria as liquid Bio-fertilizers supplemented (25 ml) at two or three times to investigate the yield and fruit quality of Picual olive trees. Data indicated that the yield of Picual olive trees fertilized with feldspar and liquid Bio-fertilizer increase to 70% over control. Picual olive trees treated with (4 kg Feldspar + 25 ml liquid Bio-fertilizer at three doses) achieved the highest increase in the yield for the both seasons and also gave the highest fruit weight, volume and fruit flesh/stone ratio of olive trees Picual cv. Concerning leaf nitrogen percentage (N%) The highest percentage of N% for the both years was recorded by treatment (6 kg Feldspar + 25 ml liquid Bio-fertilizer at three doses). With respect to leaf phosphor and potassium percentage, it could be seen that most treatments gave more or less results similar to the control
Impact of Natural stimulates and Antioxidant compounds on fruit quality and quantity of pomegranate trees cv. H116
This experiment was carried out during two seasons (2022 and 2023) to examine the effect of spraying pomegranate trees cv. H116 with extract algae at (2cm/l), salicylic acid at (0.5 g/l) and ascorbic acid at (0.5 g/l) either alone or in combination together on productivity, fruit properties and leaf mineral content. All treatments were applied as a foliar three times: at full blooming, after fruit sit and after 30 days from fruit set. The results showed that all treatments caused a significant increase in the yield and the leaf mineral content of trees compared to the control. Treated pomegranate trees with 0.5 g/l ascorbic acid + 0.5 g/l salicylic acid was the best treatment for enhancing the yield (kg), fruit weight (g) and fruit peel (%). Foliar l application on pomegranate trees with (2 cm/l algae extract + 0.5 g/l ascorbic acid + 0.5 g/l salicylic acid) was the best treatment in this study to improve the fruit juice content and its quality and obtain maximum leaf content of nitrogen, phosphorus and potassium
Association of jasmonic acid priming with multiple defense mechanisms in wheat plants under high salt stress
Salinity is a global conundrum that negatively affects various biometrics of agricultural crops. Jasmonic acid (JA) is a phytohormone that reinforces multilayered defense strategies against abiotic stress, including salinity. This study investigated the effect of JA (60 μM) on two wheat cultivars, namely ZM9 and YM25, exposed to NaCl (14.50 dSm−1) during two consecutive growing seasons. Morphologically, plants primed with JA enhanced the vegetative growth and yield components. The improvement of growth by JA priming is associated with increased photosynthetic pigments, stomatal conductance, intercellular CO2, maximal photosystem II efficiency, and transpiration rate of the stressed plants. Furthermore, wheat cultivars primed with JA showed a reduction in the swelling of the chloroplast, recovery of the disintegrated thylakoids grana, and increased plastoglobuli numbers compared to saline-treated plants. JA prevented dehydration of leaves by increasing relative water content and water use efficiency via reducing water and osmotic potential using proline as an osmoticum. There was a reduction in sodium (Na+) and increased potassium (K+) contents, indicating a significant role of JA priming in ionic homeostasis, which was associated with induction of the transporters, viz., SOS1, NHX2, and HVP1. Exogenously applied JA mitigated the inhibitory effect of salt stress in plants by increasing the endogenous levels of cytokinins and indole acetic acid, and reducing the abscisic acid (ABA) contents. In addition, the oxidative stress caused by increasing hydrogen peroxide in salt-stressed plants was restrained by JA, which was associated with increased α-tocopherol, phenolics, and flavonoids levels and triggered the activities of superoxide dismutase and ascorbate peroxidase activity. This increase in phenolics and flavonoids could be explained by the induction of phenylalanine ammonia-lyase activity. The results suggest that JA plays a key role at the morphological, biochemical, and genetic levels of stressed and non-stressed wheat plants which is reflected in yield attributes. Hierarchical cluster analysis and principal component analyses showed that salt sensitivity was associated with the increments of Na+, hydrogen peroxide, and ABA contents. The regulatory role of JA under salinity stress was interlinked with increased JA level which consequentially improved ion transporting, osmoregulation, and antioxidant defense
ЭТЕРИФИКАЦИЯ ЛИМОННОЙ КИСЛОТЫ АЛИФАТИЧЕСКИМИ СПИРТАМИ С2-С5
There is an ever increasing interest in the production and use of bio-based chemical substances for the preparation of polymeric materials designed to obtain children's toys, medical equipment and materials that are in contact with food. Trifunctional esters of citric acid - trialkyl citrates - obtained by esterification of citric acid with aliphatic alcohols C2-C5 are of hazard class 4, good plasticizing ability, compatibility with the polymer, a high flash temperature. Citric acid (global production of 0.85-1 mtpa) is a readily available raw material obtained from the waste of sugar production. The purpose of the study is to develop a model of citric acid esterification with linear alcohols C2-C5 in the presence of homogeneous acidic catalysts for the creation of a domestic technology for the synthesis of plasticizing compositions based on renewable raw materials. The selected catalyst is sulfuric acid. The correlation of reactivity of the aliphatic alcohols C2-C5 with their structure in the esterification of citric acid was studied. Some kinetic parameters (lnk0, Ea) were estimated. Synthesis modes were proposed. The reaction time of the preparation of trialkyl citrates at 96% conversion of citric acid (t = 90 °C) was calculated. Laboratory samples were obtained and tested. Some operational characteristics were determined.Существует постоянно растущий интерес к производству и использованию биовозобновляемых химических веществ в производстве полимерных материалов, предназначенных для получения детских игрушек, медицинского оборудования, находящихся в контакте с пищевыми продуктами. Тризамещенные эфиры лимонной кислоты - триалкилцитраты, получаемые этерификацией лимонной кислоты алифатическими спиртами C2-C5, имеют 4 класс опасности, хорошую пластифицирующую способность, совместимость с полимером, высокую температуру вспышки. Лимонная кислота (объем мирового производства 0.85-1 млн. т/год) является доступным сырьем, получаемым из отходов сахарных производств. Цель исследования - разработка модели этерификации лимонной кислоты линейными спиртами С2-С5 на гомогенных кислотных катализаторах для создания отечественной технологии синтеза пластифицирующих композиций на основе возобновляемого сырья. Выбран катализатор - серная кислота; изучена связь реакционной способности алифатических спиртов С2-С5 с их строением в реакции этерификации лимонной кислоты, оценены некоторые кинетические параметры (lnk0, Eакт), предложены режимы синтеза, рассчитано время реакции получения триалкилцитратов при конверсии лимонной кислоты 96% (t = 90 ºС). Наработаны лабораторные образцы и определены некоторые эксплуатационные характеристики
