    Asn12 and Asn278: Critical Residues for In Vitro Biological Activity of Reteplase

    Reteplase (rPA) is a thrombolytic agent used for the treatment of acute myocardial infarction. We studied the expression of rPA and its selected asparagine mutants after integration into the Pichia genome. Though methanol induction of the native and the rPA mutants showed similar expression levels (~200–250 mg/L), the mutants displayed significant loss of protease activity. Strikingly, the clot lysis activities of these mutants were considerably different. While mutation of Asn12 (N12P) of the Kringle 2 domain showed delayed clot lysis activity (t1/2 = 38 min) compared to the native rPA (t1/2 = 33 min), a faster rate of clot lysis (t1/2 = 27 min) was observed when the Asn278 (N278S) of the serine protease domain was mutated. Interestingly, the slowest clot lysis activity (t1/2 = 49 min) demonstrated by the double mutant (N12P, N278S) suggests the dominant role of Asn12 in regulating the fibrinolytic activity of rPA. The results presented in this paper indicate that the fibrinolytic and the proteolytic activities of rPA are independent of each other

    Growth and yield of transplanted rice as affected by different cultivars and weed management practices

    An experiment was conducted to study the competitive ability of rice cultivars in different weed management practices in transplanted rice. Five cultivars viz. BPT-5204, Malviya 36, HUBR2-1, Swarna and Adamchini were tested for three weed management practices viz. Hand weeding, Bispyribac-sodium@25g ha-1 and Weedy check. Among the different cultivars tested, the Swarna proved to be the best selection in producing higher grain (4.6 t ha-1) and straw yield (5 t ha-1) as well as harvest index (44.4%) due to more no. of tillers hill-1, dry matter accumulation, no. of panicles hill-1, no. of grains/panicle and test weight (20.3 g). The grain and straw yield in Swarnawere 130 and 43% higher than the lowest yielding cultivar Adamchini respectively. The best weed control was achieved by hand weeding thrice at 25, 55 and 85 DAT. The grain yield was 58 and 27 % higher through hand weeding than that of weedy check and Bispyribac-sodium applied treatment, respectively. To examine the performance of different rice cultivars in terms of growth and yield potential, evaluation of these cultivars is necessary with different weed management practices. Therefore, release of best weed suppressive rice cultivars for the farmers is important for improving the productivity as well as reduced cost of production for feeding the ever growing population of the country in the year to come

    Evaluation of saturated hydraulic conductivity from soil properties in an Inceptisol using different land cover and depths

    hree soil profiles from Regional Research Station of Bidhan Chandra Krishi Viswavidyalaya, Gayeshpur situated in New Alluvial zone of Nadia district, West Bengal were studied to assess the predictability of the hydraulic conductivity of the soil as influenced by different physical and chemical and properties of cultivated and forest land. The various statistical procedures were employed on the measured laboratory based data for comprehensive agree-ment of dependent hydraulic conductivity of soils as a model function of independent soil variables that is likely to be useful for different land cover systems. Soils are neutral in reaction, silty clay to silty clay loam in nature. Forest soil contained greater organic carbon (OC) (5.9 ± 0.16 g kg-1) compared to cultivated soil (4.4 ± 0.34 g kg-1). Jhau plan-tation recorded the highest value (6.8 g kg-1) of OC due to soil texture and cation exchange capacity (CEC). Soil hydraulic conductivity was greater in soil for cabbage and Sagun tree among the cultivated and forest soil studied with values 2.80 and 1.10 cmh -1. Correlation study showed a positive and negative relation with hydraulic conductiv-ity for sand (r= 0.68; P > 0.05) and clay (r= - 0.71; P > 0.05) respectively. Further, principal component analysis con-cluded that addition of bulk density with clay and sand can predict the hydraulic conductivity for different land uses

    Enhancing physiological metrics, yield, zinc bioavailability, and economic viability of Basmati rice through nano zinc fertilization and summer green manuring in semi–arid South Asian ecosystem

    During the summer and rainy seasons (April-October) of 2020 and 2021, two consecutive field experiments were conducted at the research farm of the ICAR-Indian Agricultural Research Institute, New Delhi, India. In this study, we examined the effects of summer green manuring crops (GM) and a variety of zinc fertilizers (ZnF) on Basmati rice (Oryza sativa L.) growth, physiological development, yield response, zinc nutrition and economic returns. A combination of GM residues and nano zinc fertilization helped significantly enhancing Basmati rice’s growth and its physiological development. Following the incorporation of Sesbania aculeata (Sesbania), successive Basmati rice physiological parameters were significantly improved, as well as grain, straw, biological yields, harvest index and economic returns. The highest Zn content of 15.1 mg kg -1 and the lowest of 11.8 mg kg -1 in milled rice grain were recorded in Sesbania green manuring (G2) and control i.e., in the fallow (G1), respectively. Coating onto urea with 0.2% nano zinc oxide (NZnCU) was observed to be more effective than other zinc sources in terms of growth parameters, yield attributes, zinc nutrition, grain and straw yields for succeeding Basmati rice crop; however, the effects were comparable to those of bulk zinc oxide-coated urea (BZnCU) of 1%. The highest Zn content of 15.1 mg kg -1 was recorded with the application of 1% BZnCU and the lowest of 11.96 mg kg -1 with the soil application of 5 kg Zn ha -1 through bulk ZnO in the milled rice grain. Application of 1% BZnCU led to a 26.25% increase in Zn content of milled rice grain compared to soil application of 5 kg Zn ha -1 through bulk ZnO. As a result, the combination of inclusion of Sesbania aculeata (Sesbania) residue and 0.2% NZnCU was identified as the most effective treatment, for Basmati rice growth and physiological development. A combination of nano Zn fertilization in conjunction with the incorporation of green manure can be advocated for better growth, physiological performance, zinc dense grains, and higher profitability of Basmati rice for farmers and consumers

    Not AvailableThe experiment was conducted at Central Tobacco Research Institute (CTRI), Rajahmundry, Andra Pradesh, 2013 to investigate the role of soil texture in weed seed germination and eff ect of depth of soil profi le on emergence of weed seeds. The soil samples were collected from 3 places viz. Rajahmundry, Katheru farm under CTRI both belong to East Godavari district and Chainnaigudem village in West Godavari district with sandy, clay and sandy loam in texture respectively. Soil samples were collected from 0-10, 10-20, 20-30 and 30-40 cm depth of soil profi le using core sampler. Each site represents were approximately 1300 m2 area and 32 samples from 4 depths for every site. So, total 144 samples were collected to conduct the experiment. Germinated weeds identifi ed and counted every week and 10 weeks study was carried out. Sandy, sandy loam and clay soil texture found signifi cant non-linear relationship between weed germination and soil depth. All three places recorded signifi cant interaction between depth and seed germination. Soil depth upto 20 cm recorded maximum weed emergence both monocotyledonous and dicotyledonous; however, dicotyledonous weeds recorded more in number than monocotyledon in four consecutive depths.
