    Protective effects of Scutellaria baicalensis Georgi against hydrogen peroxide-induced DNA damage and apoptosis in HaCaT human skin keratinocytes

    Oxidative stress due to excessive accumulation of reactive oxygen species (ROS) is one of the risk factors for the development of several chronic diseases. In this study, we investigated the protective effects of Scutellaria bai- calensis rhizome ethanol extract (SBRE) against oxidative stress-induced cellular damage and elucidated the un- derlying mechanisms in the HaCaT human skin keratinocyte cell line. Our results revealed that treatment with SBRE prior to hydrogen peroxide (H2O2) exposure significantly increased viability of aCaT cells. SBRE also effectively attenuated H2O2-induced comet tail formation and inhibited the H2O2-induced phosphorylation levels of the histone ÎłH2AX, as well as the number of apoptotic bodies and Annexin V-positive cells. In addition, SBRE exhibited scavenging activity against intracellular ROS generation and restored the mitochondrial membrane po- tential loss by H2O2. Moreover, H2O2 enhanced the cleavage of caspase-3 and degradation of poly (ADP-ribose)- polymerase, a typical substrate protein of activated caspase-3, as well as DNA fragmentation; however, these events were almost totally reversed by pretreatment with SBRE. Furthermore, SBRE increased the levels of heme oxygenase-1 (HO-1), which is a potent antioxidant enzyme, associated with the induction of nuclear fac- tor-erythroid 2-related factor 2 (Nrf2). According to our data, SBRE is able to protect HaCaT cells from H2O2- induced DNA damage and apoptosis through blocking cellular damage related to oxidative stress through a mech-anism that would affect ROS elimination and activating the Nrf2/HO-1 signaling pathway

    Prognostic Value of Metastatic Tumoral Caveolin-1 Expression in Patients with Resected Gastric Cancer

    Objective. Caveolin-1 (Cav-1), as the main component of caveolae, has complex roles in tumourigenesis in human malignancies. We investigated Cav-1 in primary and metastatic tumor cells of gastric cancer (GC) and its association with clinical outcomes. Methods. We retrieved 145 cases of GC who had undergone curative gastrectomy. The expression levels of Cav-1 was evaluated by immunohistochemistry, and its association with clinicopathological parameters and patient survival was analyzed. Results. High expression of Cav-1 protein of the GC in the stomach and metastatic lymph node was 12.4% (18/145) and 16.5% (15/91). In the multivariate analysis, tumoral Cav-1 protein in metastatic lymph node showed prognostic significance for relapse-free survival (RFS, HR, 3.934; 95% CI, 1.882–8.224; P=0.001) and cancer-specific survival outcome (CSS, HR, 2.681; 95% CI, 1.613–8.623; P=0.002). Among the GCs with metastatic lymph node, it remained as a strong indicator of poor prognosis for RFS (HR, 3.136; 95% CI, 1.444–6.810; P=0.004) and CSS (HR, 2.509; 95% CI, 1.078–5.837; P=0.032). Conclusion. High expression of tumoral Cav-1 protein in metastatic lymph node is associated with unfavorable prognosis of curative resected GC, indicating the potential of novel prognostic markers

    Gas Temperature Effect on Discharge-Mode Characteristics of Atmospheric-Pressure Dielectric Barrier Discharge in a Helium–Oxygen Mixture

    For a better understanding of gas temperature effects on plasma characteristics, a numerical study has been carried out for a dielectric barrier discharge (DBD) with a helium–oxygen mixture at atmospheric pressure. A one-dimensional time-dependent simulation code has been developed to solve continuity equations for plasma species and Poisson's equation for electric field calculation for a parallel-plate DBD reactor. To include temperature effects, gas heating by enthalpy change and Joule heating with ionic current movement are considered in the helium–oxygen plasma including 13 species reacting with one another according to 34 reactions depending on the gas temperature. Varying the ambient temperature from 300 K to 500 K, the plasma characteristics are calculated for the temporal variations and spatial distributions of electric field and species densities in the DBD region, and the different features of discharge modes are described by the voltage–current characteristic curves. A glowlike mode, which typically shows the formation of cathode fall, Faraday dark space, negative glow, and positive column in the spatial distributions of electric field and plasma density, is found in the discharge at a low ambient temperature, while a Townsend discharge mode with moderate electric field intensity and lower electron density is characterized at higher ambient temperatures. The temperature-dependent reactions strongly influence the generation and loss of species in the DBD plasma, and the decomposition of O3 into O or O2 and the quenching of metastable helium by the resultant O or O2 play an important role in determining the distinct discharge mode in the DBD of a He-O2 mixture. Furthermore, it is understood that the discharge-mode transition is controllable by the coupled effects of oxygen additive concentration, frequency, and gas temperature. A small amount of O2 additive or a high-frequency operation exhibits a glow mode in a specific range of ambient temperature, of which reason can be explained by density variation and quenching of helium metastable species caused by the produced oxygen-related species

    Comparison of air pollution and the prevalence of allergy-related diseases in Incheon and Jeju City

    PurposeA high level of air pollutants can increase the number of patients with allergy-related diseases such as asthma and allergic rhinitis (AR). To analyze the association between air pollution and allergic disease, we investigated 2 areas in Korea: Incheon, an industrial area, and Jeju, a non-industrialized area.MethodsSecond grade students at elementary schools (11 schools in Incheon and 45 schools in Jeju) were examined in a cross-sectional study. A questionnaire was used and a skin prick test was performed. The levels of NO2, CO2, O3, particulate matter (PM) PM10/2.5, formaldehyde, tVOCs, and dust mites in the classrooms and grounds were determined.ResultsThe levels of outdoor CO, PM10, and PM2.5 were significantly higher in Incheon (P<0.01). The levels of indoor CO, CO2, PM10, PM2.5 were significantly higher in Incheon (P<0.01). The prevalence rates of AR symptoms at any time, AR symptoms during the last 12 months, diagnosis of rhinitis at any time, and AR treatment during the last 12 months were significantly higher in Incheon (P<0.01). The prevalence rate of wheezing or whistling at any time, and wheezing during the last 12 months were significantly higher in Incheon (P<0.01).ConclusionWe found that the children living in Incheon, which was more polluted than Jeju, had a higher rate of AR and asthma symptoms compared to children in Jeju. To determine the effect of air pollution on the development of the AR and asthma, further studies are needed

    A Study of Parameters Related to the Etch Rate for a Dry Etch Process Using NF 3

    The characteristics of the dry etching of SiNx:H thin films for display devices using SF6/O2 and NF3/O2 were investigated using a dual-frequency capacitively coupled plasma reactive ion etching (CCP-RIE) system. The investigation was carried out by varying the RF power ratio (13.56 MHz/2 MHz), pressure, and gas flow ratio. For the SiNx:H film, the etch rates obtained using NF3/O2 were higher than those obtained using SF6/O2 under various process conditions. The relationships between the etch rates and the usual monitoring parameters—the optical emission spectroscopy (OES) intensity of atomic fluorine (685.1 nm and 702.89 nm) and the voltages VH and VL—were investigated. The OES intensity data indicated a correlation between the bulk plasma density and the atomic fluorine density. The etch rate was proportional to the product of the OES intensity of atomic fluorine (I(F)) and the square root of the voltages (Vh+Vl) on the assumption that the velocity of the reactive fluorine was proportional to the square root of the voltages

    Cyclic Loading Test of Structural Walls with Small Openings

    This study aimed to investigate the effects of small openings on the structural performance of reinforced-concrete (RC) structural walls. Cyclic lateral loading tests were conducted on five RC walls with an aspect ratio of 2.6 and small openings. The main test parameter was the size of the small openings. The specimens were designed to fail after flexural yielding, considering the typical failure mode of slender RC walls. The structural performances of the test specimens were analyzed based on the test results in terms of the load-carrying capacity (flexural strength), hysteretic behavior, strain distribution, and the size of the openings. The specimens showed flexural yielding regardless of the size of the openings, and the flexural strength and deformation capacity were not significantly affected by the small openings. This result indicates that small openings do not affect the flexural behavior of slender walls if the walls have sufficient shear resistance and the small openings are located away from the extreme compressive end and in the compression zone where compressive stress does not decrease.This work was fnancially supported by the National Research Foundation of Korea (NRF) Grant funded by the Korea Government (MSIT) (No. 2018R1A2B6007559

    The Effect of Total Thyroidectomy on the Speech Production

    ObjectivesVoice and speech alternations that can occur after total thyroidectomy are usually due to recurrent or superior laryngeal nerve injury. These alterations may also be associated with other extralaryngeal factors, such as neck muscle dysfunction and scar contracture of the neck. We performed a prospective acoustic analysis on speech changes after surgery, in the absence of laryngeal nerve injury.MethodsPatients aged 19 to 58 years undergoing total thyroidectomy, in the absence of laryngeal/pulmonary disease, previous neck surgery, or other malignant diseases, were recruited prospectively. For the running speech analysis, the speaking fundamental frequencies (SFo), range of SFo and speaking intensity were evaluated before surgery, 7 days, and 1 and 3 months after surgery. For consonant analysis, the acoustic distinctions of stop consonant, the voice onset time (VOT), vowel duration and closure duration were evaluated at the same periods.ResultsSFo and range of SFo were specifically diminished after surgery, while speaking intensities were not changed significantly after surgery. The thyroidectomized speakers displayed systematically varied VOT for the consonant production, which was phonetically representative. However, VOT after surgery could be longer in the strong aspirated and glottalized stops, but not in the lax stop than before surgery. The vowel and closure durations were not affected before and after surgery.ConclusionPatients with thyroidectomy have some difficulty of pitch control and consonant articulation during speaking. VOT is also one of the meaningful acoustic parameters and provide a reference for comparing acoustic measures before and after thyroidectomy
