Análise dos receptores de acetilcolina (nAChRs) na musculatura esquelética de ratos com insuficiência cardÃaca submetidos ao treinamento aeróbico
A insuficiência cardÃaca (IC) é uma sÃndrome iniciada por uma redução na função cardÃaca e caracterizada pela ativação de mecanismos compensatórios e atrofia. Fadiga e dispnéia são os sintomas mais comuns desta doença, e estão associados à miopatia do músculo esquelético; ocorre redução da capacidade oxidativa, mudança nas fibras do tipo I para tipo II, alteração na expressão dos fatores reguladores miogênicos (MRFs) e na JNM. O treinamento fÃsico aeróbico é considerado uma conduta amplamente aceita para minimizar as consequências dos sintomas causados pela IC, com melhora na qualidade de vida e pode reverter anormalidades do músculo cardÃaco e esquelético, além de modular a junção neuromuscular. O músculo diafragma é o mais importante músculo inspiratório dos mamÃferos sendo um dos mais acometidos na IC; o treinamento fÃsico aeróbico altera as propriedades eletrofisiológicas do diafragma durante a fadiga e favorece a resistência muscular. A hipótese do presente estudo é que as alterações fenotÃpicas musculares que ocorrem na IC estão associadas com alterações na distribuição e expressão dos nAChRs e que o treinamento fÃsico aeróbico atenue estas alterações. O objetivo deste trabalho foi avaliar a distribuição e expressão dos nAChRs em ratos com IC submetidos ao treinamento aeróbico. Foram utilizados 44 ratos Wistar machos (90 a 100g), divididos em quatro grupos experimentais: controle (C), treinamento aeróbico (TR), estenose aórtica (EAo) e estenose aórtica com treinamento aeróbico (EAoTR). A estenose aórtica foi induzida pela inserção de um clip de prata de 0.6 mm de diâmetro na aorta ascendente. Os demais animais foram divididos aleatoriamente em...Heart failure (HF) is a syndrome initiated by a reduction in cardiac function and characterized by the activation of compensatory mechanisms and muscle myopathy. Fatigue and dyspnea are the most common symptoms of this disease, associated with the reduction of the oxidative capacity, changes in the fibers type I to type II, alterations in the expression of myogenic regulatory factors (MRFs) and in the neuromuscular junction (NMJs). Aerobic training is considered a widely accepted practice to minimize the consequences of the symptoms caused by HF, improving the life quality and can reverse abnormalities of cardiac and skeletal muscle, and modulate the NMJ. The diaphragm is the most important inspiratory muscle in mammals and is one of the most affected in HF; aerobic training changes the electrophysiological properties of the diaphragm during fatigue and promotes muscle endurance. Thus, the hypothesis of this study is that the changes that occur in HF are associated with alterations in the NMJs, in the expression of nAChRs and aerobic training can attenuate these changes. The aim of this study was to evaluate the distribution and the expression of nAChRs subunits in rats with HF submitted to an aerobic training. Expressão gênica dos receptores nicotÃnicos de acetilcolina (nAChR) e caracterização morfológica das junções neuromusculares em músculo esquelético de ratos com insuficiência cardÃaca
A insuficiência cardÃaca (IC) está associada à miopatia dos músculos esqueléticos com aumento da expressão das isoformas rápidas da cadeia pesada de miosina e alterações na matriz extracelular. Os fatores de regulação miogência(MRF),como a MyoD e MRF4, pertecem a uma famÃlia de fatores transcricionais que controlam vários genes músculo-especÃficos.Esses fatores forma heterodÃmeros com proteÃnas HLH e ligam-se a seqüências de DNA conhecidas como Ebox, presentes na região promotora de vários genes músculo–especÃficos, incluindo todos aqueles que codificam as subunidades dos receptores nicotÃnicos de acetilcolina (nAChR) da junção neuromuscular (JNM). Baseado no fato de que na IC há uma diminuição na expressão da MyoD e MRF4 no músculo sóleo de ratos com IC induzida por monocrotalina, o objetivo do presente trabalho é verificar possÃveis alterações na expressão dos receptores de acetilcolina, bem como realizar uma análise morfológica e morfométrica das JNMs. Neste estudo foram utilizados ratos Wistar, machos, divididos em grupos controle e experimental e a Insuficiência cardÃaca (IC) foi induzida pela injeção de monocrotalina intra-peritoneal.Quando os sintomas de IC foram visualizados (após 22 dias) os animais foram sacrificados pentobarbital sódico i.p. (50 mg/Kg). A seguir foram mensurados o peso corporal (PC) dos ratos, bem como outros parâmetros. O músculo sóleo de ambos os antÃmeros foram dissecados e preparados para: avaliação da expressão gênica das subunidades ε, γ e α dos receptores de acetilcolina por PCR em Tempo Real; para análise morfológica e morfométrica da Junção Neuromuscular através da técnica de Esterase inespecÃfica; para análise ultra-estrutural da JN. Pediculose na escola - uma oportunidade para aprender e ensinar
O piolho de cabeça atualmente é um problema mundial e não está relacionado à pobreza, falta de higiene, desleixo. Sua prevalência é alta na idade escolar de três a doze anos de idade. Quando uma criança o adquire tem um sentimento de vergonha, deixa de ir à escola e raramente é encaminhada para cuidados médicos. Geralmente para o controle, as famÃlias utilizam substâncias tóxicas, inseticidas domiciliares, produtos inflamáveis e ervas na cabeça das crianças. E ainda, acreditam que o piolho pode ser transmissor de doenças. Há uma falta de informação e neste caso, a educação se faz necessária. O objetivo deste trabalho foi elaborar um instrumento de avaliação de professores das séries iniciais do ensino fundamental I, da rede pública de ensino, da cidade de Botucatu – SP, relacionado à pediculose. Esta análise ocorreu através de um questionário constituÃdo de 10 perguntas abertas e 14 fechadas (concordo e discordo) com abordagem educacional (ensino sobre piolho - conhecimento, atitude e prática) e com uma abordagem didática sobre biologia, diagnóstico, prevenção, tratamento, transmissão sobre o piolho de cabeça. Este trabalho foi realizado em 9 escolas da rede pública do ensino fundamental I com 108 professores.Todos os dados foram tabulados no programa Microsoft Excel 2000 versões 7.0 e as análises foram realizadas com base na estatÃstica descritiva, não – paramétrica, segundo o método estatÃstico que consiste em técnica de análise de variância não paramétrica para o modelo com um fator – Teste de Kruskal-Wallis e por tabelas de freqüência. Effects of Nandrolone Decanoate on Skeletal Muscle and Neuromuscular Junction of Sedentary and Exercised Rats
Background and Objectives: Nandrolone decanoate (ND) is the most widely used among the anabolic androgenic steroids (AAS), synthetic substances derived from testosterone, to improve muscular and health gains associated with exercises. The AAS leads to physical performance enhancement and presents anti-aging properties, but its abuse is associated with several adverse effects. Supraphysiological doses of AAS with or without physical exercise can cause morphological and functional alterations in neuromuscular interactions. This study aims to investigate the effects of ND supraphysiological doses in neuromuscular interactions, focusing on the soleus muscle and its neuromuscular junctions (NMJs) in rats, associated or not with physical exercise. Materials and Methods: Forty male Sprague Dawley rats were divided into four groups: sedentary and exercised groups, with or without ND at the dose of 10 mg/kg/week. The animals were treated for eight weeks, with intramuscular injections, and the soleus muscle was collected for morphological analyses. Results: The supraphysiological doses of ND in the sedentary group caused muscle degeneration, evidenced by splitting fibers, clusters of small fibers, irregular myofibrils, altered sarcomeres, an increase in collagen deposition and in the number of type I muscle fibers (slow-twitch) and central nuclei, as well as a decrease in fibers with peripheral nuclei. On the other hand, in the ND exercise group, there was an increase in the NMJs diameter with scattering of its acetylcholine receptors, although no major morphological changes were found in the skeletal muscle. Thus, the alterations caused by ND in sedentary rats were partially reversed by physical exercise. Conclusions: The supraphysiological ND exposure in the sedentary rats promoted an increase in muscle oxidative pattern and adverse morphological alterations in skeletal muscle, resulting from damage or post-injury regeneration. In the ND-exercised rats, no major morphological changes were found. Thus, the physical exercise partially reversed the alterations caused by ND in sedentary rats
Aerobic training attenuates nicotinic acetylcholine receptor changes in the diaphragm muscle during heart failure
Introduction: Heart failure (HF) is a
progressive myopathy, with clinical signs of fatigue and
limb weakness that can damage the nerve-muscle
interaction, altering synaptic transmission and nicotinic
acetylcholine receptors (nAChR) in neuromuscular
junctions (NMJs). The diaphragm is composed of a
mixed proportion of muscle fibres, and during HF, this
muscle becomes slower and can alter its function. As
exercise training is an accepted practice to minimise
abnormalities of skeletal muscle during HF, in this study,
we evaluated the hypothesis that aerobic training
attenuates alterations in the expression of nAChR
subunits in NMJs diaphragm during heart failure.
Objective: The aim of this study was to evaluate the
distribution and expression of nAChR subunits in the
diaphragm muscle fibres of rats subjected to an aerobic
training programme during HF. Methods: Control
(Sham), control training (ShamTR), aortic stenosis (AS)
and aortic stenosis training (ASTR) groups were
evaluated. The expression of nAChR subunits (γ, α1, ε,
β1 and δ) was determined by qRT-PCR, and NMJs were
analysed using confocal microscopy. Results: We
observed increased expression of the γ, α1 and β1
subunits in the AS group compared with the ASTR
group. The distribution of NMJs was modulated in these
groups. Discussion: HF alters the mRNA expression of
nAChR subunits and the structural characteristics of
diaphragm NMJs. In addition, aerobic training did notalter NMJs morphology but attenuated the alterations in
heart structure and function and in nAChR subunit
mRNA expression. Our findings demonstrate the
beneficial effects of aerobic exercise training in
maintaining the integrity of the neuromuscular system in
the diaphragm muscle during HF and may be critical for
non-pharmacological therapy to improve the quality of
life for patients with this syndrome
Regulation of cardiac microRNAs induced by aerobic exercise training during heart failure
Exercise training (ET) has beneficial effects on the myocardium in heart failure (HF) patients and in animal models of induced cardiac hypertrophy and failure. We hypothesized that if microRNAs (miRNAs) respond to changes following cardiac stress, then myocardial profiling of these miRNAs may reveal cardio-protective mechanisms of aerobic ET in HF. We employed ascending aortic stenosis (AS) inducing HF in Wistar rats. Controls were sham-operated animals. At 18 weeks after surgery, rats with cardiac dysfunction were randomized to 10 weeks of aerobic ET (HF-ET) or to a heart failure sedentary group (HF-S). ET attenuated cardiac remodeling, as well as clinical and pathological signs of HF with maintenance of systolic and diastolic function when compared to HF-S. Global miRNA expression profiling of the cardiac tissue revealed 56 miRNAs differentially regulated in animals in the HF-ET, but only 12 miRNAs were differentially regulated in the HF-S. Out of 23 miRNAs that were differentially regulated in both groups, 17 miRNAs exhibited particularly high increases in expression, including miR-598, miR-429, miR-224, miR-425, and miR-221. From the initial set of deregulated miRNAs, 14 miRNAs with validated targets expressed in cardiac tissue that respond robustly to ET in HF were used to construct a miRNA-mRNA regulatory networks that revealed a set of 203 miRNA-target genes involved in programmed cell death, TGF-β signaling, cellular metabolic processes, cytokine signaling, and cell morphogenesis.
