Dynamics of Primordial Black Hole Formation
We present a numerical investigation of the gravitational collapse of
horizon-size density fluctuations to primordial black holes (PBHs) during the
radiation-dominated phase of the Early Universe. The collapse dynamics of three
different families of initial perturbation shapes, imposed at the time of
horizon crossing, is computed. The perturbation threshold for black hole
formation, needed for estimations of the cosmological PBH mass function, is
found to be rather than the generally employed
, if is defined as \Delta M/\mh, the
relative excess mass within the initial horizon volume. In order to study the
accretion onto the newly formed black holes, we use a numerical scheme that
allows us to follow the evolution for long times after formation of the event
horizon. In general, small black holes (compared to the horizon mass at the
onset of the collapse) give rise to a fluid bounce that effectively shuts off
accretion onto the black hole, while large ones do not. In both cases, the
growth of the black hole mass owing to accretion is insignificant. Furthermore,
the scaling of black hole mass with distance from the formation threshold,
known to occur in near-critical gravitational collapse, is demonstrated to

In situ measurements of rock salt permeability changes due to nearby excavation
The Small-Scale Mine-By was an in situ experiment to measure changes in brine and gas permeability of rock salt as a result of nearby excavation. A series of small-volume pressurized brine- and gas-filled test intervals were established 8 m beneath the floor of Room L1 in the WIPP underground. The test intervals were isolated in the bottom of the 4.8-cm diameter monitoring boreholes with inflatable rubber packers, and are initially pressurized to about 2 MPa. Both brine- and gas-filled test intervals were located 1.25, 1.5, 2, 3, and 4 r from the center of a planned large-diameter hole, where r is the radius of the large-diameter hole. Prior to the drilling of the large-diameter borehole, the responses of both the brine- and gas-filled test intervals were consistent with the formation modeled as a very low permeability, low porosity porous medium with a significant pore (brine) pressure and no measurable gas permeability. The drilling of the mine-by borehole created a zone of dilated, partially saturated rock out to about 1.5 r. The formation pressure increases from near zero at 1.5 r to the pre-excavation value at 4 r. Injection tests reveal a gradient of brine permeabilities from 5 {times} 10{sup {minus}18} m{sup 2} at 1.25 r to about the pre-excavation value (10{sup {minus}21} m{sup 2}) by 3 r. Discrete vortex representation of magnetohydrodynamics
We present an alternative approach to statistical analysis of an intermittent ideal MHD fluid in two dimensions, based on the hydrodynamical discrete vortex model applied to the Elsasser variables. The model contains negative temperature states which predict the formation of magnetic islands, but also includes a natural limit under which the equilibrium states revert to the familiar twin-vortex states predicted by hydrodynamical turbulence theories. Numerical dynamical calculations yield equilibrium spectra in agreement with the theoretical predictions

Tests of Fermilab built 40 mm aperture full length SSC dipole magnets
Several 40 mm aperture, 17 m long dipoles have been built by Fermilab as developmental prototypes for the Superconducting Super Collider. These magnets differ from those manufactured at Brookhaven National Laboratory in that they have an external inner-outer coil splice design, a collet style end clamp assembly, a new, analytically designed minimum stress coil end design, and a new insulation system which does not employ shims or shoes''. In addition, the magnets were built using production-style tooling. The magnets were tested at the Fermilab Magnet Testing Facility. Quench testing and mechanical measurement results are presented and analyzed with emphasis on the new design and fabrication features of these magnets. 13 refs., 5 figs
Characteristics of the nuclear (18S, 5.8S, 28S and 5S) and mitochondrial (12S and 16S) rRNA genes of Apis mellifera (Insecta: Hymenoptera): structure, organization, and retrotransposable elements
As an accompanying manuscript to the release of the honey bee genome, we report the entire sequence of the nuclear (18S, 5.8S, 28S and 5S) and mitochondrial (12S and 16S) ribosomal RNA (rRNA)-encoding gene sequences (rDNA) and related internally and externally transcribed spacer regions of Apis mellifera (Insecta: Hymenoptera: Apocrita). Additionally, we predict secondary structures for the mature rRNA molecules based on comparative sequence analyses with other arthropod taxa and reference to recently published crystal structures of the ribosome. In general, the structures of honey bee rRNAs are in agreement with previously predicted rRNA models from other arthropods in core regions of the rRNA, with little additional expansion in non-conserved regions. Our multiple sequence alignments are made available on several public databases and provide a preliminary establishment of a global structural model of all rRNAs from the insects. Additionally, we provide conserved stretches of sequences flanking the rDNA cistrons that comprise the externally transcribed spacer regions (ETS) and part of the intergenic spacer region (IGS), including several repetitive motifs. Finally, we report the occurrence of retrotransposition in the nuclear large subunit rDNA, as R2 elements are present in the usual insertion points found in other arthropods. Interestingly, functional R1 elements usually present in the genomes of insects were not detected in the honey bee rRNA genes. The reverse transcriptase products of the R2 elements are deduced from their putative open reading frames and structurally aligned with those from another hymenopteran insect, the jewel wasp Nasonia (Pteromalidae). Stretches of conserved amino acids shared between Apis and Nasonia are illustrated and serve as potential sites for primer design, as target amplicons within these R2 elements may serve as novel phylogenetic markers for Hymenoptera. Given the impending completion of the sequencing of the Nasonia genome, we expect our report eventually to shed light on the evolution of the hymenopteran genome within higher insects, particularly regarding the relative maintenance of conserved rDNA genes, related variable spacer regions and retrotransposable elements
Allelic Heterogeneity at the CRP Locus Identified by Whole-Genome Sequencing in Multi-ancestry Cohorts
Whole-genome sequencing (WGS) can improve assessment of low-frequency and rare variants, particularly in non-European populations that have been underrepresented in existing genomic studies. The genetic determinants of C-reactive protein (CRP), a biomarker of chronic inflammation, have been extensively studied, with existing genome-wide association studies (GWASs) conducted in >200,000 individuals of European ancestry. In order to discover novel loci associated with CRP levels, we examined a multi-ancestry population (n = 23,279) with WGS (âŒ38Ă coverage) from the Trans-Omics for Precision Medicine (TOPMed) program. We found evidence for eight distinct associations at the CRP locus, including two variants that have not been identified previously (rs11265259 and rs181704186), both of which are non-coding and more common in individuals of African ancestry (âŒ10% and âŒ1% minor allele frequency, respectively, and rare or monomorphic in 1000 Genomes populations of East Asian, South Asian, and European ancestry). We show that the minor (G) allele of rs181704186 is associated with lower CRP levels and decreased transcriptional activity and protein binding in vitro, providing a plausible molecular mechanism for this African ancestry-specific signal. The individuals homozygous for rs181704186-G have a mean CRP level of 0.23 mg/L, in contrast to individuals heterozygous for rs181704186 with mean CRP of 2.97 mg/L and major allele homozygotes with mean CRP of 4.11 mg/L. This study demonstrates the utility of WGS in multi-ethnic populations to drive discovery of complex trait associations of large effect and to identify functional alleles in noncoding regulatory regions

Politics ahead of patients: The battle between medical and chiropractic professional associations over the inclusion of chiropractic in the American Medicare System
Health care professions struggling for legitimacy, recognition, and market share can become disoriented to their priorities. Health care practitioners are expected to put the interests of patients first. Professional associations represent the interests of their members. So when a professional association is composed of health care practitioners, its interests may differ from those of patients, creating a conflict for members. In addition, sometimes practitionersâ perspectives may be altered by indoctrination in a belief system, or misinformation, so that a practitioner could be confused about the reality of patient needs. Politicians, in attempting to find an expedient compromise, can value a âwinâ in the legislative arena over the effects of that legislation. These forces all figure into the events that led to the acceptance of chiropractic into the American Medicare system. Two health care systems in a political fight lost sight of their main purpose: to provide care to patients without doing harm.

Genetic determinants of telomere length from 109,122 ancestrally diverse whole-genome sequences in TOPMed
Genetic studies on telomere length are important for understanding age-related diseases. Prior GWASs for leukocyte TL have been limited to European and Asian populations. Here, we report the first sequencing-based association study for TL across ancestrally diverse individuals (European, African, Asian, and Hispanic/Latino) from the NHLBI Trans-Omics for Precision Medicine (TOPMed) program. We used whole-genome sequencing (WGS) of whole blood for variant genotype calling and the bioinformatic estimation of telomere length in n = 109,122 individuals. We identified 59 sentinel variants (p < 5 Ă 10â9) in 36 loci associated with telomere length, including 20 newly associated loci (13 were replicated in external datasets). There was little evidence of effect size heterogeneity across populations. Fine-mapping at OBFC1 indicated that the independent signals colocalized with cell-type-specific eQTLs for OBFC1 (STN1). Using a multi-variant gene-based approach, we identified two genes newly implicated in telomere length, DCLRE1B (SNM1B) and PARN. In PheWAS, we demonstrated that our TL polygenic trait scores (PTSs) were associated with an increased risk of cancer-related phenotypes
