    Ku70 alleviates neurodegeneration in drosophila models of Huntington's disease

    DNA damage accumulates in genome DNA during the long life of neurons, thus DNA damage repair is indispensable to keep normal functions of neurons. We previously reported that Ku70, a critical molecule for DNA double strand break (DSB) repair, is involved in the pathology of Huntington's disease (HD). Mutant huntingtin (Htt) impaired Ku70 function via direct interaction, and Ku70 supplementation recovered phenotypes of a mouse HD model. In this study, we generate multiple Drosophila HD models that express mutant huntingtin (Htt) in eye or motor neuron by different drivers and show various phenotypes. In such fly models, Ku70 co-expression recovers lifespan, locomotive activity and eye degeneration. In contrast, Ku70 reduction by heterozygous null mutation or siRNA-mediated knock down accelerates lifespan shortening and locomotion disability. These results collectively support that Ku70 is a critical mediator of the HD pathology and a candidate therapeutic target in HD

    Production of interleukin-10 by alveolar macrophages from lung cancer patients

    AbstractInterleukin (IL)-10 is known to be an autoregulatory factor of functions of monocyte macrophages. The purpose of this study was to determine whether IL-10 production by alveolar macrophages (AMs) is altered in patients with lung cancer. AMs were obtained by bronchoalveolar lavage from 25 patients with lung cancer and 14 control patients. The production of IL-10 by AMs was quantitated by enzyme immunoassay with or without stimulation with lipopolysaccharide (LPS). No significant difference in spontaneous and LPS-stimulated IL-10 production by AMs was observed between lung cancer patients and control patients (mean ± sem; 288·0 ± 56·7 vs. 249·6 ± 58·4 pg ml−1). IL-10 production of LPS-stimulated AMs was not impaired even in lung cancer patients with systemic metastasis. IL-4 failed to suppress LPS-induced production of IL-10 by AMs both in control patients and in lung cancer patients. In eight patients with lung cancer, IL-10 production by AMs was estimated before and after systemic chemotherapy and IL-10 production by LPS-stimulated AMs tended to increase after systemic chemotherapy from 152·3 ± 51·9 to 278·0 ± 112·8 pg ml−1. As IL-10 is a potent inhibitor of tumour angiogenesis, an important process of tumour progression, these results suggest that, even in advanced cancer patients, macrophages can produce potent angiogenesis inhibitor and systemic chemotherapy may augment this inhibitory activity in the lung

    The aims of this research is to produce inulinfructotransferase enzyme with potentially benefit of bacteria Actinomycetes which was isolated from Indonesia soils to produce Difructoanhydride-Ill (DFA III), a compound has physiology activities to enhance calsium absorption of rats. cows and human colons so that the materials can be applied as antiosteoporosis. The processing of inulinfructotransferase enzyme from Nonomuraea sp by usingshakeflask level and study the influence of carbohydrate source were conducted. This enzyme has optimum temperature at 65rtC,higher than optimum temperature which has found before and the activities was stable at 70dc for 20 minutes. So that the enzyme has potentially applicable for industry scale. The results indicated that optimation condition for processing inulinfructotransferase enzyme of Nonomuraea sp : temperature sore. incubation time 48 hours.carbon source of inulin 20 g/L, nitrogen source 5g/L, noyeast extract added, the enzyme activities was 7,3 Unit/ml; However,when medium contains yeast extract 0,2 gIL,the inulinfructotransferase enzyme activity was 14,6 Unit/mL

    The TAO-Gen Algorithm for Identifying Gene Interaction Networks with Application to SOS Repair in E. coli

    One major unresolved issue in the analysis of gene expression data is the identification and quantification of gene regulatory networks. Several methods have been proposed for identifying gene regulatory networks, but these methods predominantly focus on the use of multiple pairwise comparisons to identify the network structure. In this article, we describe a method for analyzing gene expression data to determine a regulatory structure consistent with an observed set of expression profiles. Unlike other methods this method goes beyond pairwise evaluations by using likelihood-based statistical methods to obtain the network that is most consistent with the complete data set. The proposed algorithm performs accurately for moderate-sized networks with most errors being minor additions of linkages. However, the analysis also indicates that sample sizes may need to be increased to uniquely identify even moderate-sized networks. The method is used to evaluate interactions between genes in the SOS signaling pathway in Escherichia coli using gene expression data where each gene in the network is over-expressed using plasmids inserts

    The Boundary Convergence of the Steady Zero-Temperature-Driven Hard Spheres

    We study the fundamental problem of two gas species in two dimensional velocity space whose molecules collide as hard circles in the presence of a flat boundary and with dependence on only one space dimension. The case of three-dimensional velocity space is a generalization. More speciffically the linear problem arising when the second gas dominates as a flow with constant velocity (and hence zero temperature) is considered. The boundary condition adopted consists of prescribing the outgoing velocity distribution at the wall. It is discovered that the presence of the boundary under general assumptions on the outgoing distribution ensures the convergence of a series of path integrals and thus a convenient representation for the solution is obtained.Comment: 13 pages, 2 figure
