    Developmental Trajectories of Aggression from Toddlerhood to Early Adolescence in Boys and Girls: Exploring Early Predictors and Later Outcomes

    Early behavior problems, such as childhood aggression, emerge in the second year of life and decrease prior to school entry for typically developing children. However, some children show frequent and persistent aggression and may be at risk for subsequent difficulties that lead to poor school and life outcomes. The current study aimed to identify aggression patterns in children from toddlerhood to early adolescence for boys and girls together and separately. This study also explored early influences on aggressive behaviors at age two, such as mothers’ parenting behaviors, cumulative family risk, and early child characteristics. A range of difficulties in early adolescence were investigated, including poor social skills, low academic success, internalizing problems (e.g., depression and anxiety), and delinquent behaviors. Data from a previous study of 3,000 families and children, the Early Head Start Research and Evaluation longitudinal study, which followed families and children from infancy to early adolescence were examined. Four aggression patterns were found, characterized by changes over time in the amount of aggressive behaviors exhibited, applicable to both boys and girls: low-stable, moderate-decreasing, moderate-increasing, and high-stable. Early parenting and development during toddlerhood discriminated these distinctive aggression patterns. Compared with children in the low-stable group, boys in the moderate-decreasing and high-stable groups, and girls in the high-stable group were less likely to have mothers using positive parenting behaviors. Boys in the moderate-decreasing group and girls in the high-stable group were more likely to show delayed language development. Boys in the moderate-decreasing and high-stable groups were more likely to indicate poor emotional regulation. Moreover, compared with children in the low-stable group, those in the moderate-decreasing, moderate-increasing, or high-stable groups tended to show difficulties in early adolescence, such as lack of social skills, lower academic success, more internalizing problems, and delinquent behaviors. Altogether, young children exhibiting high levels of aggression over time were at the highest risk for later social, behavioral, and academic problems when, at age two, they had mothers with less positive parenting behaviors or when they showed poor language development or poor emotional regulation

    BEHAVIOUR AND UPTAKE OF ENGINEERED GOLD NANOP ARTICLES IN AQUATIC SYSTEMS

    Nanotechnology is a rapidly growing industry of global economic importance. However, there is concern that the unique properties of engineered nanoparticles (ENPs) which can make them useful to society could mean that they pose a risk to the natural environment. There are still many uncertainties around the behaviour of ENPs in aquatic systems, their capacity to be taken up by aquatic organisms, and their potential toxic effects. This study therefore used four different surface functionalised gold (Au) nanoparticles as exemplars and explored their behaviour in various aquatic systems and their uptake into aquatic organisms. Studies into the behaviour of the study ENPs in standardized ecotoxicity test media showed that the particles exhibit very different aggregation behaviour depending on the test media type, the chemical composition of the test media and surface capping of the nanoparticles. Based on the results of the aggregation studies in a range of natural waters, a series of methods to predict size of Au particles in different water chemistries were developed. Results showed that there would likely be big differences in the aggregation of the different Au nanoparticle types in UK water types which implies that aggregation of ENPs will vary widely across surface waters. The uptake of the four Au nanoparticles into the aquatic invertebrate, Gammarous pulex, did not show obvious relationship between the aggregation state in a treatment and uptake suggesting that the widely accepted assumption that ENP uptake is related to particle size does not hold for the range of aggregation states studied. The results of this thesis showed that the degree of aggregation of ENPs and uptake into aquatic organisms would vary depending on the surface functionalisation of the ENPs and water chemistry. The results imply that aggregation of ENPs will vary widely across surface waters which make assessment of risks a challenge. Additionally, the uptake data indicate that factors other than particle size determine uptake of ENPs into organisms. Therefore, it may be necessary to develop new paradigms and models for risk assessment of ENPs

    Pressure changes in the endotracheal tube cuff in otorhinolaryngologic surgery: a prospective observational study

    ObjectiveInflation of the endotracheal tube cuff is needed for providing ventilation. Cuff pressure should be maintained inside the appropriate range to prevent critical airway complications. The purpose of this study is to evaluate the pressure changes in the endotracheal tube cuff during otorhinolaryngologic surgery.Design and methodThis single-center observational study was conducted at Severance Hospital in Korea between April 2020 and November 2020. Patients aged >20 years scheduled to undergo otorhinolaryngological surgical procedures were enrolled. Patients undergoing planned tracheostomy and those who were slated for uncuffed endotracheal tube use were excluded. Intubation was performed after the induction of general anesthesia. A pressure transducer was connected to the pilot balloon of the endotracheal tube, and cuff pressure was continuously monitored until extubation. If the cuff pressure was not appropriate for more than 5 min, it was adjusted to the appropriate range by injecting or removing air. The percentage of time for which the cuff pressure remained within the appropriate range was calculated and defined as the time in the therapeutic range (TTR). The presumed cause for the rise or fall in cuff pressure was identified.ResultsIn total 199 patients, alterations in cuff pressure outside the appropriate range occurred in 191 patients (96.0%). The mean TTR was 79.7% (SD 25.0%), and head and neck surgery had the lowest mean TTR of 69.0% compared to ear and nose surgeries (94.2 and 82.1%, respectively). Sixty-eight patients (34.2%) demonstrated inadequate endotracheal tube cuff pressure for more than 20% of the total anesthesia time. Twenty-six patients (13.1%) demonstrated optimal endotracheal tube cuff pressure for less than 50% of the total anesthesia time. The causative factors inducing inappropriate cuff pressure were found to vary, including positional changes, surgical procedure, anatomical manipulation, and anesthetic procedure.ConclusionIn otorhinolaryngologic surgery, cuff pressure increased or decreased outside the appropriate range due to various factors. Therefore, we suggest close continuous monitoring of cuff pressure during anesthesia for otorhinolaryngologic surgery.Clinical trial registrationclinicaltrials.gov, identifier NCT03938493

    Detection of Absorbing Aerosol Using Single Near-UV Radiance Measurements from a Cloud and Aerosol Imager

    The Ultra-Violet Aerosol Index (UVAI) is a practical parameter for detecting aerosols that absorb UV radiation, especially where other aerosol retrievals fail, such as over bright surfaces (e.g., deserts and clouds). However, typical UVAI retrieval requires at least two UV channels, while several satellite instruments, such as the Thermal And Near infrared Sensor for carbon Observation Cloud and Aerosol Imager (TANSO-CAI) instrument onboard a Greenhouse gases Observing SATellite (GOSAT), provide single channel UV radiances. In this study, a new UVAI retrieval method was developed which uses a single UV channel. A single channel aerosol index (SAI) is defined to measure the extent to which an absorbing aerosol state differs from its state with minimized absorption by aerosol. The SAI qualitatively represents absorbing aerosols by considering a 30-day minimum composite and the variability in aerosol absorption. This study examines the feasibility of detecting absorbing aerosols using a UV-constrained satellite, focusing on those which have a single UV channel. The Vector LInearized pseudo-spherical Discrete Ordinate Radiative Transfer (VLIDORT) was used to test the sensitivity of the SAI and UVAI to aerosol optical properties. The theoretical calculations showed that highly absorbing aerosols have a meaningful correlation with SAI. The retrieved SAI from OMI and operational OMI UVAI were also in good agreement when UVAI values were greater than 0.7 (the absorption criteria of UVAI). The retrieved SAI from the TANSO-CAI data was compared with operational OMI UVAI data, demonstrating a reasonable agreement and low rate of false detection for cases of absorbing aerosols in East Asia. The SAI retrieved from TANSO-CAI was in better agreement with OMI UVAI, particularly for the values greater than the absorbing threshold value of 0.7

    Temporal variability of surface air pollutants in megacities of South Korea

    This study investigated the various temporal (weekly, monthly, and inter-annual) variability of air pollutants (PM10, SO2, NO2, O-3, CO) in seven megacities in South Korea (Seoul, Busan, Incheon, Daegu, Gwangju, Daejeon, and Ulsan). We found that the general decreasing trend of PM10, SO2, NO2, and CO. An exceptional pollutant is O-3, showing a clear increasing trend consistently in all seven megacities. Seasonally PM10, SO2, NO2, and CO have the highest level in winter due to the large fossil-fuel combustion for the heating demand, but O-3 shows the maximum peak in summer related to the intensified photochemistry. Based on the analysis for percentile values of air pollutants, we recognized that some patterns of air pollutants in Korean megacities are overlooked: O-3 increase is not perfectly related to the NO2 pattern, somewhat high SO2 in the coastal cities, ambiguous weekly pattern on Monday (as a weekday) and Sunday (as a weekend). Through this comprehensive analysis of multiple air pollutants using the percentile values, the characteristic for various temporal change of air pollutants in Korean megacities can be better understood, and some useful ideas for the air quality control in the urban region can be also excavated

    Development of Machine Learning Models Predicting Estimated Blood Loss during Liver Transplant Surgery

    The incidence of major hemorrhage and transfusion during liver transplantation has decreased significantly over the past decade, but major bleeding remains a common expectation. Massive intraoperative hemorrhage during liver transplantation can lead to mortality or reoperation. This study aimed to develop machine learning models for the prediction of massive hemorrhage and a scoring system which is applicable to new patients. Data were retrospectively collected from patients aged >18 years who had undergone liver transplantation. These data included emergency information, donor information, demographic data, preoperative laboratory data, the etiology of hepatic failure, the Model for End-stage Liver Disease (MELD) score, surgical history, antiplatelet therapy, continuous renal replacement therapy (CRRT), the preoperative dose of vasopressor, and the estimated blood loss (EBL) during surgery. The logistic regression model was one of the best-performing machine learning models. The most important factors for the prediction of massive hemorrhage were the disease etiology, activated partial thromboplastin time (aPTT), operation duration, body temperature, MELD score, mean arterial pressure, serum creatinine, and pulse pressure. The risk-scoring system was developed using the odds ratios of these factors from the logistic model. The risk-scoring system showed good prediction performance and calibration (AUROC: 0.775, AUPR: 0.753)

    Resting-state prefrontal EEG biomarker in correlation with postoperative delirium in elderly patients

    Postoperative delirium (POD) is associated with adverse outcomes in elderly patients after surgery. Electroencephalography (EEG) can be used to develop a potential biomarker for degenerative cerebral dysfunctions, including mild cognitive impairment and dementia. This study aimed to explore the relationship between preoperative EEG and POD. We included 257 patients aged >70 years who underwent spinal surgery. We measured the median dominant frequency (MDF), which is a resting-state EEG biomarker involving intrinsic alpha oscillations that reflect an idle cortical state, from the prefrontal regions. Additionally, the mini-mental state examination and Montreal cognitive assessment (MoCA) were performed before surgery as well as 5 days after surgery. For long-term cognitive function follow up, the telephone interview for cognitive statusβ„’ (TICS) was performed 1 month and 1 year after surgery. Fifty-two (20.2%) patients were diagnosed with POD. A multivariable logistic regression analysis that included age, MoCA score, Charlson comorbidity index score, Mini Nutritional Assessment, and the MDF as variables revealed that the MDF had a significant odds ratio of 0.48 (95% confidence interval 0.27–0.85). Among the patients with POD, the postoperative neurocognitive disorders could last up to 1 year. Low MDF on preoperative EEG was associated with POD in elderly patients undergoing surgery. EEG could be a novel potential tool for identifying patients at a high risk of POD

    Presence of plasmid-mediated quinolone resistance (PMQR) genes in non-typhoidal Salmonella strains with reduced susceptibility to fluoroquinolones isolated from human salmonellosis in Gyeonggi-do, South Korea from 2016 to 2019

    Non-typhoidal salmonellosis remains a pressing public health problem worldwide. Quinolones, particularly fluoroquinolones, are widely used to treat various infections, including non-typhoidal salmonellosis, which can be a serious illness. The emergence of fluoroquinolone-resistant Salmonella has resulted in treatment failure and high mortality rates. In this study, we estimated the presence of plasmid-mediated quinolone resistance (PMQR) genes in Salmonella enterica isolated from human salmonellosis patients in South Korea from 2016 to 2019. We evaluated the association of these genes with fluoroquinolone susceptibility. Antimicrobial susceptibility tests for Salmonella isolates were performed using the Vitek II system, and the minimum inhibitory concentrations (MIC) of ciprofloxacin and levofloxacin were determined using the E-test method. Plasmid-mediated quinolone resistance (PMQR) genes were detected by PCR amplification and quinolone resistance-determining regions (QRDRs) of the gyrA and parC genes were analyzed following Sanger sequencing of the PCR products. Thirty-four Salmonella strains with reduced susceptibility to fluoroquinolones (ciprofloxacin MIC β‰₯ 0.125 Β΅g/mL and levofloxacin MIC β‰₯ 0.25 Β΅g/mL) were selected from 208 human clinical Salmonella isolates. Among them, 22 Salmonella strains harbored one PMQR gene (qnrA, qnrB, or qnrS), and three Salmonella strains carried two PMQR genes (qnrS and aac(6β€²)-Ib-cr or qnrA and qnrB). qnrS was the most common PMQR gene. Serotyping revealed that Salmonella 4,[5]12:i:- (32.4%, 11/34) and Salmonella Typhimurium (29.4%, 10/34) were the two most predominant serovars, and Multi-locus sequence typing (MLST) showed that ST19 and ST34 were the most frequent sequence types. In conclusion, qnr gene-positive Salmonella 4,[5],12:i:- and Salmonella Typhimurium were the main serovars responsible for reduced susceptibility to fluoroquinolones. Therefore, our findings suggest that PMQR-positive Salmonella strains, which can be isolated from various samples including human, food, and the environment, should be carefully monitored.This research was supported by a Grant (19162MFDS037) from the Ministry of Food and Drug Safety in 2020

    Basic Fibroblast Growth Factor Activates MEK/ERK Cell Signaling Pathway and Stimulates the Proliferation of Chicken Primordial Germ Cells

    BACKGROUND: Long-term maintenance of avian primordial germ cells (PGCs) in vitro has tremendous potential because it can be used to deepen our understanding of the biology of PGCs. A transgenic bioreactor based on the unique migration of PGCs toward the recipients' sex cord via the bloodstream and thereby creating a germline chimeric bird has many potential applications. However, the growth factors and the signaling pathway essential for inducing proliferation of chicken PGCs are unknown. METHODOLOGY/PRINCIPAL FINDINGS: Therefore, we conducted this study to investigate the effects of various combinations of growth factors on the survival and proliferation of PGCs under feeder-free conditions. We observed proliferation of PGCs in media containing bFGF. Subsequent characterization confirmed that the cultured PGCs maintained expression of PGC-specific markers, telomerase activity, normal migrational activity, and germline transmission. We also found that bFGF activates the mitogen-activated protein kinase kinase/extracellular-signal regulated kinase (MEK/ERK) signaling. Also, the expression of 133 transcripts was reversibly altered by bFGF withdrawal. CONCLUSIONS/SIGNIFICANCE: Our results demonstrate that chicken PGCs can be maintained in vitro without any differentiation or dedifferentiation in feeder free culture conditions, and subsequent analysis revealed that bFGF is one of the key factors that enable proliferation of chicken PGCs via MEK/ERK signaling regulating downstream genes that may be important for PGC proliferation and survival
