    Comparison of self-reported and accelerometer-assessed measurements of physical activity according to socio-demographic characteristics in Korean adults

    OBJECTIVES Previous studies have shown relatively low correlations between self-reported and accelerometer-assessed physical activity (PA). However, this association differs by socio-demographic factors, and this relationship has not been fully investigated in the general population. Thus, we investigated the correlation between self-reported and accelerometer-assessed PA and whether it differed by demographic and socioeconomic factors among the Korean general population. METHODS This cross-sectional study included 623 participants (203 men and 420 women) aged 30 to 64 years, who completed a PA questionnaire and wore a wrist-worn accelerometer on the non-dominant wrist for 7 days. We examined the agreement for metabolic equivalent task minutes per week (MET-min/wk) between the 2 measures and calculated Spearman correlation coefficients according to demographic and socioeconomic factors. RESULTS The kappa coefficient between tertiles of self-reported and accelerometer-assessed total MET-min/wk was 0.16 in the total population, suggesting overall poor agreement. The correlation coefficient between the 2 measurements was 0.26 (p<0.001) in the total population, and the correlation tended to decrease with increasing age (p for trend <0.001) and depression scores (p for trend <0.001). CONCLUSIONS We found a low correlation between self-reported and accelerometer-assessed PA among healthy Korean adults, and the correlation decreased with age and depression score. When studying PA using accelerometers and/or questionnaires, age and depression need to be considered, as should differences between self-reported and accelerometer-assessed PA

    Characterization of the complete mitochondrial genome of the false trevally, Lactarius lactarius (Perciformes: Lactariidae)

    The complete mitochondrion DNA sequence of Lactarius lactarius was determined by next generation sequencing (NGS) platform as the first mitogenome report in the family. The mitogenome of L. lactarius wa 16,642ā€‰bp in length that contains 13 gene-encoded proteins, 2 ribosomal RNA gene (12S and 16S), 22 tRNA gene, and a control region (D-loop). Besides COX1 gene (GTG), all the other protein-coding genes showed typical (ATG) start codon. Incomplete stop condons (TA-/Tā€“) were identified in COX2, COX3, ND2, ND3, ND4 and Cyt B. A phylogenetic analysis with currently reported mitogenomes of its relative species, L. lactarius formed a Lactariidae clade distinct from other families. Two species, Pentaceros japonicus (AB739063) and Banjos banjos (KT345965) were among the most closely related species with 86.61% and 86.33% sequence identity, respectively

    Although spatiotemporal analysis of the cephalopod larvae provides the useful information for the effective management of their resources, it has been difficult mainly due to their low numbers in the mixed zooplankton net samples and difficulty in morphological identification. In order to analyze the planktonic cephalopods using next-generation sequencing (NGS), we have designed a cephalopod-specific universal (CPD) primer set targeting a region covering mitochondrial cytochrome b and ND6 genes based on the currently identified 36 complete cephalopod mitochondrial genome sequences in the GenBank database. The expected amplicon sizes by CPD primers were between 465 and 471 bp, which was applicable to the MiSeq system (Illumina, San Diego, CA, USA). NGS results of pooled DNAs from 8 months (including 739 zooplankton net samples) collected from Korean waters in 2016 showed the exclusive cephalopod sequences with little contaminant sequences supporting the specificity of CPD primer set. Total 47 representative cephalopod haplotypes (seven families and 10 genera) were obtained from 1,439,414 merged reads. Among the total analyzed haplotypes, Watasenia scintillans, Todarodes pacificus, and Sepiola birostrata were the most abundant species in Korean waters. Two ā€œunidentifiedā€ clades in order Oegopsida were identified, which was showed less than 90% sequence identity but closely related to Enoploteuthidae and Idiosepiidae, respectively. Monthly changes in proportions of each haplotype were also identified, which may reflect its reproduction and spawning period. The larvae of W. scintillans was dominant from February to June, while high proportions of other cephalopod taxa were also identified from August to November. Only single haplotype was dominant in W. scintillans (Type 2) throughout the year, while two distinct haplotypes showed seasonal differences in T. pacificus

    The first mitochondrial genome of Ophiocara porocephala was determined by the combination of next-generation sequencing (NGS) and Sanger sequencing methods. A complete circular mitogenome of O. porocephala (16,529ā€‰bp) consisted of 13 protein-coding genes, 22 transfer RNAs, two ribosomal RNAs, and two non-coding regions, including a control region (D-loop) and a light strand origin of replication (OL). Two start codons (ATG and GTG) and four stop codons (TAG, TAA, TAā€“, and Tā€“) were used in all the PCGs. Except for ND6 and eight transfer RNAs (tRNAs), all the other genes were encoded in the heavy strand. Based on phylogenetic analysis, O. porocephala formed a clade with three other species in the subfamily Butinae, while the other 10 made a subfamily Eleotrinae clade

    A Detection of Convectively Induced Turbulence Using in Situ Aircraft and Radar Spectral Width Data

    A commercial aircraft, departing from Seoul to Jeju Island in South Korea, encountered a convectively induced turbulence (CIT) at about z = 2.2 km near Seoul on 28 October 2018. At this time, the observed radar reflectivity showed that the convective band with cloud tops of z = 6ā€“7 km passed the CIT region with high values of spectral width (SW; larger than 4 m sā€“1). Using the 1 Hz wind data recorded by the aircraft, we estimated an objective intensity of the CIT as a cube root of eddy dissipation rate (EDR) based on the inertial range technique, which was about 0.33ā€“0.37 m2/3 sāˆ’1. Radar-based EDR was also derived by lognormal mapping technique (LMT), showing that the EDR was about 0.3ā€“0.35 m2/3 sāˆ’1 near the CIT location, which is consistent with in situ EDR. In addition, a feasibility of the CIT forecast was tested using the weather and research forecast (WRF) model with a 3 km horizontal grid spacing. The model accurately reproduced the convective band passing the CIT event with an hour delay, which allows the use of two methods to calculate EDR: The first is using both the sub-grid and resolved turbulent kinetic energy to infer the EDR; the second is using the LMT for converting absolute vertical velocity (and its combination with the Richardson number) to EDR-scale. As a result, we found that the model-based EDRs were about 0.3ā€“0.4 m2/3 sāˆ’1 near the CIT event, which is consistent with the estimated EDRs from both aircraft and radar observations

    The complete mitochondrial genome sequence of the Icefish, Chionobathyscus dewitti was determined by the Next Generation Sequencing (NGS) analysis. The complete mitogenome was 17,452ā€‰bp in length, which encoded the canonical 13 protein-coding genes, 22 tRNAs, two rRNAs, and two non-coding regions. As shown in the other notothenids, translocation of ND6 and an additional nonā€“coding region were identified, which is different from the typical vertebrate mitochondrial genomes. The C. dewitti was clustered distinctly from the those in the Chinodraco and Chaenocephalus, which supported the idea that this species should be classified in the different genus, Chionobathyscus in the family Channichthyidae
