    Down Regulation of Plasma and Tissue Biomarkers by Homocastasterone

    Homocastasterone is a ketosteroid and a member of the brassinosteroid family of plant hormones. Earlier studies with 28-homobrasslinolide, an aldosteroid, had indicated that rat blood and tissue biochemical parameters studied were affected by this compound, resulting in altered homeostasis and cellular phosphorylation status, rendering this plant oxysterol inappropriate for high energy related work activities. The use of the ketosteroid in this study presents evidence for renormalization of elevated plasma lipid content in diabetic rat, antiglycemic potency, increase in liver glycogen and glucose level and diminished ALT and AST enzyme activities. A role for this ketosteroid in rat liver gluconeogenesis and in lipid homeostasis is suggested while the aldo and keto forms regulated glucose homeostasis in the rat. The observed differences in the effects of homobrasslinolide and homocastasterone as exogenous oxysterols on normal and diabetic rat plasma lipid level suggests the possibility of differential influence by endogenous aldo and keto oxycholesterol forms on glucose and lipid homeostasis in mammalian physiology

    Brassinosterols (BS) are ubiquitous pluripotent growth regulator present in plants. They exist in isoforms of epi and homobrassinolides (HB).  BS act as potent stimulators of root and shoot elongation, cell division, DNA and RNA polymerase activity, ethylene production and of stress tolerance to temperature, water scarcity and salinity in plants. It is also used to increase the yield of crop and to protect the plants against pesticides. Consumption of plant material as diet and used as growth regulator in animals, and application of BS in agriculture would increases its availability to the host tissues. In the present study, the effect of 28-HB, an isomer of brassinosterol on serum, liver and kidney marker enzymes, lipid peroxidation, tissue histology and the blood parameters of rat were investigated. The rats were given the compound by intradermal mode at the concentration of 75µg, 150µg and 300µg as single dose and the effects were observed after 4 hr to study the immediate response of the animal. The treatment of rats with 28-HB, caused different effects on the serum, liver and kidney parameters of this study. In conclusion, the present study showed that 28-HB affects the structure and function of rat tissues in a dose dependent manner.Keywords: Brassinosterols; homobrassinolides; 28-HB; lipid peroxidatio

    Acute effect of gibberellic acid on serum enzymes and blood markers in male albino rats

    This study was designed to evaluate the influence of a phytohormone, gibberellic acid (GA3) on marker enzymes and biomarkers of serum, and blood hemoglobin and its blood cells counts of rat. In order to evaluate the positive/negative effects, the rats were administered 75μg, 150μg and 300μg of GA3/kg body weight as a single dose. GA3 treatments produced differential effects on the different parameters at dose dependent manner after 4 hours.The down regulation in specific activities of ALT, ALP, GGT and amylase were noted against the control with significant up regulation of AST activity. GA3 also produced dose dependent effect on biomarkers. There is a substantial reduction in the quantity of glucose, urea, creatinine, calcium, phosphorus, sodium and potassium was recorded against the control. On the other hand, the insignificant increase in content of total protein, albumin and uric acid was observed at all dose of GA3 treatment against the control. GA3 increased the RBC, WBC and neutrophil by decreasing the lymphocyte total numbers. Platelets, monocytes and oesinophils count were not altered by any dose of GA3.In conclusion, GA3 produced dose dependent effect on different parameters of rat blood serum.Keywords: phytohormone; gibberellic acid; creatinine; neutrophi

    Biochemical evaluation of low dose methyl 2-benzimidazole carbamate fungicide on male albino rats

    Methyl 2-benzimidazole carbamate (carbendazim) is one of the synthetic fungicides that controlled organisms that caused plant diseases of different types. It is widely used as a preservative in leather, paint, textile, fruits and papermaking industry. It is also used as an anticancer drug in chemical medicine. In the present study low concentrations of carbendazim was administered at 5, 10, 25 and 50mM doses intradermally to male albino rats. At the end of 6 hr, 12hr and 24hr duration, blood samples were collected from the animal for the analysis of biochemical and haematological parameters. Carbendazim caused an increase of cholesterol, uric acid, glucose and creatinine while serum phosphorous content was decreased. However, mean hemoglobin, WBC, E, and platelet counts increased and total RBC, N and L counts decreased. These results indicated that low dose level carbendazim contributed to toxicological effects in the rat tissues.Keywords: Methyl 2-benzimidazole carbamate; Fungicide; Rat tissue

    HOMOCASTASTERONE: A NOVEL PLANT KETOSTEROID INDUCING HAEMATOLOGICAL CHANGES IN NORMAL AND DIABETIC MALE RAT

    Objective: To study the effect of brassinosteroid keto isoform homocastasterone, in diabetic male wistar rat as an antihyperglycemic factor and to evaluate its effects on the hemodynamic parameters in rat blood.Methods: Diabetes was induced in a group (n=6) of rats with a single peritoneal injection of streptozotocin at 60 mg/kg. bw. With a treatment schedule of 15 consecutive days, control (n=6) and diabetic rats received 666µg/kg bw, of homocastasterone. Circulating blood glucose, cell count, cell indices, and MDA level was assessed.Results: Significant reduction (p<0.05) in blood glucose level and increase inRBCs, WBCs, granulocytes, lymphocytes, monocytes and platelets count(p<0.05) along with improved functional indices for HCT, PCV, MCV, MCH, MCHC, MPV, PDW, PCT in homocastasterone treated diabetic group was noted. A significant reduction in RBC-MDA level (p<0.001) in the treated group was noted.Conclusion: It is suggested that brassinosteroid keto isoform homocastasterone exhibits antiglycemic effect in diabetic rat, and improves RBC, WBC, Platelet counts, haemoglobin level, and cell indices, while reducing peroxidative cell damage in RBCs.Â

    Prehistoric Painted Rock Shelter or Chalukyan Workshop? New Discovery of a Rock Art Site near Aihole and Its Examination in Context

    We report the discovery of a hitherto unreported rock art site in the neighbourhood of a recently uncovered single‐celled shrine at Benakanawari, near Aihole. The shrine is located deep inside a cleft of the sandstone ranges near Benakanawari, between Pattadakal and Aihole, at the foot of an escarpment. The painted rock shelter, which is roughly at a height of 25m in the cliffs above the temple, is formed by two large slabs of sandstone, collapsed and resting against a natural overhang, and contains a painted image as well as painted inscriptions in ochre and white. The inscriptions are in 8th century Kannada script as well as modern Kannada script, the former being definitely of Early Chalukyan provenance.     Evaluation and Characterization of Tamarind Gum Polysaccharide: The Biopolymer

    Polymers from natural sources are widely used as excipients in the formulation of pharmaceutical dosage forms. The objective of this study was to extract and further characterize the tamarind gum polysaccharide (TGP) obtained from Tamarindus indica as an excipient for biomedical applications. Double distilled water was used as a solvent for the extraction of gum while Ethyl alcohol was used as an antisolvent for the precipitation. The results of the Hausner ratio, Carr’s index and angle of repose were found to be 0.94, 6.25, and 0.14, respectively, which revealed that the powder is free-flowing with good flowability. The gum was investigated for purity by carrying out chemical tests for different phytochemical constituents and only carbohydrates were found to be present. The swelling index was found to be 87 ± 1%, which shows that TGP has good water intake capacity. The pH of the 1% gum solution was found to be neutral, approximately 6.70 ± 0.01. The ash values such as total ash, sulphated ash, acid insoluble ash, and water-soluble ash were found to be 14.00 ± 1.00%, 13.00 ± 0.05%, 14.04 ± 0.57% and 7.29 ± 0.06%, respectively. The IR spectra confirmed the presence of alcohol, amines, ketones, anhydrides groups. The contact angle was <90°, indicating favorable wetting and good spreading of liquid over the surface The scanning electron micrograph (SEM) revealed that the particle is spherical in shape and irregular. DSC analysis shows a sharp exothermic peak at 350 °C that shows its crystalline nature. The results of the evaluated properties showed that TGP has acceptable properties and can be used as a excipient to formulate dosage forms for biomedical applications
