    Comparative Evaluation of Biomarkers of Inflammation Among Indian Women With Polycystic Ovary Syndrome (PCOS) Consuming Vegetarian vs. Non-vegetarian Diet

    Background: Sub-inflammation and insulin resistance characterize women with PCOS. Data on dietary modulation of inflammation among PCOS women is scant, particularly from Indian subcontinent. The present study aimed to assess the effect of plant based vs. animal origin diets on serum markers of inflammation (primary outcome measure).Methods: This observational case-control study compared age and BMI matched PCOS and apparently healthy women from two populations following different dietary practices. The vegetarian women from New-Delhi (n = 82 PCOS and n = 179 healthy) and non-vegetarian women from Srinagar (n = 62 PCOS and n = 141 healthy) formed the groups. Using a uniform methodology, detailed clinical, biochemical, hormonal, and inflammatory marker assessment was undertaken.Results: The mean age of the overall cohort was 26.23 Ā± 4.59 years with a mean BMI of 24.39 Ā± 3.72 kg/m2. Overall pro-inflammatory markers (TNF-Ī±, IL-6, IL-1Ī², hs-CRP and serum resistin) were significantly higher (p ā‰¤ 0.05) and anti-inflammatory markers (IL-10 and adiponectin) were lower among women with PCOS than healthy subjects. On comparing vegetarian women with non-vegetarians, higher daily calorie intake (1895.46 Ā± 258.19 vs. 1860.13 Ā± 323.96 Kcal) with a higher protein and fat and lower carbohydrate intake was recorded in the latter, although the percent energy derived from carbohydrates was higher among vegetarians. Clinical and biochemical parameters were comparable among the groups except mFG score, total serum testosterone and serum lipid levels which were higher among non-vegetarian women as compared to their vegetarian counterparts from both categories (PCOS and healthy). Interestingly, vegetarian women with PCOS and healthy women had higher serum pro-inflammatory and lower anti-inflammatory markers compared to their non-vegetarian counterparts.Conclusion: Women with PCOS consuming Indian vegetarian diet have higher pro-inflammatory and lower anti-inflammatory marker levels than their age and BMI matched healthy non-vegetarian counterparts. This interesting observation can be attributed to the dietary composition, among other factors and needs confirmation from well-designed randomized studies on a larger cohort

    Enhanced production of Ī²- glucosidase by locally isolated fungal strain employing submerged fermentation

    Ī²-glucosidase has wide spectrum of biotechnological applications in different industries including food, textile, laundry detergents, pulp and paper, pharmaceutical and biofuel industry. The present investigation related to isolation, screening, and process optimization of fungal strain for enhanced production of Ī²-glucosidase (BGL). For this purpose, different fungal stains were isolated from different sources including soil, fruits, bark of tree as well as from the compost. The screening of fungal strain for BGL production was carried out via submerged fermentation. All the tested strains were identified on the basis of micro and macroscopic features. The fungal strain having greater ability for BGL synthesis among tested ones was identified as Aspergillus niger and given the code SBT-15. The process parameter including fermentation media, temperature, pH, rate of fermentation, carbon and nitrogen sources, volume of media were optimized. Five different fermentation media were evaluated M3 medium gave maximum production. The optimal conditions for BGL production was 72 hours of incubation at 40Ā°C, pH 6 and 50 ml fermentation medium. Glucose (1%) and ammonium sulphate (3%) were optimized as best carbon and nitrogen sources, respectively

    A novel pH-responsive hydrogel system based on Prunus armeniaca gum and acrylic acid: Preparation and evaluation as a potential candidate for controlled drug delivery

    pH-responsive hydrogels have become effective and attractive materials for the controlled release of drugs at pre-determined destinations. In the present study, a novel hydrogel system based on Prunus armeniaca gum (PAG) and acrylic acid (AA) was prepared by a free radical mechanism using N, N-methylene bisacrylamide (MBA) as cross-linker and potassium persulfate (KPS) as initiator. A series of hydrogels varying PAG, AA, and MBA concentration was developed to determine the impact of these components. Formulated hydrogels were characterized for pH-responsive swelling, drug release, gel content, and porosity. Structural analysis was performed by FTIR, XRD, and SEM analysis. TGA study was applied to assess thermal stability. Oral acute toxicity and in vivo drug release were performed in rabbits. Hydrogels exhibited pH-dependent swelling and drug release. Swelling, drug loading and release, and porosity increased by increasing PAG and AA concentration while decreased by increasing MBA. The gel content of formulations was increased by increasing all three components. FTIR studies confirmed the development of copolymeric networks and the loading of drug. XRD studies revealed that hydrogels were amorphous, and the crystalline drug was changed into an amorphous form during loading. TGA results indicated that hydrogels were stable up to 600 Ā°C. Acute oral toxicity results confirm that hydrogels were nontoxic up to a dose of 2 g/kg body weight in rabbits. The pharmacokinetic evaluation revealed that hydrogels prolonged the availability of the drug and the peak plasma concentration of the drug was obtained in 6 h as compared to the oral solution of the drug. Tramadol hydrochloride (THC) was used as a model drug. Hence, pH-responsive swelling and release, nontoxic nature and improved pharmacokinetics support that PAG-based hydrogels may be considered as potential controlled-release polymeric carriers

    Studying the Impact of Initialization for Population-Based Algorithms with Low-Discrepancy Sequences

    To solve different kinds of optimization challenges, meta-heuristic algorithms have been extensively used. Population initialization plays a prominent role in meta-heuristic algorithms for the problem of optimization. These algorithms can affect convergence to identify a robust optimum solution. To investigate the effectiveness of diversity, many scholars have a focus on the reliability and quality of meta-heuristic algorithms for enhancement. To initialize the population in the search space, this dissertation proposes three new low discrepancy sequences for population initialization instead of uniform distribution called the WELL sequence, Knuth sequence, and Torus sequence. This paper also introduces a detailed survey of the different initialization methods of PSO and DE based on quasi-random sequence families such as the Sobol sequence, Halton sequence, and uniform random distribution. For well-known benchmark test problems and learning of artificial neural network, the proposed methods for PSO (TO-PSO, KN-PSO, and WE-PSO), BA (BA-TO, BA-WE, and BA-KN), and DE (DE-TO, DE-WE, and DE-KN) have been evaluated. The synthesis of our strategies demonstrates promising success over uniform random numbers using low discrepancy sequences. The experimental findings indicate that the initialization based on low discrepancy sequences is exceptionally stronger than the uniform random number. Furthermore, our work outlines the profound effects on convergence and heterogeneity of the proposed methodology. It is expected that a comparative simulation survey of the low discrepancy sequence would be beneficial for the investigator to analyze the meta-heuristic algorithms in detail

    The Pakistan risk of myocardial infarction study: A resource for the study of genetic, lifestyle and other determinants of myocardial infarction in south Asia

    The burden of coronary heart disease (CHD) is increasing at a greater rate in South Asia than in any other region globally, but there is little direct evidence about its determinants. The Pakistan Risk of Myocardial Infarction Study (PROMIS) is an epidemiological resource to enable reliable study of genetic, lifestyle and other determinants of CHD in South Asia. By March 2009, PROMIS had recruited over 5,000 cases of first-ever confirmed acute myocardial infarction (MI) and over 5,000 matched controls aged 30-80 years. For each participant, information has been recorded on demographic factors, lifestyle, medical and family history, anthropometry, and a 12-lead electrocardiogram. A range of biological samples has been collected and stored, including DNA, plasma, serum and whole blood. During its next stage, the study aims to expand recruitment to achieve a total of about 20,000 cases and about 20,000 controls, and, in subsets of participants, to enrich the resource by collection of monocytes, establishment of lymphoblastoid cell lines, and by resurveying participants. Measurements in progress include profiling of candidate biochemical factors, assay of 45,000 variants in 2,100 candidate genes, and a genomewide association scan of over 650,000 genetic markers. We have established a large epidemiological resource for CHD in South Asia. In parallel with its further expansion and enrichment, the PROMIS resource will be systematically harvested to help identify and evaluate genetic and other determinants of MI in South Asia. Findings from this study should advance scientific understanding and inform regionally appropriate disease prevention and control strategies

    Investigating the Impact of Derivatives on Stability in the Financial Performance of the Banking Sector of Pakistan

    Purpose: The aim of the paper is twofold, to contribute to the ongoing debate regarding highlighting the determinants of derivative usage by commercial banks specifically from a dual banking economy perspective, and to discuss the influence of derivatives usage on the value and stability in the financial performance of the banking sector of Pakistan. Design/Methodology/Approach: The study used the Z-Score and CAMELS Index to compare the stability analysis from different dimensions. Further, the impact of bank-controlled variables on the use of derivatives has also been analyzed. Findings: By conducting Panel regression models, the results revealed that derivatives used by banks reduce the profitability and ultimately stability of banking and increase the bank-specific risk. Implications/Originality/Value: These findings point towards serious intentions regarding using derivative instruments in an economy where dual banking is practiced

    Qualitative and semi-quantitative analysis of health-care pharmaceutical products using laser-induced breakdown spectroscopy

    Laser-induced breakdown spectroscopy (LIBS) is a sensitive optical technique that is capable of rapid multi-elemental analysis. The development of this technique for elemental analysis of pharmaceutical products may eventually revolutionize the field of human health. Under normal circumstances, the elemental analysis of pharmaceutical products based on chemical methods is time-consuming and complicated. In this investigation, the principal aim is to develop an LIBS-based methodology for elemental analysis of pharmaceutical products. This LIBS technique was utilized for qualitative as well as quantitative analysis of the elements present in Ca-based tablets. All the elements present in the tablets were detected and their percentage compositions were verified in a single shot, using the proposed instrument. These elements (e.g., Ca, Mg, Fe, Zn, and others) were identified by the wavelengths of their spectral lines, which were verified using the NIST database. The approximate amount of each element was determined based on their observed peaks and the result was in exact agreement with the content specification. The determination of the composition of prescription drug for patients is highly important in numerous circumstances. For example, the exploitation of LIBS may facilitate elemental decomposition of medicines to determine the accuracy of the stated composition information. Moreover, the approach can provide element-specific, meaningful, and accurate information related to pharmaceutical products.     Association of polycystic ovary syndrome and Gravesā€² disease: Is autoimmunity the link between the two diseases

    Introduction: Polycystic ovary syndrome (PCOS) is a common endocrinopathy of women of child-bearing age. Although some studies have suggested an association between PCOS and autoimmune thyroiditis, to our knowledge, only a few cases indicating association between PCOS and Gravesā€² disease are reported. Objective: We aim to describe this first case series of six women presenting with PCOS and Gravesā€² disease together. Materials and Methods: Women attending the endocrinology clinic at a tertiary care centre in north India and fulfilling the AE-PCOS criteria for diagnosis of PCOS were studied using a predefined proforma for any clinical, biochemical and imaging features of Gravesā€² disease. Results: The series consisted of six women with a mean age of 27.5 years and menarche as 12.6 years. All women were lean with mean BMI of 22.73 kg / m 2 and three out of six had waist circumference <80 cm. The mean FG score of subjects was 16.66 and average total testosterone was 77.02 ng / dl (25.0-119.64). All the patients had suppressed TSH, the average being 0.052 Ī¼IU/ml (0.01-0.15). Thyroid gland was enlarged in all clinically and on ultrasonography and imaging with 99m Tc and/or RAIU revealed diffuse increased uptake. Conclusions: The association of a rare disorder like Gravesā€² disease with a relatively common disorder like PCOS is unlikely to be because of a chance alone and may point to a common aetiopathogenic linkage leaving a scope for molecular characterization
