Osmoregulatory reactions of frog erythrocytes under conditions of activation and blockade of Ca²+-channels

    Get PDF
The kinetics of cell osmoregulatory reactions under conditions of activation and blockade of Ca2+-channels was studied on a model of frog polyfunctional nucleated erythrocytes. Both activation and blockade of Ca2+-channels has been established to promote swelling of nuclei and an increase of the nucleocytoplasmic ratios under conditions of hypotonic exposur

Seasonal activity of frog erythrocytes by data of electrophoretic mobility

The peculiarities of frog erythrocyte electrophoretic mobility, coupled to the seasonal course of temperatures, have been studied. At the periods of anabiosis and of burst of hemopoiesis, in the vascular bed there increases the portion of functionally young erythrocytes (up to 22%) with increased values of the cell membrane surface charge. Preparation to winter is accompanied by a rise of the number of circulating functionally worn down blood cells (up to 60%) with low values of the superficial charge and low mobility in electrical field. Use of the cell microelectrophoresis method of evaluation of seasonal activity of frog erythrocytes allows obtaining objective data about the cellular surface charge and its depending functional cell activity without submitting the erythrocytes to modifying action

Estimation of surface potential of lymphocytes from patients with leukemia using kelvin probe mode

The feasibilities of the Kelvin probe mode were shown to estimate a surface potential of lympho cytes from the patients with leukemia. Having compared the values of the surface potential obtained by the traditional method of microelectrophoresis and by the Kelvin probe mode, it was established that the use of the atomic force microscope made it possible to obtain data which were of specified diagnostic importance while studying lymphocyte functional status at different stages in leucopoiesis and indicated progression of the pathological proces

Evaluation of the elastic properties and topography of leukocytes’ surface in patients with type 2 diabetes mellitus using atomic force microscope

The aim of study was to examine some morphometrical parameters (height, diameter) of the leukocytes (white blood cells - WBCs), their specific surface morphology (globular prominences and depression in WBCs) as well as their local elastic properties (Young’s modulus) in healthy persons and in patients with type 2 diabetes mellitus (T2DM) by means of the atomic force microscopy (AFM). Morphological and morphometrical parameters of human leukocytes were evaluated by AFM in tapped mod

Changes of the biophysical properties of blood corpuscles from the elderly under mechanical stress in vitro

Features of biophysical properties in blood cells from the elderly under mechanical stress applied in vitro have been studied. This stress was reported to trigger the signaling pathway engaged by purinergic receptor activation. The stiffness of the cell surfaces of red blood cells, granulocytes, lymphocytes, and platelets decreased, and the surface potential became more positive. Our findings add to the knowledge of the effect of mechanical deformation on leukocytes and platelets, which are the key regulators of homeostatic processes in the microvasculature, and on red blood cells involved in the regulation of vascular tone in arterioles and tissue oxygenation in the elderl

Effects of nanodispersed iron on the morphofunctional parameters of the blood system

    Get PDF
The effects of nanodispersed iron forms on the morphology and function of the blood system were studied. Maghemite and lepidocrocite caused a leukocytic shift towards segmented neutrophil forms, reduction of lymphocyte rigidity, and stimulated their compactization. In addition, the counts of small hyperchromatic erythrocytes with high rigidity increased in the blood flow. The results indicated that a single dose of nanodispersed iron-containing drugs improved the blood respiratory function and its microrheolog

Application of technologies of atomic force microscope investigation for evaluation of structure and properties of blood cells’ surfaces

With application of techniques of atomic force microscope investigation, we studied properties and topography of blood cells’ surfaces in patients with acute lymphoblastic and myeloblastic leucosis. It was stated that development of acute forms of lymphoblastic and myeloblastic types of proliferation is accompanied with reduction of stiffness of cell surface and increase of surface potentia

The stiffness of cell membrane was found to be one of the factors determining resistance of a cell in vitro to antibiotic doxorubicin action. The data obtained have important prognostic value in studying drug resistance of tumour blood cells and can be used as objective markers of efficiency of the antitumor therap
