Members of different cross?sections and perforations were evaluated numerically via ANSYS and GBTul, with emphasis on distortional buckling mode. The ultimate loads of the numerical analysis were compared to the Direct Strength Method (DSM) equations of ABNT NBR 14762:2010. Finally, changes were proposed in the DSM equations so it is possible calculate the predicted compressive strengths of columns with perforated cross?sections studied

    Numerical analysis of truss systems with stiffened flattened end-bars.

    Circular hollow sections are usually used in long-span roof truss systems. One of the typology for connecting elements in such structures involves the flattening of bar ends. This article presents the numerical analysis of a plane truss composed of circular hollow sections, in which diagonal bars have flattened ends. In this sense, a new flattening typology called stiffened flattening is proposed, characterized by a non-flat geometry, with the creation of stiffeners in the lateral edges of the diagonal flattened ends. The diagonal connecting system with the chord members uses connecting plates. The plates are welded to the chords and the diagonals are connected to latter through a single bolt. The numerical analysis using finite elements method was developed in two stages through ANSYS software with the Parametric Design Language (APDL), in which parameters such as geometry, materials, element types, boundary conditions and loads are specified. A non-linear analysis was performed using shell elements on the chords, diagonals, plates and welds, and contact elements between the diagonals with stiffened flattened ends and the connecting plates. Initially, a numerical study of the connecting node and the stiffened flattened end was performed, and the results directed the modeling of the plane truss. The numerical results were calibrated with the experimental truss results in full scale. The numerical result of the plane truss was also compared to a theoretical study, considering the axial load eccentricity applied in the diagonal with stiffened flattened ends. The study was based on the consideration of combined effects of axial force and bending moment provided by the Brazilian standard ABNT NBR 8800:2008. The final results indicate that the numerical model proposed is efficient and has good correlation with the experimental and theoretical results

    Consórcios de caupi e milho em cultivo orgânico para produção de grãos e espigas verdes.

    No período de outono-inverno-primavera de 2007, foi conduzido um estudo em Seropédica, Região Metropolitana do estado do Rio de Janeiro (Baixada Fluminense), com o objetivo de avaliar diferentes tipos de consórcio entre caupi (cv. Mauá) e milho (cv. AG-1051), em sistema orgânico de produção. O experimento foi instalado em área de Argissolo Vermelho-Amarelo no delineamento de blocos ao acaso, com quatro repetições. Os tratamentos constaram de diferentes épocas ou intervalos de tempo de semeadura do caupi em relação à do milho, a saber: (E1) 21 dias antes do milho; (E2) 14 dias antes do milho; (E3) 7 dias antes do milho; e (E4) no mesmo dia do milho. Tratamentos correspondentes aos cultivos solteiros do caupi e do milho foram incluídos, ambos semeados na data do tratamento E4. O cultivo consorciado com o caupi não interferiu na produtividade do milho em espigas verdes e também em termos de comprimento e diâmetro basal dessas espigas, independentemente do intervalo entre semeaduras. Com referência ao caupi, a produtividade em grãos verdes no cultivo solteiro foi superior à dos consórcios com o milho. Os valores obtidos para os Índices de Equivalência de Área (IEA), foram todos acima de 1,0, indicando que os consórcios foram eficientes quanto ao desempenho agronômico/biológico. Considerando, ainda a produtividade de cada cultura participante do consórcio, a semeadura do caupi antecipada de 21 dias em relação à do milho afigura-se mais adequada ao manejo orgânico adotado e às condições edafoclimáticas da região

    Detection and identification of Xanthomonas pathotypes associated with citrus diseases using comparative genomics and multiplex PCR.

    Background. In Citrus cultures, three species of Xanthomonas are known to cause distinct diseases. X. citri subsp. citri patothype A, X. fuscans subsp. aurantifolii pathotypes B and C, and X. alfalfae subsp. citrumelonis, are the causative agents of cancrosis A, B, C, and citrus bacterial spots, respectively. Although these species exhibit different levels of virulence and aggressiveness, only limited alternatives are currently available for proper and early detection of these diseases in the fields. The present study aimed to develop a new molecular diagnostic method based on genomic sequences derived from the four species of Xanthomonas. Results. Using comparative genomics approaches, primers were synthesized for the identification of the four causative agents of citrus diseases. These primers were validated for their specificity to their target DNA by both conventional and multiplex PCR. Upon evaluation, their sensitivity was found to be 0.02 ng/?l in vitro and 1.5 ? 104 CFU ml?1 in infected leaves. Additionally, none of the primers were able to generate amplicons in 19 other genomes of Xanthomonas not associated with Citrus and one species of Xylella, the causal agent of citrus variegated chlorosis (CVC). This denotes strong specificity of the primers for the different species of Xanthomonas investigated in this study. Conclusions. We demonstrated that these markers can be used as potential candidates for performing in vivo molecular diagnosis exclusively for citrus-associated Xanthomonas. The bioinformatics pipeline developed in this study to design specific genomic regions is capable of generating specific primers. It is freely available and can be utilized for any other model organism

    The inventory of geological heritage of the state of São Paulo, Brazil: Methodological basis, results and perspectives

    An inventory of geological sites based on solid and clear criteria is a first step for any geoconservation strategy. This paper describes the method used in the geoheritage inventory of the State of São Paulo, Brazil, and presents its main results. This inventory developed by the geoscientific community aimed to identify geosites with scientific value in the whole state, using a systematic approach. All 142 geosites representative of 11 geological frameworks were characterised and quantitatively evaluated according to their scientific value and risk of degradation, in order to establish priorities for their future management. An online database of the inventory is under construction, which will be available to be easily consulted and updated by the geoscientific community.     The Mutational Spectrum in a Cohort of Charcot-Marie-Tooth Disease Type 2 among the Han Chinese in Taiwan

    BACKGROUND: Charcot-Marie-Tooth disease type 2 (CMT2) is a clinically and genetically heterogeneous group of inherited axonal neuropathies. The aim of this study was to extensively investigate the mutational spectrum of CMT2 in a cohort of patients of Han Chinese. METHODOLOGY AND PRINCIPAL FINDINGS: Genomic DNA from 36 unrelated Taiwanese CMT2 patients of Han Chinese descent was screened for mutations in the coding regions of the MFN2, RAB7, TRPV4, GARS, NEFL, HSPB1, MPZ, GDAP1, HSPB8, DNM2, AARS and YARS genes. Ten disparate mutations were identified in 14 patients (38.9% of the cohort), including p.N71Y in AARS (2.8%), p.T164A in HSPB1 (2.8%), and p.[H256R]+[R282H] in GDAP1 (2.8%) in one patient each, three NEFL mutations in six patients (16.7%) and four MFN2 mutations in five patients (13.9%). The following six mutations were novel: the individual AARS, HSPB1 and GDAP1 mutations and c.475-1G>T, p.L233V and p.E744M mutations in MFN2. An in vitro splicing assay revealed that the MFN2 c.475-1G>T mutation causes a 4 amino acid deletion (p.T159_Q162del). Despite an extensive survey, the genetic causes of CMT2 remained elusive in the remaining 22 CMT2 patients (61.1%). CONCLUSIONS AND SIGNIFICANCE: This study illustrates the spectrum of CMT2 mutations in a Taiwanese CMT2 cohort and expands the number of CMT2-associated mutations. The relevance of the AARS and HSPB1 mutations in the pathogenesis of CMT2 is further highlighted. Moreover, the frequency of the NEFL mutations in this study cohort was unexpectedly high. Genetic testing for NEFL and MFN2 mutations should, therefore, be the first step in the molecular diagnosis of CMT2 in ethnic Chinese

    Background: Paracoccidioides brasiliensis causes paracoccidioidomycosis, one of the most prevalent systemic mycosis in Latin America. Thus, understanding the characteristics of the protective immune response to P. brasiliensis is of interest, as it may reveal targets for disease control. The initiation of the immune response relies on the activation of pattern recognition receptors, among which are TLRs. Both TLR2 and TLR4 have been implicated in the recognition of P. brasiliensis and regulation of the immune response. However, the role of TLR9 during the infection by this fungus remains unclear.J.F.
