    The virtual ethnographic study by Kathryn Stam and Michael Scialdone seeks to understand social interaction through the Massively Multiplayer Online Role-Playing Games (MMORPG) communities of Furcadia and Runescape. Their article on “Where Dreams and Dragons Meet. An Ethnographic Analysis of two Examples of Massive Multiplayer Online Role-Playing Games“ attempt to understand some key aspects of this genre of online activity and interaction. Using an adaptation of Grounded Theory and analytic induction methods, they explored the nature of these two online communities and the interaction between characters. In this context they discuss some of the main methodological and ethical issues such as the difference between public versus private spheres, the concept of consent in challenging environments, new considerations of reciprocity within the game, the building of online trust, and representation of the researcher of him- or herself

    Spectral and Temporal Variability of Earth Observed in Polarization

    We present a comprehensive set of spectropolarimetric observations of Earthshine as obtained by FORS2 at the VLT for phase angles from 50degree to 135degree (Sun-Earth-Moon angle), covering a spectral range from 430nm to 920nm. The degree of polarization in BVRI passbands, the differential polarization vegetation index, and the equivalent width of the O2A polarization band around 760nm are determined with absolute errors around 0.1 percent in the degree of polarization. Earthshine polarization spectra are corrected for the effect of depolarization introduced by backscattering on the lunar surface, introducing systematic errors of the order of 1 percent in the degree of polarization. Distinct viewing sceneries such as observing the Atlantic or Pacific side in Earthshine yield statistically different phase curves. The equivalent width defined for the O2A band polarization is found to vary from -5nm to +2nm. A differential polarized vegetation index is introduced and reveals a larger vegetation signal for those viewing sceneries that contain larger fractions of vegetated surface areas. We corroborate the observed correlations with theoretical models from the literature, and conclude that the Vegetation Red Edge(VRE) is a robust and sensitive signature in polarization spectra of planet Earth. The overall behaviour of polarization of planet Earth in the continuum and in the O2A band can be explained by existing models. Biosignatures such as the O2A band and the VRE are detectable in Earthshine polarization with a high degree of significance and sensitivity. An in-depth understanding of Earthshines temporal and spectral variability requires improved models of Earths biosphere, as a prerequisite to interpret possible detections of polarised biosignatures in earthlike exoplanets in the future.Comment: 19 pages, 14 figures, 3 table

    DE-STRESS:A user-friendly web application for the evaluation of protein designs

    De novo protein design is a rapidly growing field, and there are now many interesting and useful examples of designed proteins in the literature. However, most designs could be classed as failures when characterised in the lab, usually as a result of low expression, misfolding, aggregation or lack of function. This high attrition rate makes protein design unreliable and costly. It is possible that some of these failures could be caught earlier in the design process if it were quick and easy to generate information and a set of high-quality metrics regarding designs, which could be used to make reproducible and data-driven decisions about which designs to characterise experimentally. We present DE-STRESS (DEsigned STRucture Evaluation ServiceS), a web application for evaluating structural models of designed and engineered proteins. DE-STRESS has been designed to be simple, intuitive to use and responsive. It provides a wealth of information regarding designs, as well as tools to help contextualise the results and formally describe the properties that a design requires to be fit for purpose

    The Impact of DSS Use and Information Load on Errors and Decision Quality

    This paper uses a laboratory experiment to examine the effect of DSS use on the decision maker‘s error patterns and decision quality. The DSS used in our experiments is the widely used Expert Choice (EC) implementation of the Analytic Hierarchy Process. Perhaps surprisingly, our experiments do not provide general support for the often tacit assumption that the use of a DSS such as EC improves decision quality. Rather, we find that, whereas a DSS can help decision makers develop a better understanding of the essence of a decision problem and can reduce logical errors (especially if the information load is high), it is also susceptible to introducing accidental effects such as mechanical errors. In some cases, as in our study, the accidental errors may outweigh the benefits of using a DSS, leading to lower quality decisions

    An NMRA-like protein regulates gene expression in Phytophthora capsici to drive the infection cycle on tomato

    Phytophthora spp. cause devastating disease epidemics on important crop plants and pose a grave threat to global crop production. Critically, Phytophthora pathogens represent a distinct evolutionary lineage in which pathogenicity has been acquired independently. Therefore, there is an urgent need to understand and disrupt the processes that drive infection if we aspire to defeat oomycete pathogens in the field. One area that has received little attention thus far in this respect is the regulation of Phytophthora gene expression during infection. Here, we characterize PcNMRAL1 (Phyca11_505845), a homolog of the Aspergillus nidulans nitrogen metabolite repression regulator NMRA and demonstrate a role for this protein in progression of the Phytophthora capsici infection cycle. PcNmrAL1 is coexpressed with the biotrophic marker gene PcHmp1 (haustorial membrane protein 1) and, when overexpressed, extends the biotrophic infection stage. Microarray analyses revealed that PcNmrAL1 overexpression in P. capsici leads to large-scale transcriptional changes during infection and in vitro. Importantly, detailed analysis reveals that PcNmrAL1 overexpression induces biotrophy-associated genes while repressing those associated with necrotrophy. In addition to factors controlling transcription, translation, and nitrogen metabolism, PcNMRAL1 helps regulate the expression of a considerable effector repertoire in P. capsici. Our data suggests that PcNMRAL1 is a transcriptional regulator that mediates the biotrophy to necrotrophy transition. PcNMRAL1 represents a novel factor that may drive the Phytophthora disease cycle on crops. This study provides the first insight into mechanisms that regulate infection-related processes in Phytophthora spp. and provides a platform for further studies aimed at disabling pathogenesis and preventing crop losses. </jats:p

    Functional Independence and Health-related Functional Status Following Spinal Cord Injury:A Prospective Study of the Association with Physical Capacity

    Objective: To determine changes in functional independence following spinal cord injury and to evaluate the association between functional independence and physical capacity. Design: Multi-centre prospective cohort study.Subjects: Patients with spinal cord injury admitted for initial rehabilitation.Methods: The motor Functional Independence Measure (FIMmotor) was determined at the start of rehabilitation (n = 176), 3 months later (n = 124), at discharge (n = 160) and one year after discharge from inpatient rehabilitation (n = 133). One year after discharge, physical and social dimensions of health-related functional status (Sickness Impact Profile 68; SIP68) were determined. On each occasion, physical capacity was established by measuring arm muscle strength, peak power output and peak oxygen uptake.Results: Multi-level random coefficient analyses revealed that FIMmotor improved during inpatient rehabilitation, but stabilized thereafter. Changes in FIMmotor were associated with peak power output. Multiple regression models showed that FIMmotor and peak power output at discharge were associated with FIMmotor one year after discharge (R-2 = 0.85), and that peak power output at discharge was associated with the social dimension of the SIP68 (R-2 = 0.18) one year after discharge.Conclusion: Functional independence improves during inpatient rehabilitation, and functional independence is positively associated with peak power output
