Kualitas Semen Beku Kuda Dalam Pengencer Susu Skim Dan Dimitropoulos Dengan Dimetilformamida Sebagai Krioprotektan

Equine semen are far less tolerant in the freezing and thawing process than bull semen. The stallion spermatozoa are known susceptible to cold-shock relating with the content of their fatty acid on the plasma membrane. The extender is one of determining factors in the success of stallion semen cryopreservation, as an energy source and protector the cell from harmfull effect of cold shock. The common cryoprotective agent (CPA) for mammalian spermatozoa was glycerol, but for stallion semen cryopreservation, dimethyl formamide (DMF) was more suitable. This research was conducted to compare the success of the stallion semen cryopreservation in skim milk and dimitropoulos (DV) extender with DMF as cryoprotectant. Semen from three sexualy mature stallions was collected twice a week using an artificial vagina. Semen was evaluated macro- and microscopically and then divided into two tubes, diluted each of them with skim milk dan DV extender (1:1), centrifuged at 3 000 rpm for 15 minutes. The supernatant was removed and the pellet (spermatozoa) was re-diluted in skim DMF (SDMF) and DVDMF extender with the concentration of spermatozoa was 200x106 ml-1. The semen then packed in 0.3ml minitub straw equilibrated for two hours at 4-50C and frezee in liquid N2 vapor for 10 minutes. The assessment of sperm quality was conducted based on the percentage of sperm motility and viability. In this research, post-thawed semen in DVDMF showed the percentages of the motility (36.2%) and the viability (59.3%) higher (P < 0.05) than SDMF (28.5 and 48.0 %). In conclusion, the DVDMF extender provided better post-thawed semen quality than SDMF

Ovarian Dynamic in Ongole Grade Cattle After GnRH Injection in Ovsynch Protocol Based on Progesterone Device

PO cattle have weaknesses to show clear estrus signs which cause difficulty in artificial insemination implementation. The present study was designed to obtain ovarian dynamic as effect of GnRH injection in ovsynch protocol based on progesterone intravaginal device. Heifers (18) and cows (n= 12) were allocated to one of three groups. Cuemate-PGF2α (CP) group inserted with a Cuemate on day 0-7 and injected with prostaglandin on day 7. Cuemate-PGF2α-GnRH (CPG) group was treated as CP group with the addition of GnRH injection on day 9. GnRH-Cuemate-PGF2α-GnRH (GCPG) group was treated as CPG group with addition of GnRH injection on day 0. Ultrasonography was performed on days 0-3, day 7 until ovulation and 7 days after ovulation. Percentage of ovulation synchronization increased significantly (P&lt;0.01) between CP, CPG, and GCPG, respectively, both in heifers (16%, 50%, and 85%, respectively) and cows (0%, 60%, and 100%, respectively), on day 11. Preovulatory follicle diameters between CP, CPG, and GCPG treatments were not different significantly both in heifers (11.9±0.5, 11.9±0.5, and 12.1±0.6 mm, respectively) and cows (11.7±0.4, 11.8±0.7, and 11.1±0.6 mm, respectively). This study concluded that GCPG protocol increased the synchrony of ovulation rate both in cows and heifers, without affecting the follicle preovulatory and CL diameters

This study aimed to determine the amino acid sequencehomology, functional domains, the model predictionof protein structure and characteristics of genesencoding anti WSSV in black tiger shrimp (Penaeusmonodon) originated from Indramayu District. Theresearch method was qualitative descriptive analysis bycomparing the results of PCR isolation of genes encodinganti WSSV with GenBank sequence data. The resultsshowed that the sequence gene encoding anti WSSV inblack tiger shrimp DNA originated Indramayu genesequences homologous with Penaeus monodon AV gene,complate cds (no. Accession DQ641258.1) and Penaeusmonodon PMAV (PmAV) mRNA, complete cds (no.Accession AY3202750.1). C -type lectin is a functionaldomain of the amino acid sequences of anti WSSVin black tiger shrimp. The structure of the proteinencoding anti WSSV (c -type lectin) is characterized bythe presence of a signal peptide, alpha helix clusters, betastrand and there are in three cells zones. C-type lectinindicated could help improved the resistance of blacktiger shrimp from Indramayu district against WSSVand the c -type lectin could be found in various typesof organisms to be developed as an immunostimulatorysubstance

Persediaan merupakan bagian penting untuk melaksanakan kegiatan operasional dalam rangka pengabdian kepada masyarakat. Persediaan adalah aset lancar berupa barang atau perlengkapan yang digunakan untuk menunjang kegiatan operasional, seperti barang habis pakai, barang tidak habis pakai, dan barang bekas. Tanpa adanya persediaan, instansi pemerintah akan kesulitan dalam melakukan pelayanan kepada masyarakat yang secara populasi memiliki jumlah yang besar dan dari segi wilayah yang begitu luas. Aktifitas di lingkungan instansi pemerintahan juga terganggu tanpa adanya persediaan, karena persediaan juga meliputi barang-barang yang mendukung kegiatan administrasi dan kantor di instansi pemerintahan. Tujuan dari penelitian ini adalah untuk mengetahui bagaimana penerapan pernyataan standar akuntansi pemerintah 05 tentang akuntansi persediaan pada kantor wilayah kementerian agama di provinsi Sulawesi Utara. Metode yang digunakan adalah metode deskriptif analisis. Hasil penelitian menunjukkan bahwa penerapan pernyataan standar akuntansi 05 tentang akuntansi persediaan pada Kanwil Kementerian Agama Provinsi Sulawesi Utara telah sesuai dengan penerapannya dan aturan yang berlaku

Pengaruh Permainan Gobak Sodor terhadap Perkembangan Motor Ability SD Negeri 1 Ledo Kabupaten Bengkayang

The purpose of this research is to know the effective of gobak sodor game to motor development ability. The method of this reseach used pre- experimental. The sample in this research is fifth grade Elementary School of 1 Ledo in academic Year of 2012/2013 , the totally are 25 students. Research was generally consists of three stages. Giving the initial test is given before treatment , treatment in the research is giving learning the game gobak Sodor, and post-test after treatment. Based on the data analysis of obtained and after the t-test on the pre - test and post - test, the results obtained can get results is 2.622 T will be compared with the t table is 2.064. So count T > T table 5 % , T = 2.622 > 2.064 . T count value that is greater than the value of the T table. Thus, in this research there is improvement gobak sodor game against the motor development ability. Furthermore, this research has a percentage increase of 15.72 %

Penggunaan Media Alternatif pada Kultur In Vitro Jahe (Zingiber Officinale Rosc.) Varietas Gajah

The problem faced in cultivation of ginger is the availability of uniform plant propagation materials. Ginger is propagated through underground rhizomes. Growth of the rhizome is not uniform, since the shoots do not sprout at the same time. Most of the rhizomes were attacked by several diseases such as bacterial wilt, soft rot, and nematodes. Propagation of plant material through in-vitro culture is also an obstacle since the price of pure agar and chemicals is very expensive. Therefore, production of cheap, uniform and disease-free plant materials with rapid multiplication rate is necessary for the successful ginger cultivation. The use of alternative media on in-vitro culture of ginger (Zingiber officinale Rosc.) cv. Gajah was conducted in order to substitute cheaper alternative media for in-vitro culture of ginger. An experiment using two basic composition of media (MS and liquid fertilizer) and three different types of agar (seaweed, Swallow and Oxoid agar) was done at the Tissue Culture Laboratory of Center for Research and Development of Isotope and Radiation Technology, Jakarta. The result showed that the highest shoot height, number of shoot and leaf was obtained from medium composition of sea weed and MS. Whilst the medium of Oxoid agar and MS produced the longest and the highest number of root. The cheapest medium was found from seaweed and liquid fertilizer composition, whereas the most expensive medium was from Oxoid agar and MS composition. Seaweed and Swallow agar in MS media showed similar growth performance as well as Oxoid agar in development of ginger plantlets

Suatu penelitian dengan tujuan untuk mengkaji Perubahan nilai indeks morfo-anatomi dan penyimpanan energi pada beberapa organ tubuh ikan senggaringan selama masa perkembangan gonad telah dilaksanakan di Sungai Klawing, Purbalingga, Jawa Tengah. Parameter morfo-anatomi meliputi faktor kondisi (CF), indeks jaringan viseral (VSI), indeks sirip lemak (AFI), indeks jaringan hati (HSI), indeks jaringan gonad (GSI), dan kandungan energi pada organ/jaringan otot punggung, sirip lemak, organ viseral, hati, dan gonad telah diukur berdasarkan tingkat perkembangan gonadnya. Hasil penelitian menunjukkan bahwa selama masa perkembangan gonad terjadi Perubahan nilai-nilai parameter morfo-anatomi dan kandungan energi pada organ yang diukur, terjadi proses penyimpanan, perpindahan dan Perubahan materi berenergi baik untuk keperluan pematangan gonad maupun untuk aktivitas pemijahan
