    The Amazing European Bison Journey prototype clip shows an extract of the timeline of this species coming back from their near disappearance. It starts in the Białowieża Forest in Poland in 1919, when they went extinct in the wild. And runs up to 2013 in Zuid-Kennemerland in the Nether-lands, where an initial herd of 6 European bison, translocated in 2007 from Poland, had quadrupled to 24 individuals living in the Dutch dunes. The process of production of this animated documentary is de-scribed starting with the ecological research required. Review of literature, contact with specialist scientists and visitation of Poland and the Netherlands are some of the starting points of the process. Subsequently, the preparation for the animation is described. The initial storyboard was constructed out of the preceding ecological research. Afterwards, the development of the script and its respective narration was recorded. The construction of the sets, props and characters is described; specific emphasis is given to the construction of the European bison. Finally, the assemblage of all the elements is explained and the final results of the animation prototype are revealed. This animation forms part of a broader project, which has research into the effectiveness of a set of visual communication formats at its core, and uses rewilding as its ecological platform. A range of animations, infographics and virtual reality pieces will be eventually compared for their effectiveness using several communication research tools

    Behavioral response of naĂŻve and non-naĂŻve deer to wolf urine

    Large carnivores are recolonizing many regions in Europe, where their ungulate prey have lived without them for >150 years. Whether the returning large carnivores will modify ungulate behavior and indirectly affect lower trophic levels, depends on the ability of ungulates to recognize risk based on past encounters and cues indicating carnivore presence. In two case studies, we tested, by means of camera trapping, the behavioral response of deer to wolf urine. The first case study was in the Netherlands where deer (still) live in absence of wolves, and the second in Poland with long-term wolf presence. As controls we used water (no scent) and all-purpose soap (unfamiliar scent). Deer vigilance level on control plots was 20% in both case studies indicating that wolf occupancy per se does not lead to a consistent difference in behavior. Placing wolf urine did not significantly affect deer behavior in either the wolf-absent or the wolf-present area. More intense cues, or a combination of cues, are likely needed to affect deer behavior. Moreover, we found an unexpected reaction of deer towards all-purpose soap of reduced foraging (and tendency for increased vigilance) in the wolf-present area, whereas it did not affect deer behavior in the wolf-absent area. We hypothesize that deer associate all-purpose soap with human presence, causing no response in human-dominated landscapes (the Netherlands), but triggering a behavioral reaction in more remote areas (Poland). This illustrates attention should be paid to controls used in scent experiments as they may be associated differently than intended

    Impediments affect deer foraging decisions and sapling performance

    Impediments, such as tree logs, can prevent access to saplings for deer, and can increase perceived predation risk by blocking view and escape possibilities for deer in areas with large carnivores. Therefore, impediments can influence deer foraging decisions and the trade-off between safety and food of different quality indirectly influencing tree regeneration. The aim of our study was to test how the presence of an impediment affects deer foraging behavior and tree sapling performance of eight species that differ in preference by deer. We planted saplings without, nearby and inside impediments and followed their fate for three consecutive years in Białowieża forest, Poland. We constructed 1 m high impediments of 5 × 5 m that would still allow deer to enter the impediment and forage from the saplings planted inside. However, we never recorded deer inside the impediments. Near the impediments deer visitation rate and cumulative visitation time was reduced. As a result, browsing intensity of all tree species was lower nearby and especially inside the impediment. Deer did not select different tree species without, near or inside the impediment. Due to the overall lower browsing intensity, tree saplings increased in height near the impediment and heights increased significantly within the impediment. The palatable, but browsing intolerant, Acer platanoides benefited most from the impediment as this species was highly selected and heavily browsed without an impediment. Followed by the palatable, but more browse tolerant Tilia cordata and Pyrus pyraster. In comparison, the presence of an impediment had a smaller effect on the less preferred Alnus glutinosa, Picea abies and Pinus sylvestris which survived well without an impediment. Our study showed that impediments modified deer behavior as they visited these plots less frequently and thereby indirectly reduced the browsing impact on the preferred tree species. Therefore, the potential for successful recruitment of preferred tree species is higher near an impediment, and especially when surrounded by impediment structures. In the long-term, the presence of natural impediments like tree logs allows browsing intolerant tree species to escape browsing, ultimately leading to a more diverse forest composition

    Prognostic significance of delirium subtypes in critically ill medical and surgical patients:a secondary analysis of a prospective multicenter study

    BACKGROUND: The prognostic implication of delirium subtypes in critically ill medical and surgical patients is scarcely investigated. The objective was to determine how delirium subtypes are associated with hospital mortality and other clinical outcomes. METHODS: We performed a secondary analysis on data from a prospective multicenter study aimed at implementation of delirium-oriented measures, conducted between 2012 and 2015 in The Netherlands. We included adults (≥ 18 years) admitted to the medical or surgical intensive care unit (ICU). Exclusion criteria were neurological admission diagnosis, persistent coma or ICU readmissions. Delirium was assessed using the Confusion Assessment Method-ICU or Intensive Care Delirium Screening Checklist, and delirium subtypes (hypoactive, hyperactive, or mixed) were classified using the Richmond Agitation–Sedation Scale. The main outcome was hospital mortality. Secondary outcomes were ICU mortality, ICU length of stay, coma, mechanical ventilation, and use of antipsychotics, sedatives, benzodiazepines and opioids. RESULTS: Delirium occurred in 381 (24.4%) of 1564 patients (52.5% hypoactive, 39.1% mixed, 7.3% hyperactive). After case-mix adjustment, patients with mixed delirium had higher hospital mortality than non-delirious patients (OR 3.09, 95%CI 1.79–5.33, p = 0.001), whereas hypoactive patients did not (OR 1.34, 95%CI 0.71–2.55, p = 0.37). Similar results were found for ICU mortality. Compared to non-delirious patients, both subtypes had longer ICU stay, more coma, increased mechanical ventilation frequency and duration, and received more antipsychotics, sedatives, benzodiazepines and opioids. Except for coma and benzodiazepine use, the most unfavourable outcomes were observed in patients with mixed delirium. CONCLUSIONS: Patients with mixed delirium had the most unfavourable outcomes, including higher mortality, compared with no delirium. These differences argue for distinguishing delirium subtypes in clinical practice and future research. Trial registration ClinicalTrials.gov NCT01952899.     Development of an HIV-1 Specific Microbicide Using Caulobacter crescentus S-Layer Mediated Display of CD4 and MIP1α

    The development of alternative strategies to prevent HIV infection is a global public health priority. Initial efforts in anti-HIV microbicide development have met with poor success as the strategies have relied on a non-specific mechanism of action. Here, we report the development of a microbicide aimed at specifically blocking HIV entry by displaying molecular components of the HIV/host cell attachment complex on the surface of Caulobacter crescentus, a harmless aquatic bacterium. This bacterium can be readily manipulated to present heterologous proteins at high density on its surface by genetic insertion into its crystalline surface layer protein [1], [2]. In separate constructions, we generated bacteria displaying domain 1 of CD4 and MIP1α. Each moiety reacted with specific antibodies by Western immunoblot and immuno-fluorescence microscopy. Microbicide functionality was assessed using an HIV pseudotype virus assay system representing Clade B subtypes. Bacteria displaying MIP1α reduced infectivity by 35–78% depending on the specific subtype while CD4 display reduced infection by as much as 56%. Combinations of both constructs reduced infectivity by nearly 98%. We demonstrated that HIV infection could be inhibited using a strategy aimed at HIV-specific molecular interactions with Caulobacter surface protein display, and that sufficient protein folding and conformation could be mimicked to bind and block entry. Further, this is the first demonstration that Caulobacter surface protein display may be a useful approach to preventing HIV infection or other viruses as a microbicide. We propose that this harmless bacterium, which is inexpensive to produce and formulate, might be suitable for topical applications as a viable alternative in the search for effective microbicides to counteract the world wide incidence of HIV infection
